Jr. A1 - Schöning, Michael Josef T1 - Associating biosensing properties with the morphological structure of multilayers containing carbon nanotubes on field-effect devices JF - Physica Status Solidi (A) N2 - The control of molecular architecture provided by the layer-by-layer (LbL) technique has led to enhanced biosensors, in which advantageous features of distinct materials can be combined. Full optimization of biosensing performance, however, is only reached if the film morphology is suitable for the principle of detection of a specific biosensor. In this paper, we report a detailed morphology analysis of LbL films made with alternating layers of single-walled carbon nanotubes (SWNTs) and polyamidoamine (PAMAM) dendrimers, which were then covered with a layer of penicillinase (PEN). An optimized performance to detect penicillin G was obtained with 6-bilayer SWNT/PAMAM LbL films deposited on p-Si-SiO2-Ta2O5 chips, used in biosensors based on a capacitive electrolyte-insulator-semiconductor (EIS) and a light-addressable potentiometric sensor (LAPS) structure, respectively. Field-emission scanning electron microscopy (FESEM) and atomic force microscopy (AFM) images indicated that the LbL films were porous, with a large surface area due to interconnection of SWNT into PAMAM layers. This morphology was instrumental for the adsorption of a larger quantity of PEN, with the resulting LbL film being highly stable. The experiments to detect penicillin were performed with constant-capacitance (ConCap) and constant-current (CC) measurements for EIS and LAPS sensors, respectively, which revealed an enhanced detection signal and sensitivity of ca. 100 mV/decade for the field-effect sensors modified with the PAMAM/SWNT LbL film. It is concluded that controlling film morphology is essential for an enhanced performance of biosensors, not only in terms of sensitivity but also stability and response time. Direct exploration of such extraterrestrial aquatic ecosystems requires advanced access and sampling technologies with a high level of autonomy. A new technological approach has been developed as part of the collaborative research project Enceladus Explorer (EnEx). The concept is based upon a minimally invasive melting probe called the IceMole. The force-regulated, heater-controlled IceMole is able to travel along a curved trajectory as well as upwards. Hence, it allows maneuvers which may be necessary for obstacle avoidance or target selection. Maneuverability, however, necessitates a sophisticated on-board navigation system capable of autonomous operations. The development of such a navigational system has been the focal part of the EnEx project. The original IceMole has been further developed to include relative positioning based on in-ice attitude determination, acoustic positioning, ultrasonic obstacle and target detection integrated through a high-level sensor fusion. Transmitted and elastic scattering intensity of the microspheres (radius 500 μm, refractive index 1.77) on an optical fiber half coupler is analyzed at 1510 nm. The 0.43 nm angular mode spacing of the resonances is comparable to the angular mode spacing value estimated using the optical size of the microsphere. The spectral linewidths of the resonances are in the order of 0.01 nm, which corresponds to quality factors of approximately 105. A polydopamine layer is used as a functionalizing agent on sapphire microspherical resonators in view of biosensor implementation. The varying layer thickness on the microsphere is determined as a function of the resonance wavelength shift. It is shown that polymer functionalization has a minor effect on the quality factor. This is a promising step toward the development of an optical biosensor. Consequently, within the coal industry, there is a growing recognition of the potential of microbial applications in fostering innovative technologies. Microbial-based coal solubilization, coal beneficiation, and coal dust suppression are green alternatives to traditional thermochemical and leaching technologies and better meet the need for ecologically sound and economically viable choices. Surfactant-mediated approaches have emerged as powerful tools for modeling, simulation, and optimization of coal-microbial systems and continue to gain prominence in clean coal fuel production, particularly in microbiological co-processing, conversion, and beneficiation. Surfactants (surface-active agents) are amphiphilic compounds that can reduce surface tension and enhance the solubility of hydrophobic molecules. A wide range of surfactant properties can be achieved by either directly influencing microbial growth factors, stimulants, and substrates or indirectly serving as frothers, collectors, and modifiers in the processing and utilization of coal. This review highlights the significant biotechnological potential of surfactants by providing a thorough overview of their involvement in coal biodegradation, bioprocessing, and biobeneficiation, acknowledging their importance as crucial steps in coal consumption. Therefore, the domain of easy-read and plain language is currently restricted to the bare minimum of necessary information. Thus, even though there is a tendency to broaden the domain of easy-read and plain language, the inaccessibility of a significant amount of textual information excludes the target audience from partaking or entertainment and restricts their ability to live life autonomously. Large language models can solve a vast variety of natural language tasks, including the simplification of standard language texts to easy-read or plain language. Moreover, with the rise of generative models like GPT, easy-read and plain language may be applicable to all kinds of natural language texts, making formerly inaccessible information accessible to marginalized groups like, a.o., non-native speakers, and people with mental disabilities. In this paper, we argue for the feasibility of text simplification and generation in that context, outline the ethical dimensions, and discuss the implications for researchers in the field of ethics and computer science. In this work we established a cell-based gas biosensor for the detection of toxic substances in air, by adapting a commercial sensor chip (Bionas®), previously used for the measurement of pollutants in liquids. Cells of the respiratory tract (A549, RPMI 2650, V79), which survive at a gas phase in a natural context, are used as biological receptors. The physiological cell parameters acidification, respiration and morphology are continuously monitored in parallel. Ammonia was used as a highly water-soluble model gas to test the feasibility of the sensor system. Infrared measurements confirmed the sufficiency of the medium draining method. This sensor system provides a basis for many sensor applications such as environmental monitoring, building technology and public security. Previously, we presented computational analysis of the blood pump flow. From the analysis, we were able to identify regions of high shear and recirculating flow that may cause blood damage, for example, deformation and fragmentation of the red blood cell (RBC). This mechanical hemolysis can be predicted using a tensor-based blood damage model that is based on the physical properties of the RBCs, for example, the relaxation time of the RBC membrane. However, an extensive and detailed analysis was complicated by the fact that the previous method predicts hemolysis along a finite number of pathlines traversed by the RBCs, possibly omitting parts of the flow domain. Furthermore, it is computationally expensive and is not easily parallelizable. Here, we propose a new method to estimate hemolysis. The method is based on treating the shape of droplet (tensor) as a field variable, like velocity in the Navier-Stokes system. The governing equation for the RBC shape is treated by least-squares finite element method and the volume conservation of the RBC is augmented by Lagrangian multiplier. Unlike the previous method, the proposed method can visualize areas of high RBC strain that is potentially dangerous for mechanical hemolysis. Also, the amount of plasma-free hemoglobin and, consequently, normalized index of hemolysis can be computed as a byproduct. The method is tested in a simple shear flow for validation and an artery graft flow is chosen to show its potential usefulness. We study monolingual and multilingual open-source text embeddings to reliably detect sexism and misogyny in Germanlanguage online comments from an Austrian newspaper. We observed classifiers trained on text embeddings to mimic closely the individual judgements of human annotators. Our method showed robust performance in the GermEval 2024 GerMS-Detect Subtask 1 challenge, achieving an average macro F1 score of 0.597 (4th place, as reported on Codabench). It also accurately predicted the distribution of human annotations in GerMS-Detect Subtask 2, with an average Jensen-Shannon distance of 0.301 (2nd place). The computational efficiency of our approach suggests potential for scalable applications across various languages and linguistic contexts. Sapphire chips, beads, and powder were first modified with (3-aminopropyl) triethoxysilane (APTES), followed by succinic anhydride (SA), and finally single-stranded probe DNA was EDC coupled to the functionalized layer. The presence of the APTES-SA layer on sapphire powders was confirmed by thermogravimetric analyis and Fourier-transform infrared spectroscopy. Using planar sapphire chips as substrates and X-ray photoelectron spectroscopy (XPS) as surface-sensitive tool, the sequence of individual layers was analyzed with respect to their chemical state, enabling the quantification of areal densities of the involved molecular units. Fluorescence microscopy was used to demonstrate the hybridization of fluorescently tagged target DNA to the probe DNA, including denaturation- and re-hybridization experiments. Due to its high thermal conductivity, synthetic sapphire is especially suitable as a chip material for the heat-transfer method, which was employed to distinguish complementary- and non-complementary DNA duplexes containing single-nucleotide polymorphisms. Due to the impact of the surface resistance, both types of IDE structures display a non-linear behavior in low-ionic strength solutions. The experimental data were fitted to an electrical equivalent circuit and interpreted taking into account the surface-charge-governed properties. The effect of a charged polyelectrolyte layer electrostatically assembled onto the sensor surface on the surface resistance in solutions with different KCl concentration is studied. In case of the same electrode footprint, 3D-IDEs show a larger cell constant and a higher sensitivity to molecular adsorption than that of planar IDEs. The obtained results demonstrate the potential of 3D-IDEs as a new transducer structure for a direct label-free sensing of charged molecules. This paper deals with the comparison of an isokinematic (motion with constant velocity) and an isotonic (motion against constant weight) training algorithm. Both algorithms are designed for a robotic research platform consisting of a 3D force plate and a high payload industrial robot, which allows leg extension training with arbitrary six-dimensional motion trajectories. In the isokinematic as well as the isotonic training algorithm, individual paths are defined i n C artesian s pace by sufficient s upport p oses. I n t he i sotonic t raining s cenario, the trajectory is adapted to the measured force as the robot should only move along the trajectory as long as the force applied by the user exceeds a minimum threshold. In the isotonic training scenario however, the robot’s acceleration is a function of the force applied by the user. To validate these findings, a simulative experiment with a simple linear trajectory is performed. For this purpose, the same force path is applied in both training scenarios. The results illustrate that the algorithms differ in the force dependent trajectory adaption. However, the risk of training-induced injuries requires the continuous monitoring and controlling of the forces applied to the musculoskeletal system as well as the velocity along the motion trajectory and the range of motion. In this paper, an adaptive norm-optimal iterative learning control algorithm to minimize the knee joint loadings during the leg extension training with an industrial robot is proposed. The response of the algorithm is tested in simulation for patients with varus, normal and valgus alignment of the knee and compared to the results of a higher-order iterative learning control algorithm, a robust iterative learning control and a recently proposed conventional norm-optimal iterative learning control algorithm. Although significant improvements in performance are made compared to the conventional norm-optimal iterative learning control algorithm with a small learning factor, for the developed approach as well as the robust iterative learning control algorithm small steady state errors occur. Thus, continuous monitoring and controlling of the loadings applied to the musculoskeletal system along the motion trajectory is required. In this paper, a norm-optimal iterative learning control algorithm for the robot-assisted training is developed. The algorithm aims at minimizing the external knee joint moment, which is commonly used to quantify the loading of the medial compartment. To estimate the external knee joint moment, a musculoskeletal lower extremity model is implemented in OpenSim and coupled with a model of an industrial robot and a force plate mounted at its end-effector. The algorithm is tested in simulation for patients with varus, normal and valgus alignment of the knee. The results show that the algorithm is able to minimize the external knee joint moment in all three cases and converges after less than seven iterations.
