TY - JOUR A1 - Honarvarfard, Elham A1 - Gamella, Maria A1 - Poghossian, Arshak A1 - Schöning, Michael Josef A1 - Katz, Evgeny T1 - An enzyme-based reversible Controlled NOT (CNOT) logic gate operating on a semiconductor transducer JF - Applied Materials Today N2 - An enzyme-based biocatalytic system mimicking operation of a logically reversible Controlled NOT (CNOT) gate has been interfaced with semiconductor electronic transducers. Electrolyte–insulator–semiconductor (EIS) structures have been used to transduce chemical changes produced by the enzyme system to an electronically readable capacitive output signal using field-effect features of the EIS device. Two enzymes, urease and esterase, were immobilized on the insulating interface of EIS structure producing local pH changes performing XOR logic operation controlled by various combinations of the input signals represented by urea and ethyl butyrate. Another EIS transducer was functionalized with esterase only, thus performing Identity (ID) logic operation for the ethyl butyrate input. Both semiconductor devices assembled in parallel operated as a logically reversible CNOT gate. The present system, despite its simplicity, demonstrated for the first time logically reversible function of the enzyme system transduced electronically with the semiconductor devices. The biomolecular realization of a CNOT gate interfaced with semiconductors is promising for integration into complex biomolecular networks and future biosensor/biomedical applications. The stepwise expansion of the malate platform with the enzymes fumarate hydratase (FH, EC 4.2.1.2) and aspartate ammonia-lyase (ASPA, EC 4.3.1.1) resulted in multi-enzyme reaction cascades and, thus, augmentation of the substrate spectrum of the sensors. Electrochemical measurements were carried out in presence of the cofactor β-nicotinamide adenine dinucleotide (NAD+) and the redox mediator hexacyanoferrate (III) (HCFIII). The amperometric detection is mediated by oxidation of hexacyanoferrate (II) (HCFII) at an applied potential of + 0.3 V vs. Ag/AgCl. For each biosensor, optimum working conditions were defined by adjustment of cofactor concentrations, buffer pH, and immobilization procedure. Under these improved conditions, amperometric responses were linear up to 3.0 mM for L-malate and fumarate, respectively, with a corresponding sensitivity of 0.7 μA mM−1 (L-malate biosensor) and 0.4 μA mM−1 (fumarate biosensor). The L-aspartate detection system displayed a linear range of 1.0–10.0 mM with a sensitivity of 0.09 μA mM−1. The sensor characteristics suggest that the developed platform provides a promising method for the detection and differentiation of the three substrates. In this work, an amperometric biosensor array for simultaneous determination of ethanol, formate, d- and l-lactate is presented. Enzyme immobilization on platinum thin-film electrodes was realized by chemical cross-linking with glutaraldehyde. The optimization of the sensor performance was investigated with regard to enzyme loading, glutaraldehyde concentration, pH, cofactor concentration and temperature. Under optimal working conditions (potassium phosphate buffer with pH 7.5, 2.5 mmol L-1 NAD+, 2.0 mmol L-1 ferricyanide, 25 °C and 0.4% glutaraldehyde) the linear working range and sensitivity of the four sensor elements was improved. Simultaneous and cross-talk free measurements of four different metabolic parameters were performed successfully. The reliable analytical performance of the biosensor array was demonstrated by application in a clarified sample of inoculum sludge. Thereby, a promising approach for on-site monitoring of fermentation processes is provided. T1 - Development and Testing of a Low NOX Micromix Combustion Chamber for an Industrial Gas Turbine JF - International Journal of Gas Turbine, Propulsion and Power Systems N2 - The Micromix combustion principle, based on cross-flow mixing of air and hydrogen, promises low emission applications in future gas turbines. The Micromix combustion takes place in several hundreds of miniaturized diffusion-type micro-flames. The major advantage is the inherent safety against flash-back and low NOx-emissions due to a very short residence time of reactants in the flame region. The paper gives insight into the Micromix design and scaling procedure for different energy densities and the interaction of scaling laws and key design drivers in gas turbine integration. Numerical studies, experimental testing, gas turbine integration and interface considerations are evaluated. The aerodynamic stabilization of the miniaturized flamelets and the resulting flow field, flame structure and NOx formation are analysed experimentally and numerically. The results show and confirm the successful adaption of the low NOx Micromix characteristics for a range of different nozzle sizes, energy densities and thermal power output. Researchers explore the fine details of why diversity management is central for generating innovations in heterogeneous research groups and how it could be effectively implemented into organizations. Complex research associations that discuss topics with a high impact on society increasingly address the necessity of establishing a diverse workforce to confront the challenges of tomorrow. Characterized by complex management structures as well as hierarchies, research associations have not been a subject of investigation until now. For this reason, the presented research project aims to develop a diversity and innovation management strategy with the ultimate goal of inducing change in the corporate culture. The proposed approach consisted of six phases; the first two phases investigated the status quo of diversity in the existing organizational structures of member institutes and the variety of particular working cultures within the research association. The third and the fourth phases utilized qualitative and quantitative studies. The third phase focused on the connection of management level to diversity and innovation, and the need for diversity and innovation management, and tailor-made methods of implementing them. The first three phases have been accomplished successfully; preliminary results are already available. The fourth phase will mainly focus on exploring the mind-set of the employees. The fifth phase will consolidate the findings in the first four phases into an implementable strategy. The final phase will address the implementation of this strategy into the organization. Phases 4 to 6 have not yet been undertaken KW - diversity management KW - innovation management KW - research association KW - change management KW - engineering Y1 - 2017 SN - 1757-5877 VL - 5 IS - 2 SP - 49 EP - 72 ER - TY - JOUR A1 - Schöning, Michael Josef A1 - Bronder, Thomas A1 - Wu, Chunsheng A1 - Scheja, Sabrina A1 - Jessing, Max A1 - Metzger-Boddien, Christoph A1 - Keusgen, Michael A1 - Poghossian, Arshak T1 - Label-Free DNA Detection with Capacitive Field-Effect Devices—Challenges and Opportunities JF - Proceedings N2 - Field-effect EIS (electrolyte-insulator-semiconductor) sensors modified with a positively charged weak polyelectrolyte layer have been applied for the electrical detection of DNA (deoxyribonucleic acid) immobilization and hybridization by the intrinsic molecular charge. The EIS sensors are able to detect the existence of target DNA amplicons in PCR (polymerase chain reaction) samples and thus, can be used as tool for a quick verification of DNA amplification and the successful PCR process. Due to their miniaturized setup, compatibility with advanced micro- and nanotechnologies, and ability to detect biomolecules by their intrinsic molecular charge, those sensors can serve as possible platform for the development of label-free DNA chips. Possible application fields as well as challenges and limitations will be discussed. This is especially true for dynamically loaded silos, e.g., in case of seismic excitation. Particularly thin-walled cylindrical silos are highly vulnerable to seismic induced pressures, which can cause critical buckling phenomena of the silo shell. The analysis of silos can be carried out in two different ways. In the first, the seismic loading is modeled through statically equivalent loads acting on the shell. Alternatively, a time history analysis might be carried out, in which nonlinear phenomena due to the filling as well as the interaction between the shell and the granular material are taken into account. The paper presents a comparison of these approaches. The model used for the nonlinear time history analysis considers the granular material by means of the intergranular strain approach for hypoplasticity theory. The interaction effects between the granular material and the shell is represented by contact elements. Additionally, soil–structure interaction effects are taken into account. An equivalent macro-element constituted by a set of six trusses, two for each transverse, longitudinal and diagonal direction, is introduced. The equivalent trusses, whose stiffness is calculated by fully modeled vaults of different geometry, mechanical properties and boundary conditions, simulate the vault in both global analysis and local analysis, such as kinematic or rocking approaches. A parametric study was carried out to investigate the influence of geometrical characteristics and mechanical features on the equivalent stiffness values. The method was numerically validated by performing modal and transient analysis on a three naves-church in the elastic range. Vibration modes and displacement time-histories were compared showing satisfying agreement between the complete and the simplified models. This procedure is particularly useful in engineering practice because it allows to assess, in a simplified way, the effectiveness of strengthening interventions for reducing horizontal relative displacements between vault supports.
