Light-addressable potentiometric sensors for quantitative spatial imaging of chemical species
(2017)
A light-addressable potentiometric sensor (LAPS) is a semiconductor-based chemical sensor, in which a measurement site on the sensing surface is defined by illumination. This light addressability can be applied to visualize the spatial distribution of pH or the concentration of a specific chemical species, with potential applications in the fields of chemistry, materials science, biology, and medicine. In this review, the features of this chemical imaging sensor technology are compared with those of other technologies. Instrumentation, principles of operation, and various measurement modes of chemical imaging sensor systems are described. The review discusses and summarizes state-of-the-art technologies, especially with regard to the spatial resolution and measurement speed; for example, a high spatial resolution in a submicron range and a readout speed in the range of several tens of thousands of pixels per second have been achieved with the LAPS. The possibility of combining this technology with microfluidic devices and other potential future developments are discussed.
The invention pertains to a CellDrum electrode arrangement for measuring mechanical stress, comprising a mechanical holder (1 ) and a non-conductive membrane (4), whereby the membrane (4) is at least partially fixed at its circumference to the mechanical holder (1), keeping it in place when the membrane (4) may bend due to forces acting on the membrane (4), the mechanical holder (1) and the membrane (4) forming a container, whereby the membrane (1) within the container comprises an cell- membrane compound layer or biological material (3) adhered to the deformable membrane 4 which in response to stimulation by an agent may exert mechanical stress to the membrane (4) such that the membrane bending stage changes whereby the container may be filled with an electrolyte, whereby an electric contact (2) is arranged allowing to contact said electrolyte when filled into to the container, whereby within a predefined geometry to the fixing of the membrane (4) an electrode (7) is arranged, whereby the electrode (7) is electrically insulated with respect to the electric contact (2) as well as said electrolyte, whereby mechanical stress due to an agent may be measured as a change in capacitance.

Genetically humanized mice for proteins involved in drug metabolism and toxicity and mice engrafted with human hepatocytes are emerging as promising in vivo models for improved prediction of the pharmacokinetic, drug–drug interaction, and safety characteristics of compounds in humans. This is an overview on the genetically humanized and chimeric liver-humanized mouse models, which are illustrated with examples of their utility in drug metabolism and toxicity studies. The models are compared to give guidance for selection of the most appropriate model by highlighting advantages and disadvantages to be carefully considered when used for studies in drug discovery and development.
The capacitive electrolyte–insulator–semiconductor (EIS) structure is a typical device based on a field-effect sensor platform. With a simple silicon-based structure, EIS have been useful for several sensing applications, especially with incorporation of nanostructured films to modulate the ionic transport and the flat-band potential. In this paper, we report on ion transport and changes in flat-band potential in EIS sensors made with layer-by-layer films containing poly(amidoamine) (PAMAM) dendrimer and single-walled carbon nanotubes (SWNTs) adsorbed on p-Si/SiO 2 /Ta 2 O 5 chips with an Al ohmic contact. The impedance spectra were fitted using an equivalent circuit model, from which we could determine parameters such as the double-layer capacitance. This capacitance decreased with the number of bilayers owing to space charge accumulated at the electrolyte–insulator interface, up to three PAMAM/SWNTs bilayers, after which it stabilized. The charge-transfer resistance was also minimum for three bilayers, thus indicating that this is the ideal architecture for an optimized EIS performance. The understanding of the influence of nanostructures and the fine control of operation parameters pave the way for optimizing the design and performance of new EIS sensors.

In the present work, surface functionalization of different sensor materials was studied. Organosilanes are well known to serve as coupling agent for biomolecules or cells on inorganic materials. 3-aminopropyltriethoxysilane (APTES) was used to attach microbiological spores time to an interdigitated sensor surface. The functionality and physical properties of APTES were studied on isolated sensor materials, namely silicon dioxide (SiO2) and platinum (Pt) as well as the combined material on sensor level. A predominant immobilization of spores could be demonstrated on SiO2 surfaces. Additionally, the impedance signal of APTES-functionalized biosensor chips has been investigated.
The methodological discourse of mixed-methods research offers general procedures to combine quantitative and qualitative methods for investigating complex fields of research such as higher education. However, integrating different methods still poses considerable challenges. To move beyond general recommendations for mixed-methods research, this chapter proposes to discuss methodological issues with respect to a particular research domain. Taking current studies on the transition to higher education as an example, the authors first provide an overview of the potentials and limitations of quantitative and qualitative methods in the research domain. Second, they show the need for a conceptual framework grounded in the theory of the research object to guide the integration of different methods and findings. Finally, an example study that investigates transition with regard to the interplay of the individual student and the institutional context serves to illustrate the guiding role of theory. The framework integrates different theoretical perspectives on transition, informs the selection of the research methods, and defines the nexus of the two strands that constitute the mixed-methods design. As the interplay of individual and context is of concern for teaching and learning in general, the example presented may be fruitful for the wider field of higher education research.
Development and Testing of a Low NOX Micromix Combustion Chamber for an Industrial Gas Turbine
(2017)
The Micromix combustion principle, based on cross-flow mixing of air and hydrogen, promises low emission applications in future gas turbines. The Micromix combustion takes place in several hundreds of miniaturized diffusion-type micro-flames. The major advantage is the inherent safety against flash-back and low NOx-emissions due to a very short residence time of reactants in the flame region. The paper gives insight into the Micromix design and scaling procedure for different energy densities and the interaction of scaling laws and key design drivers in gas turbine integration. Numerical studies, experimental testing, gas turbine integration and interface considerations are evaluated. The aerodynamic stabilization of the miniaturized flamelets and the resulting flow field, flame structure and NOx formation are analysed experimentally and numerically. The results show and confirm the successful adaption of the low NOx Micromix characteristics for a range of different nozzle sizes, energy densities and thermal power output.

Gearboxes are mechanical transmission systems that provide speed and torque conversions from a rotating power source. Being a central element of the drive train, they are relevant for the efficiency and durability of motor vehicles. In this work, we present a new approach for gearbox design: Modeling the design problem as a mixed-integer nonlinear program (MINLP) allows us to create gearbox designs from scratch for arbitrary requirements and—given enough time—to compute provably globally optimal designs for a given objective. We show how different degrees of freedom influence the runtime and present an exemplary solution.
Energy-efficient components do not automatically lead to energy-efficient systems. Technical Operations Research (TOR) shifts the focus from the single component to the system as a whole and finds its optimal topology and operating strategy simultaneously. In previous works, we provided a preselected construction kit of suitable components for the algorithm. This approach may give rise to a combinatorial explosion if the preselection cannot be cut down to a reasonable number by human intuition. To reduce the number of discrete decisions, we integrate laws derived from similarity theory into the optimization model. Since the physical characteristics of a production series are similar, it can be described by affinity and scaling laws. Making use of these laws, our construction kit can be modeled more efficiently: Instead of a preselection of components, it now encompasses whole model ranges. This allows us to significantly increase the number of possible set-ups in our model. In this paper, we present how to embed this new formulation into a mixed-integer program and assess the run time via benchmarks. We present our approach on the example of a ventilation system design problem.
In times of planned obsolescence the demand for sustainability keeps growing. Ideally, a technical system is highly reliable, without failures and down times due to fast wear of single components. At the same time, maintenance should preferably be limited to pre-defined time intervals. Dispersion of load between multiple components can increase a system’s reliability and thus its availability inbetween maintenance points. However, this also results in higher investment costs and additional efforts due to higher complexity. Given a specific load profile and resulting wear of components, it is often unclear which system structure is the optimal one. Technical Operations Research (TOR) finds an optimal structure balancing availability and effort. We present our approach by designing a hydrostatic transmission system.
