Molke als Nebenprodukt der Käseherstellung wurde lange Zeit als Abfall betrachtet. Bedingt durch ihren hohen BOD (biological oxygen demand) war die direkte Einleitung in Gewässer, aber auch der mikrobielle Abbau in Kläranlagen bedenklich. Falls eine Weiterverarbeitung der Molke stattfand, so geschah dies meist zu Molkepulver oder Proteinkonzentrat. Als Untersuchungen der Molkeproteine jedoch unter pharmazeutischen Gesichtspunkten interessante Eigenschaften nahelegten, stieg das Interesse am Bioprodukt Molke und ihren Proteinen an. So stehen beispielsweise für die Molkeproteine a-Lactalbumin (ala) und b-Lactoglobulin (blg) antibakterielle, anticancerogene und diverse andere physiologische Effekte in der Diskussion. Gegenwärtig finden meist Membranverfahren zur Aufreinigung von Molkeproteinen Anwendung. Als alternatives Verfahren wurde am Institut für Bioverfahrenstechnik in Kaiserslautern ein chromatographisches Verfahren entwickelt, bei dem natürliche Tonminerale zum Einsatz kamen. Nach chemischer und physikalischer Modifikation des Ausgangsmaterials durch den Hersteller Süd-Chemie wurden drei der Adsorber für nähere Untersuchungen zur Auftrennung von Molkeproteinen aus Molkekonzentrat herangezogen. Nach einer Cross-Flow-Filtration des Molkekonzentrats erfolgte die Aufreinigung der Molkeproteine in einem FPLC-System.
Cytochrom P450 sind Häm-Proteine, die zur Enzymklasse der Oxidoreduktasen (EC 1.14.xy) gehören. Eine wichtige Reaktion ist die Hydroxylierung nichtaktivierter C–H-Bindungen, die in technischen Systemen von großem Interesse ist. Durch die Verwendung von M-IDA-2-Partikeln ist eine direkte Aufreinigung mit gleichzeitiger Immobilisierung und die Applikation der Enzyme aus dem Zelllysat möglich. Damit ist das Verfahren mehr als fünf Stunden schneller als die konventionelle Chromatographie und mehr als 80 % der Aufreinigungszeit wird gespart. Mit dem isolierten nativen Enzym konnte die Plattformchemikalie 9,10-Dihydroxystearinsäure aus Ölsäure hergestellt werden. Unter anderem für die Kunststoffindustrie können aus diesem Produkt wichtige Monomere wie z. B. Azelainsäure hergestellt werden. Die Bildung des Produkts erfolgt in einem zweiphasigen Reaktionssystem an der Grenzfläche zwischen dem Öl und der wässrigen Phase als Feststoff. Um das immobilisierte Enzym aktiv in die obere Phase zu transportieren, wurde eine neue magnetische Mischvorrichtung entwickelt. Das Reaktionsprodukt wurde mit NMR, GC-MS und HPLC-MS analysiert und mit einem chemisch synthetisierten Standard von 9,10-Dihydroxystearinsäure verglichen. Derzeit werden Studien des immobilisierten Häms des Enzyms durchgeführt.
Die selektive Isolierung von Cephalosporin C (CPC) aus komplexen Fermentationssuspensionen unter Einsatz magnetischer Separation ist das Ziel dieser Arbeit. Das Verfahren wird im frühen Stadium der Aufarbeitung genutzt, um CPC zu stabilisieren und somit die Produktausbeute zu erhöhen. Als Adsorbersysteme für CPC wurden neben einem projektinternen magnetischen Material ND 10322, dessen Oberflächenladungen spezifisch für die Bindung des Zielmoleküls synthetisiert wurden, verschiedene kommerzielle Partikelsysteme verglichen. Es konnten massenspezifische Maximalbeladungen von 51 mg g⁻¹ erreicht werden. Weiterhin wurde die Stabilität von CPC untersucht. Unter optimalen Adsorptionsbedingungen kann CPC stabilisiert werden, so dass die Geschwindigkeitskonstante der Degradation des b-Lactam-Rings unter diesen Bedingungen unter 0,005 h⁻¹ liegt. Untersuchungen zur Wiederverwertbarkeit der neuen Adsorbers zeigten eine irreversible Bindung geringer CPC-Mengen nach dem ersten Einsatz. Nach zwölf Zyklen tritt eine irreversible Bindung von CPC ein, was zu einer signifikanten Reduktion der Adsorptionsfähigkeit führt. Die Anhäufung des CPC auf dem Adsorber konnte durch IR-Untersuchungen auf die Bildung einer Peptidbindung zwischen Carboxylgruppen des CPC und Aminogruppe der Adsorberoberfläche zurückgeführt werden.
b-Lactame gehören zu den wirkungsvollsten Antibiotika, jedoch lassen sich viele nur schwierig fermentativ erzeugen. Ein Problem bei der fermentativen Produktion ist die Hydrolyse des Lactamrings im wässrigen Milieu. Das Ziel des von der DBU geförderten Projekts ist die selektive In-situ-Adsorption der b-Lactamantibiotika unter anschließender magnetischer Separation. Durch die Isolation im Hochgradientenmagnetseparator (HGMS) ist eine Fest-fest-flüssig-Trennung und somit ein erheblicher Zeitgewinn im Downstreamprozess möglich. Zusätzlich kommt es zur Einsparung an Lösungsmittel und Energie, was neben Reduzierung der Antibiotikahydrolyse auch in ökologischer Hinsicht einen interessanten Aspekt darstellt. Als Trägermaterial für die Adsorbermatrix werden magnetisierbare Eisenoxidpartikel eingesetzt, die in einer Silikamatrix eingebettet sind. Diese Adsorber sollen auf Selektivität in Wasser und verschiedenen Medien getestet werden. Zusätzlich werden die Abbauprodukte des b-Lactams analysiert, um eine Aussage über die Stabilisierung des Antibiotikums durch die selektiven Adsorber treffen zu können. Diese Ergebnisse werden mit kommerziell erhältlichen Adsorbern verglichen. Die Aufreinigung der Antibiotika soll direkt aus der Fermentationsbrühe erfolgen. Um die Trennung der magnetischen, selektiven Adsorber von der Biomasse zu gewährleisten, soll der HGMS in die Fermentation integriert werden. Das filamentöse Wachstum des Mikroorganismus erfordert eine Neuauslegung der Filtermatrix.
Aufarbeitung von Polyphenolen aus Weizenmittels Zeolithen am Beispiel der Ferulasa¨ ureAlexander Thiel1, Kai Muffler1, Nils Tippko¨ tter1, Kirstin Suck2, Ulrich Sohling2, Friedrich Ruf3und Roland Ulber1,*DOI: 10.1002/cite.201400031Bei der Ferulasa¨ure handelt es sich um einen Wertstoff, der aus Weizen gewonnen und in der Lebensmittel- und Pharma-industrie eingesetzt werden kann. Der Einsatz von Weizen als nachwachsende Rohstoffquelle ist allerdings nur dann wirt-schaftlich durchfu¨hrbar, wenn eine Prozessintegration in die bestehenden industriellen Verfahren gewa¨hrleistet oder einedirekte Konkurrenz zur Mehl- und Sta¨rkeindustrie vermieden werden kann. In diesem Artikel wird ein Verfahren aufge-zeigt, welches hohe Ausbeuten ermo¨glicht und eine Konkurrenz zu bestehenden Verwertungspfaden vermeidet.
The Monte Carlo code FLUKA is used to simulate the production of a number of positron emitting radionuclides, ¹⁸F, ¹³N, ⁹⁴Tc, ⁴⁴Sc, ⁶⁸Ga, ⁸⁶Y, ⁸⁹Zr, ⁵²Mn, ⁶¹Cu and ⁵⁵Co, on a small medical cyclotron with a proton beam energy of 13 MeV. Experimental data collected at the TR13 cyclotron at TRIUMF agree within a factor of 0.6 ± 0.4 with the directly simulated data, except for the production of ⁵⁵Co, where the simulation underestimates the experiment by a factor of 3.4 ± 0.4. The experimental data also agree within a factor of 0.8 ± 0.6 with the convolution of simulated proton fluence and cross sections from literature. Overall, this confirms the applicability of FLUKA to simulate radionuclide production at 13 MeV proton beam energy.
Air- and water-stable phenyl complexes with nitridotechnetium(V) cores can be prepared by straightforward procedures. [TcNPh2(PPh3)2] is formed by the reaction of [TcNCl2(PPh3)2] with PhLi. The analogous N-heterocyclic carbene (NHC) compound [TcNPh2(HLPh)2], where HLPh is 1,3,4-triphenyl-1,2,4-triazol-5-ylidene, is available from (NBu4)[TcNCl4] and HLPh or its methoxo-protected form. The latter compound allows the comparison of different Tc–C bonds within one compound. Surprisingly, the Tc chemistry with such NHCs does not resemble that of corresponding Re complexes, where CH activation and orthometalation dominate.
For the successful implementation of microfluidic reaction systems, such as PCR and electrophoresis, the movement of small liquid volumes is essential. In conventional lab-on-a-chip-platforms, solvents and samples are passed through defined microfluidic channels with complex flow control installations. The droplet actuation platform presented here is a promising alternative. With it, it is possible to move a liquid drop (microreactor) on a planar surface of a reaction platform (lab-in-a-drop). The actuation of microreactors on the hydrophobic surface of the platform is based on the use of magnetic forces acting on the outer shell of the liquid drops which is made of a thin layer of superhydrophobic magnetite particles. The hydrophobic surface of the platform is needed to avoid any contact between the liquid core and the surface to allow a smooth movement of the microreactor. On the platform, one or more microreactors with volumes of 10 µL can be positioned and moved simultaneously. The platform itself consists of a 3 x 3 matrix of electrical double coils which accommodate either neodymium or iron cores. The magnetic field gradients are automatically controlled. By variation of the magnetic field gradients, the microreactors' magnetic hydrophobic shell can be manipulated automatically to move the microreactor or open the shell reversibly. Reactions of substrates and corresponding enzymes can be initiated by merging the microreactors or bringing them into contact with surface immobilized catalysts.
We present a concise mini overview on the approaches to the disposal of nuclear waste currently used or deployed. The disposal of nuclear waste is the end point of nuclear waste management (NWM) activities and is the emplacement of waste in an appropriate facility without the intention to retrieve it. The IAEA has developed an internationally accepted classification scheme based on the end points of NWM, which is used as guidance. Retention times needed for safe isolation of waste radionuclides are estimated based on the radiotoxicity of nuclear waste. Disposal facilities usually rely on a multi-barrier defence system to isolate the waste from the biosphere, which comprises the natural geological barrier and the engineered barrier system. Disposal facilities could be of a trench type, vaults, tunnels, shafts, boreholes, or mined repositories. A graded approach relates the depth of the disposal facilities’ location with the level of hazard. Disposal practices demonstrate the reliability of nuclear waste disposal with minimal expected impacts on the environment and humans.
The use of transgenic animal models has transformed our knowledge of complex biochemical pathways in vivo. It has allowed disease processes to be modelled and used in the development of new disease prevention and treatment strategies. They can also be used to define cell- and tissue-specific pathways of gene regulation. A further major application is in the area of preclinical development where such models can be used to define pathways of chemical toxicity, and the pathways that regulate drug disposition. One major application of this approach is the humanisation of mice for the proteins that control drug metabolism and disposition. Such models can have numerous applications in the development of drugs and in their more sophisticated use in the clinic.
Multi-analyte biosensors may offer the opportunity to perform cost-effective and rapid analysis with reduced sample volume, as compared to electrochemical biosensing of each analyte individually. This work describes the development of an enzyme-based biosensor system for multi-parametric determination of four different organic acids. The biosensor array comprises five working electrodes for simultaneous sensing of ethanol, formate, d-lactate, and l-lactate, and an integrated counter electrode. Storage stability of the biosensor was evaluated under different conditions (stored at +4 °C in buffer solution and dry at −21 °C, +4 °C, and room temperature) over a period of 140 days. After repeated and regular application, the individual sensing electrodes exhibited the best stability when stored at −21 °C. Furthermore, measurements in silage samples (maize and sugarcane silage) were conducted with the portable biosensor system. Comparison with a conventional photometric technique demonstrated successful employment for rapid monitoring of complex media.
Die Ausbildung von Biofilmen in technischen Anlagen, wie z. B. Kühlkreisläufen, Wasseraufbereitungssystemen und Bioreaktoren, führen zu Materialschäden (Biofouling) und stark erhöhtem Energieaufwand. Im Rahmen der aktuellen Forschungsarbeiten erfolgen aktive sowie passive Bio-Modifikationen auf funktionalisierten magnetischen Mikropartikelober-flächen. Um die verschiedenen funktionalisierten magnetischen Mikropartikel zu analysieren und ihre antimikrobielle Wirkung zu testen, wird der Einsatz einer 3D-gedruckten, magnetischen Plattform für ein Fluoreszenz-basiertes Screening-System untersucht. Für den Oberflächenschutz wurden verschiedene, antimikrobiell funktionalisierte Partikelkombinationen mit dem Mikroorganismus Escherichia coli GFPmut2 in Bezug auf aktiven Oberflächenschutz verglichen. Um die antimikrobielle Oberflächeneffekte von synergistischen Kombinationen unterschiedlich funktionalisierter Partikel zu bestimmen, werden Oberflächen einem Magnetfeld ausgesetzt, das die Mikropartikel als definierte Schicht auf ihnen zurück hält. Diese modifizierten Oberflächen können sowohl durch Fluoreszenzspektroskopie als auch -mikroskopie analysiert werden.
In most beers, producers strive to minimize haze to maximize visual appeal. To detect the formation of particulates, a measurement system for sub-micron particles is required. Beer haze is naturally occurring, composed of protein or polyphenol particles; in their early stage of growth their size is smaller than 2 µm. Microscopy analysis is time and resource intensive; alternatively, backscattering is an inexpensive option for detecting particle sizes of interest.
Autoradiography is a well-established method of nuclear imaging. When different radionuclides are present simultaneously, additional processing is needed to distinguish distributions of radionuclides. In this work, a method is presented where aluminium absorbers of different thickness are used to produce images with different cut-off energies. By subtracting images pixel-by-pixel one can generate images representing certain ranges of β-particle energies. The method is applied to the measurement of irradiated reactor graphite samples containing several radionuclides to determine the spatial distribution of these radionuclides within pre-defined energy windows. The process was repeated under fixed parameters after thermal treatment of the samples. The greyscale images of the distribution after treatment were subtracted from the corresponding pre-treatment images. Significant changes in the intensity and distribution of radionuclides could be observed in some samples. Due to the thermal treatment parameters the most significant differences were observed in the ³H and ¹⁴C inventory and distribution.
The composition and physiochemical properties of aquatic-phase natural organic matter (NOM) are most important problems for both environmental studies and water industry. Laser desorption/ionization (LDI) mass spectrometry facilitated successful examinations of NOM, as humic and fulvic acids in NOM are readily ionized by the nitrogen laser. In this study, hydrophobic NOMs (HPO NOMs) from river, reservoir and waste water were characterized by this technique. The effect of analytical variables like concentration, solvent composition and laser energy was investigated. The exact masses of small molecular NOM moieties in the range of 200–1200 m/z were determined in reflectron mode. In addition, spectra of post-source-decay experiments in this range showed that some compounds from different natural NOMs had the same fragmental ions. In the large mass range of 1200–15 000 Da, macromolecules and their aggregates were found in HPO NOMs from natural waters. Highly humic HPO exhibited mass peaks larger than 8000 Da. On the other hand, the waste water and reservoir water mainly had relatively smaller molecules of about 2000 Da. The LDI-MS measurements indicated that highly humic river waters were able to form large aggregates and membrane foulants, while the HPO NOMs from waste water and reservoir water were unlikely to form large aggregates. Analyse von Lignocellulose mittels dynamischer Differenzkalorimetrie und Infrarot – Spektrometrie

This study investigated the anaerobic digestion of an algal–bacterial biofilm grown in artificial wastewater in an Algal Turf Scrubber (ATS). The ATS system was located in a greenhouse (50°54′19ʺN, 6°24′55ʺE, Germany) and was exposed to seasonal conditions during the experiment period. The methane (CH4) potential of untreated algal–bacterial biofilm (UAB) and thermally pretreated biofilm (PAB) using different microbial inocula was determined by anaerobic batch fermentation. Methane productivity of UAB differed significantly between microbial inocula of digested wastepaper, a mixture of manure and maize silage, anaerobic sewage sludge, and percolated green waste. UAB using sewage sludge as inoculum showed the highest methane productivity. The share of methane in biogas was dependent on inoculum. Using PAB, a strong positive impact on methane productivity was identified for the digested wastepaper (116.4%) and a mixture of manure and maize silage (107.4%) inocula. By contrast, the methane yield was significantly reduced for the digested anaerobic sewage sludge (50.6%) and percolated green waste (43.5%) inocula. To further evaluate the potential of algal–bacterial biofilm for biogas production in wastewater treatment and biogas plants in a circular bioeconomy, scale-up calculations were conducted. It was found that a 0.116 km2 ATS would be required in an average municipal wastewater treatment plant which can be viewed as problematic in terms of space consumption. However, a substantial amount of energy surplus (4.7–12.5 MWh a−1) can be gained through the addition of algal–bacterial biomass to the anaerobic digester of a municipal wastewater treatment plant. Wastewater treatment and subsequent energy production through algae show dominancy over conventional technologies.
Mit Palmöl beladene Bleicherde wurde auf die Anwendbarkeit als Rohstoff zur Bildung von Aceton, Ethanol und Butanol (ABE) untersucht. Nach der Abtrennung des entfärbten bzw. gebleichten Öls bestehen 20–40 % (w/w) des Bleicherderückstands aus adsorbiertem Öl. Somit stellt das Mineral einen wertvollen Rohstoff dar. Zur Produktion der Lösungsmittel wurde das gebundene Öl hydrolysiert. Das freigesetzte Glycerin diente als Substrat einer nachfolgenden anaeroben Fermentation zu den ABE-Lösungsmitteln. Die Fermentationen wurden mit verschiedenen Clostridien-Stämmen durchgeführt, von denen einige lipolytische Aktivität aufweisen. Der Ölgehalt der Bleicherde betrug 28 % der Adsorbermasse. Für die Hydrolyse wurden drei Ansätze untersucht:
1. die direkte Fermentation des Adsorbers mit lipolytisch aktiven Clostridien,
2. der Einsatz von Lipasen und die Fermentation des resultierenden Überstands und
3. die Kofermentation mit einer lipolytisch aktiven Hefe.
Jeder der drei Ansätze führte zu einerHydrolyse des Öls und Wachstum der Mikroorganismen. Der Einsatz von Lipasen resultierte in einer vollständigen und der schnellsten Hydrolyse des Öls. Eine Glycerinkonzentration von 13,4 g/L konnte erreicht werden. Die anaerobe Fermentation der verschiedenen Clostridien auf Minimalmedien verlief erfolgreich. Der Vergleich der Fermentationen der Bleicherdehydrolysate zeigte, dass das Hydrolysat wachstumsinhibierende Substanzen enthält. Unter diesen Bedingungen konnte eine Ausbeute von Y P/S = 0,11 erzielt werden. Der Einsatz eines Ionenaustauschers zur Reinigung des Hydrolysats vor der Fermentation resultierte in einer Verbesserung des Wachstums
Replacement tissues, designed to fill in articular cartilage defects, should exhibit the same properties as the native material. The aim of this study is to foster the understanding of, firstly, the mechanical behavior of the material itself and, secondly, the influence of cultivation parameters on cell seeded implants as well as on cell migration into acellular implants. In this study, acellular cartilage replacement material is theoretically, numerically and experimentally investigated regarding its viscoelastic properties, where a phenomenological model for practical applications is developed. Furthermore, remodeling and cell migration are investigated.
Die wachsende Produktpalette von z. B. Pharmazeutika geht mit einer steigenden Nachfrage für hochsensitive/schonende Aufreinigungstechniken einher. Bisherige Verfahren führen oft zu geringer Reinheit und verminderter Bioaktivität, zeigen eine Limitation der Analytengröße oder bedingen dessen Modifikation. Durch die Kombination von mikroskaligen Magnetpartikeln und spezifisch wechselwirkenden Einzelstrang-DNA-Oligonukleotiden, den sog. ssDNA-Aptameren, sind eine höhere Selektivität/Reinheit und eine Automatisierung möglich. In diesem Kontext werden zum einen ssDNA-Amplifikationstechniken und zum anderen der praktische Einsatz von Aptameren in einer Magnetseparation vorgestellt. Die ssDNA-Synthese basiert auf einem In-vivo-dsDNA-Produktionsschritt mittels eines rekombinanten Escherichia coli. Die als High-copy-Plasmid organisierte Sequenz wird in vitro durch Kombination verschiedener enzymatischer Reaktionen in die funktionelle ssDNA überführt. Diese Technik bedingt nur minimale Instrumentierung bzw. Prozessregelung. Die zweite Synthesetechnik wird in Form eines In-vitro-Amplifikationsverfahrens realisiert und beruht auf dem Prinzip einer PCR (Potenzial zu einer Automatisierung bzw. Miniaturisierung). Die gewonnenen Aptamere werden im Anschluss in einem auf Magnetpartikeln basierten Trennverfahren zur Isolationvon 6xHis-tag-Proteinen bezüglich ihrer Eigenschaften untersucht.
The interplay of albumin (BSA) and lysozyme (LYZ) adsorbed simultaneously on titanium was analyzed by gel electrophoresis and BCA assay. It was found that BSA and lysozyme adsorb cooperatively. Additionally, the isoelectric point of the respective protein influences the adsorption. Also, the enzymatic activity of lysozyme and amylase (AMY) in mixtures with BSA was considered with respect to a possible influence of protein-protein interaction on enzyme activity. Indeed, an increase of lysozyme activity in the presence of BSA could be observed. In contrast, BSA does not influence the activity of amylase.
