  abstract  = {Durch den Einsatz magnetisierbarer Partikel lassen sich Stoffwechselprodukte direkt und selektiv aus feststoffreichen Fermentationssuspensionen abtrennen. Im Gegensatz zu klassischen Adsorbermaterialien k{\"o}nnen magnetisierbare Partikel mit sehr geringen Durchmessern verwendet werden. Zur deren Abtrennung ist jedoch ein hoher Magnetfeldgradient notwendig. Dieser wird in der Regel durch in der Trennkammer bzw. dem Magnetfeld eingebrachte magnetisierbare Dr{\"a}hte realisiert. Bei der Auslegung der Drahtgitter ist ein Kompromiss zwischen Abtrennrate und Durchl{\"a}ssigkeit n{\"o}tig. Die Ausrichtung der Dr{\"a}hte in Relation zum Magnetfeld, deren Abstand sowie die geometrische Anordnung k{\"o}nnen hierbei variiert werden. Zum Verst{\"a}ndnis der Einfl{\"u}sse auf das sich ausbildende Magnetfeld und die Fluiddynamik wurden Simulationen mit der Finite-Elemente-Methode durchgef{\"u}hrt und experimentell {\"u}berpr{\"u}ft. Hierf{\"u}r wurden die Dr{\"a}hte unter Variation von Anzahl, Richtung und Anordnung in den Hochgradient-Magnetseparator eingebracht. Erste Verifizierungen der Simulationen zeigen, dass die in Magnetfeldrichtung ausgerichteten Dr{\"a}hte (x-Achse) {\"u}ber die geringste Partikelr{\"u}ckhaltef{\"a}higkeit verf{\"u}gen. Die Dr{\"a}hte der y- und z-Achse halten den gr{\"o}ßten Anteil der Magnetpartikel zur{\"u}ck, wobei die Dr{\"a}hte in y-Richtung den h{\"o}chsten Feldgradienten ausbilden. Des Weiteren konnte gezeigt werden, dass eine rhomboedrische Drahtanordnung der kubischen vorzuziehen ist.},

  abstract  = {Compared with rodents and many other animal species, the human cytochrome P450 (P450) Cyp2c gene cluster varies significantly in the multiplicity of functional genes and in the substrate specificity of its enzymes. As a consequence, the use of wild-type animal models to predict the role of human CYP2C enzymes in drug metabolism and drug-drug interactions is limited. Within the human CYP2C cluster CYP2C9 is of particular importance, because it is one of the most abundant P450 enzymes in human liver, and it is involved in the metabolism of a wide variety of important drugs and environmental chemicals. To investigate the in vivo functions of cytochrome P450 Cyp2c genes and to establish a model for studying the functions of CYP2C9 in vivo, we have generated a mouse model with a deletion of the murine Cyp2c gene cluster and a corresponding humanized model expressing CYP2C9 specifically in the liver. Despite the high number of functional genes in the mouse Cyp2c cluster and the reported roles of some of these proteins in different biological processes, mice deleted for Cyp2c genes were viable and fertile but showed certain phenotypic alterations in the liver. The expression of CYP2C9 in the liver also resulted in viable animals active in the metabolism and disposition of a number of CYP2C9 substrates. These mouse lines provide a powerful tool for studying the role of Cyp2c genes and of CYP2C9 in particular in drug disposition and as a factor in drug-drug interaction.},

  abstract  = {Drought-induced forest decline, like the Scots pine mortality in inner-Alpine valleys, will gain in importance as the frequency and severity of drought events are expected to increase. To understand how chronic drought affects tree growth and tree-ring δ13C values, we studied mature Scots pine in an irrigation experiment in an inner-Alpine valley. Tree growth and isotope analyses were carried out at the annual and seasonal scale. At the seasonal scale, maximum δ13C values were measured after the hottest and driest period of the year, and were associated with decreasing growth rates. Inter-annual δ13C values in early- and latewood showed a strong correlation with annual climatic conditions and an immediate decrease as a response to irrigation. This indicates a tight coupling between wood formation and the freshly produced assimilates for trees exposed to chronic drought. This rapid appearance of the isotopic signal is a strong indication for an immediate and direct transfer of newly synthesized assimilates for biomass production. The fast appearance and the distinct isotopic signal suggest a low availability of old stored carbohydrates. If this was a sign for C-storage depletion, an increasing mortality could be expected when stressors increase the need for carbohydrate for defence, repair or regeneration.},

"Gr{\"u}n" verheißt im Allgemeinen Licht, Luft und Sonne, Freizeit und Erholung und gilt als etwas W{\"u}nschenswertes im st{\"a}dtischen Kontext. Unbeachtet bleibt, dass dem Begriff "Gr{\"u}n" zun{\"a}chst keine Eigenschaften oder Qualit{\"a}ten zugewiesen sind - er sagt noch lange nichts {\"u}ber dessen r{\"a}umliche Qualit{\"a}t, dessen Nutzbarkeit, Erreichbarkeit und Zug{\"a}nglichkeit aus. Abstandsgr{\"u}n und Straßenbegleitgr{\"u}n ist eben auch "Gr{\"u}n". Neben einer quantitativen Argumentation, welche {\"o}kologischen Aspekte wie Biotopvernetzung, Stadt- und Mikroklima ber{\"u}cksichtigt, sollten wir verst{\"a}rkt qualitativ denken und damit die Grundlage f{\"u}r einen erweiterten und wesentlich differenzierteren Diskurs {\"u}ber urbane Freir{\"a}ume ebnen.},

So k{\"o}nnen die Tropfen als fl{\"u}ssige Mikroreaktoren eingesetzt werden. Der w{\"a}ssrige Kern dieser Mikroreaktoren besteht aus einer Substratl{\"o}sung f{\"u}r enzymatische Umsetzungen. Durch Bewegen der Mikroreaktoren k{\"o}nnen diese {\"u}ber immobilisierten Enzymen positioniert werden, um so einen enzymatischen Umsatz innerhalb der Mikroreaktoren zu realisieren. Hierf{\"u}r wurde eine neue Mikroreaktorplattform-Technologie etabliert. Die Bewegung erfolgt dabei mit einer automatisierten Aktuatorplattform, bestehend aus einer 3x3 Doppelspulenmatrix mit Magnetkernen. Als modellhaftes Reaktionssystem wird eine Enzymkaskade eingesetzt, die sich aus einer b-Glucosidase, Glucose-Oxidase und Meerrettichperoxidase zusammensetzt.   abstract  = {Variability in xylem anatomy is of interest to plant scientists because of the role water transport plays in plant performance and survival. Insights into plant adjustments to changing environmental conditions have mainly been obtained through structural and functional comparative studies between taxa or within taxa on contrasting sites or along environmental gradients. Yet, a gap exists regarding the study of hydraulic adjustments in response to environmental changes over the lifetimes of plants. In trees, dated tree-ring series are often exploited to reconstruct dynamics in ecological conditions, and recent work in which wood-anatomical variables have been used in dendrochronology has produced promising results. Environmental signals identified in water-conducting cells carry novel information reflecting changes in regional conditions and are mostly related to short, sub-annual intervals. Although the idea of investigating environmental signals through wood anatomical time series goes back to the 1960s, it is only recently that low-cost computerized image-analysis systems have enabled increased scientific output in this field. We believe that the study of tree-ring anatomy is emerging as a promising approach in tree biology and climate change research, particularly if complemented by physiological and ecological studies. This contribution presents the rationale, the potential, and the methodological challenges of this innovative approach.},

  abstract  = {Increasing Scots pine (Pinus sylvestris L.) mortality has been recently observed in the dry inner valleys of the European Alps. Besides drought, infection with pine mistletoe (Viscum album ssp. austriacum) seems to play an important role in the mortality dynamics of Scots pines, but how mistletoes promote pine decline remains unclear. To verify whether pine mistletoe infection weakens the host via crown degradation, as observed for dwarf mistletoes, we studied the negative effects of pine mistletoe infestation on the photosynthetic tissues and branch growth of pairs of infested and non-infested branches. Pine mistletoe infection leads to crown degradation in its host by reducing the length, the radial increment, the ramification, the needle length and the number of needle years of the infested branches. This massive loss in photosynthetic tissue results in a reduction in primary production and a subsequent decrease in carbohydrate availability. The significant reduction in needle length due to mistletoe infection is an indication for a lower water and nutrient availability in infested branches. Thus, mistletoe infection might lead to a decrease in the availability of water and carbohydrates, the two most important growth factors, which are already shortened due to the chronic drought situation in the area. Therefore, pine mistletoe increases the risk of drought-induced mortality of its host when growing in a xeric environment.},

  abstract  = {Die selektive Isolierung von Cephalosporin C (CPC) aus komplexen Fermentationssuspensionen unter Einsatz magnetischer Separation ist das Ziel dieser Arbeit. Das Verfahren wird im fr{\"u}hen Stadium der Aufarbeitung genutzt, um CPC zu stabilisieren und somit die Produktausbeute zu erh{\"o}hen. Als Adsorbersysteme f{\"u}r CPC wurden neben einem projektinternen magnetischen Material ND 10322, dessen Oberfl{\"a}chenladungen spezifisch f{\"u}r die Bindung des Zielmolek{\"u}ls synthetisiert wurden, verschiedene kommerzielle Partikelsysteme verglichen. Es konnten massenspezifische Maximalbeladungen von 51 mg g⁻¹ erreicht werden. Weiterhin wurde die Stabilit{\"a}t von CPC untersucht. Unter optimalen Adsorptionsbedingungen kann CPC stabilisiert werden, so dass die Geschwindigkeitskonstante der Degradation des b-Lactam-Rings unter diesen Bedingungen unter 0,005 h⁻¹ liegt. Untersuchungen zur Wiederverwertbarkeit der neuen Adsorbers zeigten eine irreversible Bindung geringer CPC-Mengen nach dem ersten Einsatz. Nach zw{\"o}lf Zyklen tritt eine irreversible Bindung von CPC ein, was zu einer signifikanten Reduktion der Adsorptionsf{\"a}higkeit f{\"u}hrt. Die Anh{\"a}ufung des CPC auf dem Adsorber konnte durch IR-Untersuchungen auf die Bildung einer Peptidbindung zwischen Carboxylgruppen des CPC und Aminogruppe der Adsorberoberfl{\"a}che zur{\"u}ckgef{\"u}hrt werden.},

In der Veranstaltung wurden wesentliche Aspekte des Forschungsdatenmanagements entlang des Datenlebenszyklus beleuchtet. Ziel war es, den Teilnehmenden praxisnahe Einblicke und Hilfestellungen zu einem effektiven Umgang mit Forschungsdaten an Hochschulen f{\"u}r Angewandte Wissenschaften (HAW) zu bieten.   abstract  = {Die tert.-Butyloxycarbonyl-Gruppe (Boc) l{\"a}ßt sich mittels reiner Trifluoressigs{\"a}ure nicht selektiv neben dem Benzyloxycarbonyl-Rest (Z) abspalten. Das gelingt auch nicht mit L{\"o}sungen von Trifluoressigs{\"a}ure bzw. Chlorwasserstoff in organischen L{\"o}sungsmitteln. Kern-substituierte Z-Gruppen wie Z(pCl), Z(mCl) oder Z(pNO₂) sind zwar stabiler, werden aber von den obengenannten Reagenzien ebenfalls angegriffen bzw. sind nicht mehr acidolytisch abspaltbar. - Mit 70proz. w{\"a}ßriger Trifluoressigs{\"a}ure gelingt die Abspaltung von Boc neben Z dagegen fast selektiv; dabei werden aber Benzylester, besonders Glutamins{\"a}ure-γ-benzylester, teilweise hydrolysiert, w{\"a}hrend Methyl- sowie {\"A}thylester nahezu best{\"a}ndig sind. Die Brauchbarkeit des Abspaltungsverfahrens wird anhand der schrittweise durchgef{\"u}hrten Synthese zweier Heptapeptid-Derivate gezeigt. - {\"A}hnlich spezifisch gelingt die Abspaltung von Boc mit Bortrifluorid-{\"a}therat in Eisessig; Benzylester sind gegen{\"u}ber diesem Reagenz stabiler als gegen w{\"a}ßrige Trifluoressigs{\"a}ure. Das Bortrifluorid-Verfahren eignet sich besonders f{\"u}r die Abspaltung von Boc-Gruppen neben s{\"a}urelabilen Thiol-Schutzgruppen (Tetrahydropyranyl- bzw. Trityl-Rest) sowie neben dem Cyclocystinyl-Rest. Die Leistungsf{\"a}higkeit der Methode wird durch die Synthese zweier Peptid-Derivate mit S-Trityl-Schutzgruppen belegt. Als Nebenreaktion ist die Acetylierung von aliphatischen Hydroxylgruppen m{\"o}glich. Sie l{\"a}ßt sich vermeiden, wenn man die Spaltung in anderen L{\"o}sungsmitteln durchf{\"u}hrt. Die als Modellverbindungen f{\"u}r Stabilit{\"a}tsuntersuchungen verwendeten Nε-acylierten Lysin-Derivate werden mit dem Aminos{\"a}ureanalysator quantitativ neben Lysin bestimmt.},

  abstract  = {In current clinical cardiovascular MR (CMR) practice cardiac motion is commonly dealt with using ECG based synchronization. However, ECG is corrupted by magneto-hydrodynamic (MHD) effects in magnetic fields. This leads to artifacts in the ECG trace and evokes severe T-wave elevations, which might be misinterpreted as R-waves resulting in erroneous triggering. At (ultra)high field strengths, the propensity of ECG recordings to MHD effects is further pronounced. Pulse oximetry (POX) being inherently sensitive to blood oxygenation provides an alternative approach for cardiac gating. However, due to the travel time of the blood the peak of maximum oxygenation and hence the trigger is delayed by approx. 300 ms with respect to the ECG's R-wave. Also the peak of maximum oxygenation shows a jitter of up to 65 ms. Alternative triggering approaches include acoustic cardiac triggering (ACT). In current clinical practice cardiac gating / triggering commonly relies on using single physiological signals only. Realizing this limitation this study proposes a combined triggering approach which exploits multiple physiological signals including ECG, POX or ACT to track cardiac activity. The feasibility of the coupled approach is examined for LV function assessment at 7.0 T. For this purpose, breath-held 2D-CINE imaging in conjunction with cardiac synchronization was performed paralleled by real time logging of physiological waveforms to track (mis)synchronization between the cardiac cycle and data acquisition. Combinations of the ECG, POX and ACT signals were evaluated and processed in real time to facilitate reliable trigger information.},

  abstract  = {Der Erhalt m{\"o}glichst hoher Zuckerkonzentrationen f{\"u}r nachfolgende Fermentationen und eine Steigerung der Produktivit{\"a}t sind Ziele der Hydrolyse bei hohen Feststoffkonzentrationen im Rahmen des Projekts „Lignocellulose Bioraffinerie". Verwendet wird durch ein Organosolv-Verfahren aufgeschlossenes Buchenholz. Die Hydrolyse des Faserstoffes erfolgt mithilfe von CTec2-Enzymen (Fa. Novozymes). Zurzeit k{\"o}nnen unter Einsatz eines neuen Feststoffreaktors Cellulosefasern in einer Konzentration bis 400 g L⁻¹ enzymatisch hydrolysiert werden. Dabei werden Ausbeuten (g Glucose/g Cellulose im Faserstoff) bis 0,86 g g⁻¹ und Glucosekonzentrationenvon 120 g L⁻¹ erreicht. Ein Nachteil ist jedoch die hierbei auftretende Abnahme der Hydrolyseausbeuten. Zahlreiche Limitierungen bez{\"u}glich der Hydrolysierbarkeit von Lignocellulose werden zurzeit diskutiert und publiziert. Ziel der Untersuchungen ist die Identifizierung hydrolysehemmender Substanzen sowie die Erh{\"o}hung der Ausbeute an Zuckermonomeren durch den Einsatz lignolytischer Enzyme. Hierbei wird eine HPLC-MS-Methode zur Charakterisierung hemmender Substanzen eingesetzt, um potenzielle Inhibitoren zu erfassen.},

  abstract  = {Tumour cell death can be evaluated in the living mouse by externally measuring the rate of loss of tumour-bound DNA tracer. By sequentially labelling the tumour-bearing animals with ¹²⁵IUdR and ¹³¹IUdR 50 h apart, the average tumour cells at the time of the second injection are labelled by ¹²⁵IUdR and the euoxic tumour cells are specifically labelled with ¹³¹IUdR. Tumour treatment at this stage of labelling permits the observation of the reaction of euoxic cells and average tumour cells and finally yields data on hypoxic cells and thus on the oxygen enhancement ratio. This information adds to results from tumour control and growth delay. With this technique effects were analysed of 60-Co γ-rays, cyclotron neutrons (E = 6 MeV), misonidazole (500 mg/kg body wt) and hyperthermia (42°C water-bath), or combinations of these. Misonidazole (15 min before irradiation) altered the oxygen enhancement ratio by a factor of 1·5 for γ-rays and of 1·1 for neutrons; when evaluated from tumour-growth delay and TCD-50 misonidazole gave a dose modifying factor of 1·47 for γ-rays and of 1·2-1·3 for neutrons. Based on percentage tumour regression 100 days after treatment, the enhancement ratio from hyperthermia (after irradiation) was 2·75 for γ-rays (at 10 Gray) and 2·2 for neutrons (at 3·2 Gray). For neutrons combined with misonidazole and hyperthermia the ratio was 2·4. These results demonstrate that effects of neutron irradiation may be modified by electron-affinic substances and/or hyperthermia.},
