This paper develops a conceptual model for the impact quantification of AM on volume and mix flexibility within production systems in the early stages of the factory-planning process. Together with the model, an application guideline is presented to help planners with the flexibility quantification and the factory design process. Following the development of the model and guideline, a case study is presented to indicate the potential impact additive technologies can have on manufacturing flexibility Within the case study, various scenarios with different production system configurations and production programs are analyzed, and the impact of the additive technologies on volume and mix flexibility is calculated. Die Dienste benötigen für die verschiedenen Medien nicht mehr so große Übertragungs- und Speicherleistungen. Bei den entwickelten Verfahren, den so genannten MPEG-(Motion Picture Experts Group-)Standards, werden die Video- und Tonsignale in die digitale Ebene überführt und anschließend unrelevante Signalanteile entfernt. Der daraus resultierende Datenstrom benötigt weniger Bandbreite bei der Übertragung zum Endkunden. An der Studie nahmen 39 Patienten teil, bei denen die Therapie bis zu vier Jahre zurückliegt. Neben einer Gesamtbetrachtung wird der Erfolg auch in Abhängigkeit von Alter, Geschlecht und sportlicher Aktivität analysiert. Insgesamt weist die Studie auf eine anhaltende Verbesserung des Gesundheitszustands mit zum Teil deutlicher Schmerzlinderung auch noch nach vier Jahren hin, jedoch mit einer leichten Schmerzzunahme gegen Ende des Untersuchungszeitraums von vier Jahren. Abstract: Based upon a questionnaire for patient-queries regarding pain-sensation and activity-restrictions during everyday task, the long-term effect of MBST® nuclear magnetic resonance therapy is studied in osteoarthritis of the knee. 39 patients, whose therapy dates back up to four years, participated in the study. In addition to an overall view, the success of this therapy is also analyzed regarding age, gender and physical activities. Overall, the study pointed out a sustained improvement in the patients´ local health-status with a significant pain-relief even after four years, but also with a slight increase in pain towards the end of that four-year period. There tends to be more positive effect on women, on elderly of also on even non-sports-active patients which suggest a possible influence by daily (over)-stressing on the success of the therapy. An additional positive effect of the therapy on bone density is also conceivable, however, this remains open. Based on a dynamic and domain-specific knowledge ontology, different types of design rules formalize this knowledge in a graph-based form. An expressive visual language provides a user-friendly, human readable representation. Finally, a consistency analysis tool enables conceptual designs to be checked against this formal conceptual knowledge. In this article, we concentrate on the knowledge specification part. Manually identified spindle annotations in EEG recordings suffer from substantial intra- and inter-rater variability, even if raters have been highly trained, which reduces the reliability of spindle measures as a research and diagnostic tool. The Massive Online Data Annotation (MODA) project has recently addressed this problem by forming a consensus from multiple such rating experts, thus providing a corpus of spindle annotations of enhanced quality. Based on this dataset, we present a U-Net-type deep neural network model to automatically detect sleep spindles. Our model’s performance exceeds that of the state-of-the-art detector and of most experts in the MODA dataset. We observed improved detection accuracy in subjects of all ages, including older individuals whose spindles are particularly challenging to detect reliably. Our results underline the potential of automated methods to do repetitive cumbersome tasks with super-human performance. Herein, an overview on the currently used sterilization processes in the global market is provided. Different sterilization techniques are grouped under a category that describes the method of treatment: radiation (gamma, electron beam, X-ray, and ultraviolet), thermal (dry and moist heat), and chemical (ethylene oxide, ozone, chlorine dioxide, and hydrogen peroxide). For each sterilization process, the typical process parameters as defined by regulations and the mode of antimicrobial activity are summarized. Finally, the recommended microorganisms that are used as biological indicators to validate sterilization processes in accordance with the rules that are established by various regulatory agencies are summarized. For this purpose, screen-printed carbon electrodes on the biodegradable substrates fibroin and PLA are modified with a glucose oxidase membrane and then encapsulated with the biocompatible material Ecoflex. The influence of different curing parameters of the carbon electrodes on the resulting biosensor characteristics is studied. The morphology of the electrodes is investigated by scanning electron microscopy, and the biosensor performance is examined by amperometric measurements of glucose (0.5–10 mM) in phosphate buffer solution, pH 7.4, at an applied potential of 1.2 V versus a Ag/AgCl reference electrode. Instead of Ecoflex, fibroin, PLA, and wound adhesive are tested as alternative encapsulation compounds: a series of swelling tests with different fibroin compositions, PLA, and Ecoflex has been performed before characterizing the most promising candidates by chronoamperometry. Therefore, the carbon electrodes are completely covered with the particular encapsulation material. For this purpose, a thin-film magnesium-based meander-like electrode is deposited onto a flexible, bioabsorbable substrate (fibroin or PLA) and encapsulated (passivated) by additional magnesium oxide layers on top and below the magnesium-based electrode. The morphology of different layered RTDs is analyzed by scanning electron microscopy. The sensor performance and lifetime of the RTD is characterized both under ambient atmospheric conditions between 30°C and 43°C, and wet tissue-like conditions with a constant temperature regime of 37°C. The latter triggers the degradation process of the magnesium-based layers. The 3-layers RTDs on a PLA substrate could achieve a lifetime of 8.5 h. In this study, a field-effect SiO2-gate electrolyte-insulator-semiconductor (EIS) sensor was utilized for the label-free electrostatic detection of tobacco mosaic virus (TMV) particles as a model plant pathogen. The capacitive EIS sensor has been characterized regarding its TMV sensitivity by means of constant-capacitance method. The EIS sensor was able to detect biotinylated TMV particles from a solution with a TMV concentration as low as 0.025 nM. A good correlation between the registered EIS sensor signal and the density of adsorbed TMV particles assessed from scanning electron microscopy images of the SiO2-gate chip surface was observed. Additionally, the isoelectric point of the biotinylated TMV particles was determined via zeta potential measurements and the influence of ionic strength of the measurement solution on the TMV-modified EIS sensor signal has been studied. Especially for seasonal storage, power-to-gas can be a promising approach. Biologically produced methane from hydrogen produced from surplus electricity can be used to substitute natural gas in the existing infrastructure. Current reactor types are not or are poorly optimized for flexible methanation. Therefore, this work proposes a new reactor type with a plug flow reactor (PFR) design. Simulations in COMSOL Multiphysics ® showed promising properties for operation in laminar flow. An experiment was conducted to support the simulation results and to determine the gas fraction of the novel reactor, which was measured to be 29%. Based on these simulations and experimental results, the reactor was constructed as a 14 m long, 50 mm diameter tube with a meandering orientation. Data processing was established, and a step experiment was performed. In addition, a kLa of 1 h−1 was determined. The results revealed that the experimental outcomes of the type of flow and gas fractions are in line with the theoretical simulation. The new design shows promising properties for flexible methanation and will be tested. In contrast to Stute and Zhu’s approach (2002) Stute & Zhu (Scandinavian J Statist 29:535–545, 2002), a transformation is not required. Thus, any problems associated with non-parametric regression estimation are avoided. As a result, the MBB method is much easier for users to implement. To illustrate the power of the MBB based tests, a small simulation study is performed. A key parameter for the potential ground risk is the kinetic impact energy of the UAS. The kinetic energy depends on the impact velocity of the UAS and, therefore, on the aerodynamic drag and the weight during free fall. Hence, estimating the impact energy of a UAS requires an accurate drag estimation of the UAS in that state. The paper at hand presents the aerodynamic drag estimation of small-scale multirotor UAS. Multirotor UAS of various sizes and configurations were analysed with a fully unsteady Reynolds-averaged Navier–Stokes approach. These simulations included different velocities and various fuselage pitch angles of the UAS. The results were compared against force measurements performed in a subsonic wind tunnel and provided good consistency. Furthermore, the influence of the UAS`s fuselage pitch angle as well as the influence of fixed and free spinning propellers on the aerodynamic drag was analysed. Free spinning propellers may increase the drag by up to 110%, depending on the fuselage pitch angle. Increasing the fuselage pitch angle of the UAS lowers the drag by 40% up to 85%, depending on the UAS. The data presented in this paper allow for increased accuracy of ground risk assessments. TEE are semi-conductor devices that can provide heating and cooling via a heat pump effect without direct noise emissions and no refrigerant use. An efficiency evaluation of the optimal operating mode is carried out for different numbers of TEE, ambient temperatures, and heating loads. The influence of an additional heat recovery unit on system efficiency and an unevenly distributed heating demand are examined. The results show that TEE can provide heat at a coefficient of performance (COP) greater than one especially for small heating demands and high ambient temperatures. The efficiency increases with the number of elements in the system and is subject to economies of scale. The best COP exceeds six at optimal operating conditions. An additional heat recovery unit proves beneficial for low ambient temperatures and systems with few TEE. It makes COPs above one possible at ambient temperatures below 0 ∘C. The effect increases efficiency by maximal 0.81 (from 1.90 to 2.71) at ambient temperature 5 K below room temperature and heating demand Q˙h=100W but is subject to diseconomies of scale. Thermoelectric technology is a valuable option for electricity-based heat supply and can provide cooling and ventilation functions. A careful system design as well as an additional heat recovery unit significantly benefits the performance. This makes TEE superior to direct current heating systems and competitive to heat pumps for small scale applications with focus on avoiding noise and harmful refrigerants. Long-chain acyl-amino acids can serve as strong surfactants and emulsifiers, with application in cosmetic industries. Heterologous expression of these enzymes, however, is often hampered, limiting their use in industrial processes. Results We identified a novel mycobacterial aminoacylase gene from Mycolicibacterium smegmatis MKD 8, cloned and expressed it in Escherichia coli and Vibrio natriegens using the T7 overexpression system. The recombinant enzyme was prone to aggregate as inclusion bodies, and while V. natriegens Vmax™ could produce soluble aminoacylase upon induction with isopropyl β-d-1-thiogalactopyranoside (IPTG), E. coli BL21 (DE3) needed autoinduction with lactose to produce soluble recombinant protein. We successfully conducted a chaperone co-expression study in both organisms to further enhance aminoacylase production and found that overexpression of chaperones GroEL/S enhanced aminoacylase activity in the cell-free extract 1.8-fold in V. natriegens and E. coli. Eventually, E. coli ArcticExpress™ (DE3), which co-expresses cold-adapted chaperonins Cpn60/10 from Oleispira antarctica, cultivated at 12 °C, rendered the most suitable expression system for this aminoacylase and exhibited twice the aminoacylase activity in the cell-free extract compared to E. coli BL21 (DE3) with GroEL/S co-expression at 20 °C. The purified aminoacylase was characterized based on hydrolytic activities, being most stable and active at pH 7.0, with a maximum activity at 70 °C, and stability at 40 °C and pH 7.0 for 5 days. The aminoacylase strongly prefers short-chain acyl-amino acids with smaller, hydrophobic amino acid residues. Several long-chain amino acids were fairly accepted in hydrolysis as well, especially N-lauroyl-L-methionine. To initially evaluate the relevance of this aminoacylase for the synthesis of N-acyl-amino acids, we demonstrated that lauroyl-methionine can be synthesized from lauric acid and methionine in an aqueous system. Conclusion Our results suggest that the recombinant enzyme is well suited for synthesis reactions and will thus be further investigated. The chemical synthesis of acyl-amino acids is conventionally performed by the Schotten-Baumann reaction using fatty acyl chlorides, but aminoacylases have also been investigated for use in biocatalytic synthesis with free fatty acids. Aminoacylases and their properties are diverse; they belong to different peptidase families and show differences in substrate specificity and biocatalytic potential. Bacterial aminoacylases capable of synthesis have been isolated from Burkholderia, Mycolicibacterium, and Streptomyces. Although several proteases and peptidases from S. griseus have been described, no aminoacylases from this species have been identified yet. In this study, we investigated two novel enzymes produced by S. griseus DSM 40236ᵀ . We identified and cloned the respective genes and recombinantly expressed an α-aminoacylase (EC 3.5.1.14), designated SgAA, and an ε-lysine acylase (EC 3.5.1.17), designated SgELA, in S. lividans TK23. The purified aminoacylase SgAA was biochemically characterized, focusing on its hydrolytic activity to determine temperature- and pH optima and stabilities. The aminoacylase could hydrolyze various acetyl-amino acids at the Nα -position with a broad specificity regarding the sidechain. Substrates with longer acyl chains, like lauroyl-amino acids, were hydrolyzed to a lesser extent. Purified aminoacylase SgELA specific for the hydrolysis of Nε -acetyl-L-lysine was unstable and lost its enzymatic activity upon storage for a longer period but could initially be characterized. The pH optimum of SgELA was pH 8.0. While synthesis of acyl-amino acids was not observed with SgELA, SgAA catalyzed the synthesis of lauroyl-methionine. However, their chemical synthesis needs acyl chlorides and does not meet sustainability criteria. Thus, the identification of biocatalysts to develop greener synthesis routes is desirable. We describe a novel aminoacylase from Paraburkholderia monticola DSM 100849 (PmAcy) which was identified, cloned, and evaluated for its N-acyl-amino acid synthesis potential. Soluble protein was obtained by expression in lactose autoinduction medium and co-expression of molecular chaperones GroEL/S. Strep-tag affinity purification enriched the enzyme 16-fold and yielded 15 mg pure enzyme from 100 mL of culture. Biochemical characterization revealed that PmAcy possesses beneficial traits for industrial application like high temperature and pH-stability. A heat activation of PmAcy was observed upon incubation at temperatures up to 80 °C. Hydrolytic activity of PmAcy was detected with several N-acyl-amino acids as substrates and exhibited the highest conversion rate of 773 U/mg with N-lauroyl-L-alanine at 75 °C. The enzyme preferred long-chain acyl-amino-acids and displayed hardly any activity with acetyl-amino acids. PmAcy was also capable of N-acyl-amino acid synthesis with good conversion rates. The best synthesis results were obtained with the cationic L-amino acids L-arginine and L-lysine as well as with L-leucine and L-phenylalanine. Exemplarily, L-phenylalanine was acylated with fatty acids of chain lengths from C8 to C18 with conversion rates of up to 75%. Meanwhile, it has been recognized that the statistics of transitional stages such as pre-REM, found between Non-REM and REM, may hold additional insight into the physiology of sleep and are now under vivid investigation. We propose a classification system based on a simple neural network architecture that scores the classical stages as well as pre-REM sleep in mice. When restricted to the classical stages, the optimized network showed state-of-the-art classification performance with an out-of-sample F1 score of 0.95 in male C57BL/6J mice. When unrestricted, the network showed lower F1 scores on pre-REM (0.5) compared to the classical stages. The result is comparable to previous attempts to score transitional stages in other species such as transition sleep in rats or N1 sleep in humans. Nevertheless, we observed that the sequence of predictions including pre-REM typically transitioned from Non-REM to REM reflecting sleep dynamics observed by human scorers. Our findings provide further evidence for the difficulty of scoring transitional sleep stages, likely because such stages of sleep are under-represented in typical data sets or show large inter-scorer variability. We further provide our source code and an online platform to run predictions with our trained network. That’s mainly because of a lack of suitable transparent investigation tunnels that are needed to apply optical methods. The paper proposes a procedure to make precise complex hollow structures from a highly transparent material using masters from generative or Rapid Prototyping processes. Taking two examples from the medical field, the upper human airways and the bronchial tree, the entire process is shown. The 3D Printing build process is illustrated as well as the silicon casting process. Finally the measuring equipment is demonstrated and sample results are given. Ein Anwendungs-Szenario wurde gleich mitgeliefert. Diese Technologie würde es möglich machen, die gesamte Ersatzteilversorgung der Amerikanischen Pazifikflotte mittels ein paar dieser Maschinen, umfangreicher Datenstätze und genügend Rohmaterial vor Ort auf einem Flugzeugträger direkt nach Bedarf zu fertigen. Diese Vorstellung definierte schon damals die direkte digitale Fertigung, das Rapid Manufacturing. In der Realität bestanden die mit diesem Verfahren hergestellten Bauteile nur aus Kunststoff, waren ungenau, bruchempfindlich und klebrig und allein in der Produktentwicklung, eben als Prototypen zu benutzen. Sie waren schnell verfügbar, weil zu Ihrer Herstellung keine Werkzeuge benötigt wurden. Folgerichtige und zudem modern hießen sie: Rapid Prototyping. Rapid Prototyping wurde schnell zum Synonym eines neuen Zweiges der Fertigungstechnik, der Generativen Fertigungstechnik. Die weitere Entwicklung brachte neue Verfahren, höhere Genauigkeiten, verbesserte Werkstoffe und neue Anwendungen. Die Herstellung von Negativen, also Werkzeugen, mit dem gleichen Verfahren wurde marketing-getrieben Rapid Tooling genannt und als die ersten Bauteile nicht mehr als Prototypen, sondern als Endprodukte eingesetzt wurden, nannte man dies Rapid Manufacturing - das Ziel war erreicht. War das Ziel wirklich erreicht? Ist es Rapid Manufacturing, wenn ein generativ gefertigtes Bauteil die gewünschte Spezifikation erreicht? N2 - As a process called stereolithography and a stereolithography apparatus (SLA) was presented in 1987, the dream of manufacturing any three-dimensional component directly from computer data and without component-specific tools became reality. An application scenario was supplied at the same time. This technology would make it possible to produce the entire spare parts requirement of the American Pacific Fleet merely through the use of a couple of such machines, extensive datasets and enough raw material on board an aircraft carrier directly as required. This image defined direct digital fabrication, rapid manufacturing, even at that time. In reality, this procedure only managed to produce components in plastic which were imprecise, fragile and sticky and only usable as prototypes in product development. They were rapidly available, because no tools were required for their manufacture. Consequentially, they are now known as Rapid Prototyping in modern jargon. Rapid Prototyping quickly became a synonym for a new branch of production engineering known as generative production engineering. Continued development brought new processes, improved accuracy, improved materials and new applications. The manufacturing of negatives, in other words tools, using the same procedure was quickly named rapid tooling by the marketing sector, and once the first components were used as final products instead of just prototypes the process was renamed "rapid manufacturing" - the goal had been reached. Was the goal really reached? Is it rapid manufacturing if a generatively manufactured component reaches the required specifications? What has to happen so that the rapid prototyping phenomenon becomes a strategy which is suitable for enabling the paradigm change from current manufacture-induced mass production of mass articles to consumer-induced (and consumer-responsible) mass production of single parts for anyone, and in all possibility makes dramatic changes in our way of working and living? This power-matching problem can be solved with a balanced hybrid propulsion system. However, there is a trade-off between wing loading, power loading, the level of hybridization, as well as range and take-off distance. An optimization method can vary design variables in such a way that a minimum of a particular objective is attained. In this paper, a comparison between the optimization results for minimum mass, minimum consumed primary energy, and minimum cost is conducted. A new initial sizing algorithm for general aviation aircraft with hybrid-electric propulsion systems is applied. This initial sizing methodology covers point performance, mission performance analysis, the weight estimation process, and cost estimation. The methodology is applied to the design of a STOL general aviation aircraft, intended for on-demand air mobility operations. The aircraft is sized to carry eight passengers over a distance of 500 km, while able to take off and land from short airstrips. Both halophilic bacteria Pontibacillus marinus DSM 16465ᵀ and Alkalibacillus haloalkaliphilus DSM 5271ᵀ and both halotolerant bacteria Metabacillus indicus DSM 16189 and Litchfieldia alkalitelluris DSM 16976ᵀ served as a source for the four new subtilisins SPPM, SPAH, SPMI and SPLA. The protease genes were cloned and expressed in Bacillus subtilis DB104. Purification to apparent homogeneity was achieved by ethanol precipitation, desalting and ion-exchange chromatography. Enzyme activity could be observed between pH 5.0–12.0 with an optimum for SPPM, SPMI and SPLA around pH 9.0 and for SPAH at pH 10.0. The optimal temperature for SPMI and SPLA was 70 °C and for SPPM and SPAH 55 °C and 50 °C, respectively. The protease gene was cloned and expressed in Bacillus subtilis DB104. The recombinant protease SPAO with 269 amino acids belongs to the subfamily of high-alkaline subtilisins. The biochemical characteristics of purified SPAO were analyzed in comparison with subtilisin Carlsberg, Savinase, and BPN'. SPAO, a monomer with a molecular mass of 27.1 kDa, was active over a wide range of pH 6.0–12.0 and temperature 20–80 °C, optimally at pH 9.0–9.5 and 55 °C. The protease is highly oxidatively stable to hydrogen peroxide and retained 58% of residual activity when incubated at 10 °C with 5% (v/v) H2O2 for 1 h while stimulated at 1% (v/v) H2O2. Furthermore, SPAO was very stable and active at NaCl concentrations up to 5.0 m. This study demonstrates the potential of SPAO for biotechnological applications in the future. Biotechnological applications include use in detergents, cosmetics, leather processing, wastewater treatment and pharmaceuticals. To identify a possible candidate for the enzyme market, here we cloned the gene of the subtilisin SPFA from Fictibacillus arsenicus DSM 15822ᵀ (obtained through a data mining-based search) and expressed it in Bacillus subtilis DB104. After production and purification, the protease showed a molecular mass of 27.57 kDa and a pI of 5.8. SPFA displayed hydrolytic activity at a temperature optimum of 80 °C and a very broad pH optimum between 8.5 and 11.5, with high activity up to pH 12.5. SPFA displayed no NaCl dependence but a high NaCl tolerance, with decreasing activity up to concentrations of 5 m NaCl. The stability enhanced with increasing NaCl concentration. Based on its substrate preference for 10 synthetic peptide 4-nitroanilide substrates with three or four amino acids and its phylogenetic classification, SPFA can be assigned to the subgroup of true subtilisins. Moreover, SPFA exhibited high tolerance to 5% (w/v) SDS and 5% H₂O₂ (v/v). The biochemical properties of SPFA, especially its tolerance of remarkably high pH, SDS and H₂O₂, suggest it has potential for biotechnological applications. The rotor blades of a wind turbine are typically made from composite material, which withstands high forces during rotation. The huge dimensions of the rotor blades complicate the inspection processes in manufacturing. The automation of inspection processes has a great potential to increase the overall productivity and to create a consistent reliable database for each individual rotor blade. The focus of this paper is set on the process of rotor blade inspection automation by utilizing an autonomous mobile manipulator. The main innovations include a novel path planning strategy for zone-based navigation, which enables an intuitive right-hand or left-hand driving behavior in a shared human–robot workspace. In addition, we introduce a new method for surface orthogonal motion planning in connection with large-scale structures. An overall execution strategy controls the navigation and manipulation processes of the long-running inspection task. The implemented concepts are evaluated in simulation and applied in a real-use case including the tip of a rotor blade form.
