The purpose of this paper is to analyse the opportunities and obstacles to implementing BIM for prefabrication. Among all other advantages of BIM, prefabrication is chosen for this paper because it plays a vital role in creating an impact on the time and cost factors of a construction project. The project stakeholders and participants can explicitly observe the positive impact of prefabrication, which enables the breakthrough of the scepticism factor among the small-scale construction companies. The analysis consists of the development of a process workflow for implementing prefabrication in building construction followed by a practical approach, which was executed with two case studies. It was planned in such a way that, the first case study gives a first-hand experience for the workers at the site on the BIM model so that they can make much use of the created BIM model, which is a better representation compared to the traditional 2D plan. The main aim of the first case study is to create a belief in the implementation of BIM Models, which was succeeded by the execution of offshore prefabrication in the second case study. Based on the case studies, the time analysis was made and it is inferred that the implementation of BIM for prefabrication can reduce construction time, ensures minimal wastes, better accuracy, less problem-solving at the construction site. It was observed that this process requires more planning time, better communication between different disciplines, which was the major obstacle for successful implementation. It also enables the production of certain foods in places where they usually dare not produced, such as production of fish or shrimps in large cities far away from the coast. Unfortunately, it is not always possible to show students such concepts and systems in real life as part of courses: visits of such industry plants are sometimes not possible because of distance or are permitted by the operator for hygienic reasons. In order to give the students the opportunity of getting into contact with such an urban farming system and its complex operation, an industrial urban farming plant was set up on a significantly smaller scale. Therefore, all needed technical components like water aeriation, biological and mechanical filtration or water circulation have been replaced either by aquarium components or by self-designed parts also using a 3D-printer. Students from different courses like mechanical engineering, smart building engineering, biology, electrical engineering, automation technology and civil engineering were involved in this project. This “miniature industrial plant” was also able to start operation and has now been running for two years successfully. Due to Corona pandemic, home office and remote online lectures, the automation of this miniature plant should be brought to a higher level in future for providing a good control over the system and water quality remotely. Since stakeholders differ regarding their preferences and time horizons, we employ a multi-criteria decision analysis approach to identify which stakeholders will support or oppose a transition path. We provide a flexible template for analyzing stakeholder preferences toward transition paths. This flexibility comes from the fact that our multi-criteria decision aid-based approach does not involve intensive empirical work with stakeholders. During parabolic flights, while 17 subjects performed drop jumps (DJs) and drop landings (DLs), electromyography (EMG) of the lower limb muscles was combined with in vivo fascicle dynamics of the gastrocnemius medialis, two-dimensional (2D) kinematics, and kinetics to measure and analyze changes in energy management. Comparisons were made between movement modalities executed in hypergravity (1.8 G) and gravity on ground (1 G). In 1.8 G, ankle dorsiflexion, knee joint flexion, and vertical center of mass (COM) displacement are lower in DJs than in DLs; within each movement modality, joint flexion amplitudes and COM displacement demonstrate higher values in 1.8 G than in 1 G. Concomitantly, negative peak ankle joint power, vertical ground reaction forces, and leg stiffness are similar between both movement modalities (1.8 G). In DJs, EMG activity in 1.8 G is lower during the COM deceleration phase than in 1 G, thus impairing quasi-isometric fascicle behavior. In DLs, EMG activity before and during the COM deceleration phase is higher, and fascicles are stretched less in 1.8 G than in 1 G. Compared with the situation in 1 G, highly task-specific neuromuscular activity is diminished in 1.8 G, resulting in fascicle lengthening in both movement modalities. Specifically, in DJs, a high magnitude of neuromuscular activity is impaired, resulting in altered energy storage. In contrast, in DLs, linear stiffening of the system due to higher neuromuscular activity combined with lower fascicle stretch enhances the buffering function of the tendon, and thus the capacity to safely dissipate energy. The present research work closes this gap and sheds light into decisive factors for purchasing products offered in Instagram stores. A theoretical research model, which contains selected constructs that are assumed to have a significant influence on Instagram user´s purchase intention, is developed. The hypotheses are evaluated by applying structural equation modelling on survey data containing 127 relevant participants. The results of the study reveal that ‘trust’, ‘personal recommendation’, and ‘usability’ significantly influences user’s buying intention in Instagram stores. The presented course "Applying Design Thinking - Sustainability, Innovation and Interculturality" was developed to teach future skills from the competency areas Digital Key Competencies, Classical Competencies and Transformative Competencies. The CDIO Standard 3.0, in particular the standards 5, 6, 7 and 8, was used as a guideline. In this report we present a technique based on a magnetic tweezers setup to measure the velocity factor of magnetically actuated individual superparamagnetic beads in a fluidic environment. Several beads can be tracked simultaneously in order to gain and improve statistics. Furthermore we show our results for different beads with hydrodynamic diameters between 200 and 1000 nm from diverse manufacturers. These measurement data can, for example, be used to determine design parameters for a magnetic separation system, like maximum flow rate and minimum separation time, or to select suitable beads for fixed experimental requirements. To ensure that this transformation creates work systems in which employees are empowered, productive, healthy, and motivated, the transformation must be guided by principles of and research on human-centered work design. Specifically, measures must be taken at all levels of work design, ranging from (1) the work tasks to (2) the working conditions to (3) the organizational level and (4) the supra-organizational level. We present selected research across all four levels that showcase the opportunities and requirements that surface when striving for human-centered work design for the Internet of Production (IoP). (1) On the work task level, we illustrate the user-centered design of human-robot collaboration (HRC) and process planning in the composite industry as well as user-centered design factors for cognitive assistance systems. (2) On the working conditions level, we present a newly developed framework for the classification of HRC workplaces. (3) Moving to the organizational level, we show how corporate data can be used to facilitate best practice sharing in production networks, and we discuss the implications of the IoP for new leadership models. Finally, (4) on the supra-organizational level, we examine overarching ethical dimensions, investigating, e.g., how the new work contexts affect our understanding of responsibility and normative values such as autonomy and privacy. Overall, these interdisciplinary research perspectives highlight the importance and necessary scope of considering the human factor in the IoP. Simultaneous detection of multiple biomolecular targets from one body fluid sample is desired. The aim of this work is to show that multiplex magnetic immunodetection based on magnetic frequency mixing by means of modular immunofiltration columns prepared for different targets is feasible. By calculations of the magnetic response signal, the required spacing between the modules was determined. Immunofiltration columns were manufactured by 3D printing and antibody immobilization was performed in a batch approach. It was shown experimentally that two different target molecules in a sample solution could be individually detected in a single assaying step with magnetic measurements of the corresponding immobilization filters. The arrangement order of the filters and of a negative control did not influence the results. Thus, a simple and reliable approach to multi-target magnetic immunodetection was demonstrated. The goal of this work is to investigate the feasibility of simultaneously detecting different superparamagnetic beads acting as markers using the magnetic frequency mixing technique. The frequency of the magnetic excitation field is scanned while the lower driving frequency is kept constant. Due to the particles’ nonlinear magnetization, mixing frequencies are generated. To record their amplitude and phase information, a direct digitization of the pickup-coil’s signal with subsequent Fast Fourier Transformation is performed. By synchronizing both magnetic beads using frequency scanning in magnetic frequency mixing technique magnetic fields, a stable phase information is gained. In this research, it is shown that the amplitude of the dominant mixing component is proportional to the amount of superparamagnetic beads inside a sample. Additionally, it is shown that the phase does not show this behaviour. Excitation frequency scans of different bead types were performed, showing different phases, without correlation to their diverse amplitudes. Two commercially available beads were selected and a determination of their amount in a mixture is performed as a demonstration for multiplex measurements. We used two different commercially available antibodies to capture CTB and for binding to superparamagnetic beads. ELISA experiments were performed to select the antibody combination. The beads act as labels for the magnetic frequency mixing detection technique. We show that the limit of detection depends on the type of magnetic beads. A nonlinear Hill curve was fitted to the calibration measurements by means of a custom-written python software. We achieved a sensitive and rapid detection of CTB within a broad concentration range from 0.2 ng/ml to more than 700 ng/ml. Dabei werden die MBs mit Hilfe von primären Antikörpern an die Zielsubstanz gebunden und mit sekundären Antikörpern an die Poren-Oberfläche eines Polyethylen-Filters fixiert (Sandwich-Immunoassay). So kann die Quantifizierung der Zielsubstanz auf eine magnetische Messung der immobilisierten MB-Marker zurückgeführt werden. Die magnetische Frequenzmischtechnik basiert auf der Anregung der Probe mit Magnetfeldern zweier verschiedener Frequenzen. Die durch die nichtlineare Magnetisierungsform der superparamagnetischen MBs entstehenden Mischfrequenzen werden typischerweise mithilfe einer zweistufigen Lock-in-Detektion analysiert (analoge Demodulation), die in einem Magnetreader als Handheldgerät realisiert wurde. Zusätzlich zu dieser Technik wurde das Prinzip der direkten Digitalisierung des gesamten Antwortsignals mit anschließender Fourier-Analyse der erzeugten Mischfrequenzen experimentell umgesetzt, um die Amplituden und Phasen mehrerer Mischfrequenzen simultan zu erfassen. Einerseits wurde eine räumliche Separation der Bindungsbereiche für verschiedene Zielsubstanzen realisiert, die sequentiell ausgewertet werden können. Andererseits wurde die Unterscheidung von verschiedenen Zielsubstanzen anhand der Charakteristika der an sie gebundenen, verschieden funktionalisierten MB-Typen untersucht. Für eine solche Unterscheidung wurde zum einen die Anregefrequenz der magnetischen Frequenzmischtechnik während einer Messung variiert. Damit konnte gezeigt werden, dass sich verschiedene MB-Sorten anhand der Phase ihrer Frequenzmischsignale voneinander unterscheiden lassen. Weiterhin wurde gezeigt, dass sich der Signalverlauf einer binären Mischung zweier verschiedener MB-Typen als gradueller Übergang der Verläufe der beiden reinen MB-Lösungen ergibt. Eine weitere Analysemethode für einen multiparametrischen Immunoassay besteht darin, ein zusätzliches einstellbares statisches magnetisches Offsetfeld zu verwenden. Hierfür wurden mehrere Aufbauten auf Basis von Permanent- und Elektromagneten simuliert, konstruiert und charakterisiert. Mithilfe von Simulationen konnte gezeigt werden, dass eine auf diesem Verfahren beruhende Unterscheidung für MBs mit unterschiedlichen magnetischen Partikelmomenten möglich ist. N2 - The qualitative and quantitative detection of target substances in an aqueous sample is of interest for many questions, for example in the detection of contaminations in drinking water in crisis situations. It is not only important that pathogens can be detected with highest possible sensitivity, but also that the analysis is carried out quickly so that safe drinking water can be provided in the event of a disaster. During such a scenario one cannot rely on a functioning laboratory infrastructure nearby. Therefore it is important that the measurement can be carried out directly on site. Within the scope of this work, it was investigated whether such a quick analysis can be performed using superparamagnetic beads (MBs) and the magnetic frequency mixing technique. The MBs are bound to the target substance with the aid of primary antibodies and fixed to the pore surfaces of a polyethylene filter with secondary antibodies (sandwich immunoassay). The quantification of the target substance can thus be traced back to a magnetic measurement of the immobilized MB markers. The magnetic frequency mixing technique is based on the excitation of the sample with magnetic fields of two different frequencies. The mixing frequencies generated due to the non-linear shape of the magnetization of the superparamagnetic MBs are typically analyzed using a two-stage Lock-in detection (analog demodulation), which was implemented in a magnetic reader as a handheld device. In addition to this technique, the principle of direct digitization of the entire response signal with subsequent Fourier analysis of the generated mixing frequencies was experimentally implemented in order to simultaneously record the amplitudes and phases of several mixing frequencies. One possibility for increasing the sensitivity is magnetic concentration. In that case, the MBs are separated from a larger sample volume by means of a magnetic field gradient before the magnetic analysis. To characterize various commercial MBs with regard to their magnetic separability, a setup for measuring their magnetophoretic mobility was implemented and their velocities in the gradient field were measured with an optical microscope.Often, a sample has to be examined not only for a single target substance, but for several different pathogens simultaneously. Various approaches have been developed and tested which enable such a multiparametric magnetic immunoassay. On the one hand, a spatial separation of the binding areas for different target substances was realized, which can be evaluated sequentially. On the other hand, a distinction among different target substances based on the magnetic characteristics of their attached different MB types was examined. For this discrimination, the excitation frequency of the magnetic frequency mixing technology was varied during measurement. It is shown that different MB types can be distinguished from one another based on the phase of their frequency mixing signals. The signal curve of a binary mixture of two different MB types is obtained as a gradual transition of the curves of the two pure MB solutions. Another method of analysis for a multiparametric immunoassay is based on an additional adjustable static magnetic offset field. For this purpose, several setups based on permanent magnets and electromagnets were simulated, designed and characterized. The simulations show that a distinction based on this method is possible for MBs with different magnetic particle moments. As a direct application of the developed magnetic reader in conjunction with digital demodulation, a magnetic assay against the B subunit of cholera toxin in drinking water was demonstrated, and a low detection limit of 0.2 ng/ml was achieved. The influence of different curing parameters of the carbon electrodes on the resulting biosensor characteristics is studied. The morphology of the electrodes is investigated by scanning electron microscopy, and the biosensor performance is examined by amperometric measurements of glucose (0.5–10 mM) in phosphate buffer solution, pH 7.4, at an applied potential of 1.2 V versus a Ag/AgCl reference electrode. Instead of Ecoflex, fibroin, PLA, and wound adhesive are tested as alternative encapsulation compounds: a series of swelling tests with different fibroin compositions, PLA, and Ecoflex has been performed before characterizing the most promising candidates by chronoamperometry. The analyses were done with limited examination windows concerning contraction level, muscle–tendon complex length, and/or intramuscular position of ultrasound imaging. With this study we aimed to investigate the correlation between fascicle arching and contraction, muscle–tendon complex length and their associated architectural parameters in gastrocnemius muscles to develop hypotheses concerning the fundamental mechanism of fascicle curving. Twelve participants were tested in five different positions (90°/105°*, 90°/90°*, 135°/90°*, 170°/90°*, and 170°/75°*; *knee/ankle angle). They performed isometric contractions at four different contraction levels (5%, 25%, 50%, and 75% of maximum voluntary contraction) in each position. Panoramic ultrasound images of gastrocnemius muscles were collected at rest and during constant contraction. Aponeuroses and fascicles were tracked in all ultrasound images and the parameters fascicle curvature, muscle–tendon complex strain, contraction level, pennation angle, fascicle length, fascicle strain, intramuscular position, sex and age group were analyzed by linear mixed effect models. Mean fascicle curvature of the medial gastrocnemius increased with contraction level (+5 m−1 from 0% to 100%; p = 0.006). Muscle–tendon complex length had no significant impact on mean fascicle curvature. Mean pennation angle (2.2 m−1 per 10°; p < 0.001), inverse mean fascicle length (20 m−1 per cm−1; p = 0.003), and mean fascicle strain (−0.07 m−1 per +10%; p = 0.004) correlated with mean fascicle curvature. Evidence has also been found for intermuscular, intramuscular, and sex-specific intramuscular differences of fascicle curving. Pennation angle and the inverse fascicle length show the highest predictive capacities for fascicle curving. Due to the strong correlations between pennation angle and fascicle curvature and the intramuscular pattern of curving we suggest for future studies to examine correlations between fascicle curvature and intramuscular fluid pressure. For operation independent from the ICE speed, the EDC compressor is decoupled from the turbine by using an electric compressor motor (CM) instead of the turbine. The algorithms not only ensure operator safety by monitoring the fuel cell and connected components, but also contribute to extending the health of the fuel cell, its durability and safe operation over its lifetime. While sensors are used to provide peripheral data surrounding the fuel cell, the internal states of the fuel cell cannot be directly measured. To overcome this restriction, Kalman Filter has been implemented as an internal state observer. Other safety conditions are evaluated using real-time data from every connected sensor and corrective actions automatically take place to ensure safety. The algorithms discussed in this paper have been validated thorough Model-in-the-Loop (MiL) tests as well as practical validation at a dedicated test bench. The most common biofuels today are biodiesel and bioethanol. Since diesel engines are rather rare in aviation this survey is focusing on ethanol admixed to gasoline products. The Directive 2003/30/EC of the European Parliament and the Council of May 8th 2003 on the promotion of the use of biofuels or other renewable fuels for transport encourage a growing admixture of biogenic fuel components to fossil automotive gasoline. Some aircraft models equipped with spark ignited piston engines are approved for operation with automotive gasoline, frequently called “MOGAS” (motor gasoline). The majority of those approvals is limited to MOGAS compositions that do not contain methanol or ethanol beyond negligible amounts. In the past years (bio-)MTBE or (bio-)ETBE have been widely used as blending component of automotive gasoline whilst the usage of low-molecular alcohols like methanol or ethanol has been avoided due to the handling problems especially with regard to the strong affinity for water. With rising mandatory bio-admixtures the conversion of the basic biogenic ethanol to ETBE, causing a reduction of energetic payoff, becomes more and more unattractive. Therefore the direct ethanol admixture is accordingly favoured. Due to the national enforcements of the directive 2003/30/EC more oxygenates produced from organic materials like bioethanol have started to appear in automotive gasolines already. The current fuel specification EN 228 already allows up to 3 % volume per volume (v/v) (bio-)methanol or up to 5 % v/v (bio-)ethanol as fuel components. This is also roughly the amount of biogenic components to comply with the legal requirements to avoid monetary penalties for producers and distributors of fuels. Since automotive fuel is cheaper than the common aviation gasoline (AVGAS), creates less problems with lead deposits in the engine, and in general produces less pollutants it is strongly favoured by pilots. But being designed for a different set of usage scenarios the use of automotive fuel with low molecular alcohols for aircraft operation may have adverse effects in aviation operation. The newly designed 1.8L turbocharged gasoline engine incorporates infinitely variable intake and outlet control timing and direct fuel injection utilizing piezo injectors centrally located. In addition, this engine uses a prototype FEV engine control system, with software that was developed and adapted entirely by FEV. The vehicle features a 160 kW engine with a maximum mean effective pressure of 22.4 bar and 34 % savings in simulated fuel consumption. During the first stage, a new electrohydraulically actuated hybrid transmission with seven forward gears and one reverse gear and a single dry starting clutch will be integrated. The electric motor of the hybrid is directly connected to the gear set of the transmission. Utilizing the special gear set layout, the electric motor can provide boost during a change of gears, so that there is no interruption in traction. Therefore, the transmission system combines the advantages of a double clutch controlled gear change (gear change without an interruption in traction) with the efficient, cost-effective design of an automated manual transmission system. Volume 1: Symposia, Parts A and B N2 - Flow separation is a phenomenon that occurs in all kinds of supersonic nozzles sometimes during run-up and shut-down operations. Especially in expansion nozzles of rocket engines with large area ratio, flow separation can trigger strong side loads that can damage the structure of the nozzle. The investigation presented in this paper seeks to establish measures that may be applied to alter the point of flow separation. In order to achieve this, a supersonic nozzle was placed at the exit plane of the conical nozzle. This resulted in the generation of cross flow surrounding the core jet flow from the conical nozzle. Due to the entrainment of the gas stream from the conical nozzle the pressure in its exit plane was found to be lower than that of the ambient. A Cold gas instead of hot combustion gases was used as the working fluid. A mathematical simulation of the concept was validated by experiment. Measurements confirmed the simulation results that due to the introduction of a second nozzle the pressure in the separated region of the conical nozzle was significantly reduced. It was also established that the boundary layer separation inside the conical nozzle was delayed thus allowing an increased degree of overexpansion. The condition established by the pressure measurements was also demonstrated qualitatively using transparent nozzle configurations. The mobility sector plays a significant role in this shift, affecting different people and groups on multiple levels. Without acceptance from these stakeholders, emission targets are out of reach. This research analyzes how the heterogeneous preferences of various stakeholders align with the transformation of the mobility sector, looking at the extent to which the German transformation paths are supported and where stakeholders are located. Under the research objective of comparing stakeholders' preferences to identify which car segments require additional support for a successful climate transition, a status quo of stakeholders and car performance criteria is the foundation for the analysis. Stakeholders' hidden preferences hinder the derivation of criteria weightings from stakeholders; therefore, a ranking from observed preferences is used. This study's inverse multi-criteria decision analysis means that weightings can be predicted and used together with a recalibrated performance matrix to explore future preferences toward car segments. Results show that stakeholders prefer medium-sized cars, with the trend pointing towards the increased potential for alternative propulsion technologies and electrified vehicles. These insights can guide the improved targeting of policy supporting the energy and mobility transformation. Additionally, the method proposed in this work can fully handle subjective approaches while incorporating a priori information. A software implementation of the proposed method completes this work and is made publicly available. Considering the currently growing enzyme market for efficient and novel biocatalysts, data mining is a promising source for novel, as yet uncharacterised enzymes, especially from halophilic or halotolerant Bacillaceae, which offer great potential to meet industrial needs. Both halophilic bacteria Pontibacillus marinus DSM 16465ᵀ and Alkalibacillus haloalkaliphilus DSM 5271ᵀ and both halotolerant bacteria Metabacillus indicus DSM 16189 and Litchfieldia alkalitelluris DSM 16976ᵀ served as a source for the four new subtilisins SPPM, SPAH, SPMI and SPLA. The protease genes were cloned and expressed in Bacillus subtilis DB104. Purification to apparent homogeneity was achieved by ethanol precipitation, desalting and ion-exchange chromatography. Enzyme activity could be observed between pH 5.0–12.0 with an optimum for SPPM, SPMI and SPLA around pH 9.0 and for SPAH at pH 10.0. The optimal temperature for SPMI and SPLA was 70 °C and for SPPM and SPAH 55 °C and 50 °C, respectively. All proteases showed high stability towards 5% (w/v) SDS and were active even at NaCl concentrations of 5 M. The four proteases demonstrate potential for future biotechnological applications. Knowledge of the extent of the impact is crucial for overall aircraft performance. An integrated simulation environment for steady aeroelastic propeller simulations is presented. The simulation environment is applied to determine the impact of elastic deformations on the aerodynamic propeller performance. The aerodynamic module includes a blade element momentum approach to calculate aerodynamic loads. The structural module is based on finite beam elements, according to Timoshenko theory, including moderate deflections. Several fixed-pitch propellers with thin-walled cross sections made of both isotropic and non-isotropic materials are investigated. The essential parameters are varied: diameter, disc loading, sweep, material, rotational, and flight velocity. The relative change of thrust between rigid and elastic blades quantifies the impact of propeller elasticity. Swept propellers of large diameters or low disc loadings can decrease the thrust significantly. High flight velocities and low material stiffness amplify this tendency. Performance calculations without consideration of propeller elasticity can lead to decreased efficiency. To avoid cost- and time-intense redesigns, propeller elasticity should be considered for swept planforms and low disc loadings.
