With the prevalence of glucosamine- and chondroitin-containing dietary supplements for people with osteoarthritis in the marketplace, it is important to have an accurate and reproducible analytical method for the quantitation of these compounds in finished products. NMR spectroscopic method based both on low- (80 MHz) and high- (500–600 MHz) field NMR instrumentation was established, compared and validated for the determination of chondroitin sulfate and glucosamine in dietary supplements. The proposed method was applied for analysis of 20 different dietary supplements. In the majority of cases, quantification results obtained on the low-field NMR spectrometer are similar to those obtained with high-field 500–600 MHz NMR devices. Validation results in terms of accuracy, precision, reproducibility, limit of detection and recovery demonstrated that the developed method is fit for purpose for the marketed products. The NMR method was extended to the analysis of methylsulfonylmethane, adulterant maltodextrin, acetate and inorganic ions. Low-field NMR can be a quicker and cheaper alternative to more expensive high-field NMR measurements for quality control of the investigated dietary supplements. High-field NMR instrumentation can be more favorable for samples with complex composition due to better resolution, simultaneously giving the possibility of analysis of inorganic species such as potassium and chloride.
Social Media - Segen oder Fluch? : Eine Aufklärungskampagne zu Auswirkungen von Social Media Nutzung

Das Projekt „Social Media – Segen oder Fluch“ beschäftigt sich mit negativen Auswirkungen, die die Nutzung Sozialer Medien haben kann. Soziale Medien werden heutzutage ständig und von fast jedem genutzt, vor allem von Jugendlichen und jungen Erwachsenen. Vielen ist aber nicht bewusst, dass sich die Nutzung negativ auf sie und ihre Gesundheit auswirken kann. Eine Kampagne soll daran etwas ändern. Soziale Medien bzw. der Content, der die Nutzer:innen negativ beeinflusst, ist wie ein Monster, vor dem sich die Nutzer:innen in acht nehmen müssen. Angesprochen werden hauptsächlich 12-29-jährige, die Soziale Medien nutzen. Die Kampagne soll durch verschiedene Maßnahmen wie Plakatwerbung und Posts auf Social Media Kanälen, den negativen Auswirkungen von Social Media Nutzung entgegenwirken. Sie soll über die Auswirkungen aufklären und zu einem bewussteren Umgang mit Sozialen Netzwerken anregen.
Amino acid-based surfactants are valuable compounds for cosmetic formulations. The chemical synthesis of acyl-amino acids is conventionally performed by the Schotten-Baumann reaction using fatty acyl chlorides, but aminoacylases have also been investigated for use in biocatalytic synthesis with free fatty acids. Aminoacylases and their properties are diverse; they belong to different peptidase families and show differences in substrate specificity and biocatalytic potential. Bacterial aminoacylases capable of synthesis have been isolated from Burkholderia, Mycolicibacterium, and Streptomyces. Although several proteases and peptidases from S. griseus have been described, no aminoacylases from this species have been identified yet. In this study, we investigated two novel enzymes produced by S. griseus DSM 40236ᵀ . We identified and cloned the respective genes and recombinantly expressed an α-aminoacylase (EC 3.5.1.14), designated SgAA, and an ε-lysine acylase (EC 3.5.1.17), designated SgELA, in S. lividans TK23. The purified aminoacylase SgAA was biochemically characterized, focusing on its hydrolytic activity to determine temperature- and pH optima and stabilities. The aminoacylase could hydrolyze various acetyl-amino acids at the Nα -position with a broad specificity regarding the sidechain. Substrates with longer acyl chains, like lauroyl-amino acids, were hydrolyzed to a lesser extent. Purified aminoacylase SgELA specific for the hydrolysis of Nε -acetyl-L-lysine was unstable and lost its enzymatic activity upon storage for a longer period but could initially be characterized. The pH optimum of SgELA was pH 8.0. While synthesis of acyl-amino acids was not observed with SgELA, SgAA catalyzed the synthesis of lauroyl-methionine.
Subtilisins from microbial sources, especially from the Bacillaceae family, are of particular interest for biotechnological applications and serve the currently growing enzyme market as efficient and novel biocatalysts. Biotechnological applications include use in detergents, cosmetics, leather processing, wastewater treatment and pharmaceuticals. To identify a possible candidate for the enzyme market, here we cloned the gene of the subtilisin SPFA from Fictibacillus arsenicus DSM 15822ᵀ (obtained through a data mining-based search) and expressed it in Bacillus subtilis DB104. After production and purification, the protease showed a molecular mass of 27.57 kDa and a pI of 5.8. SPFA displayed hydrolytic activity at a temperature optimum of 80 °C and a very broad pH optimum between 8.5 and 11.5, with high activity up to pH 12.5. SPFA displayed no NaCl dependence but a high NaCl tolerance, with decreasing activity up to concentrations of 5 m NaCl. The stability enhanced with increasing NaCl concentration. Based on its substrate preference for 10 synthetic peptide 4-nitroanilide substrates with three or four amino acids and its phylogenetic classification, SPFA can be assigned to the subgroup of true subtilisins. Moreover, SPFA exhibited high tolerance to 5% (w/v) SDS and 5% H₂O₂ (v/v). The biochemical properties of SPFA, especially its tolerance of remarkably high pH, SDS and H₂O₂, suggest it has potential for biotechnological applications.
Sustainability is playing an increasingly important role. Not least due to the definition of the sustainable development goals (SDGs) in the framework of the agenda 2030 by the United Nations (UN) in 2015 (United Nations, n.d.), it has become clear that the cooperation of different actors is needed to achieve the defined 17 goals. Industry, as a global actor, has a special role to play in this. In the course of sustainable production processes and chains, the industry is confronted with the responsibility of reflecting on the consequences of its own trade on an ecological, economic, and also social level and deriving measures that, according to the definition of sustainability (Hauff, 1987), will also enable future generations to satisfy their needs. While the ecological pillar of sustainability is already being addressed by different industrial initiatives (Deloitte, 2021), it is questionable to what extent the economic and, above all, the social pillars of sustainability also play a decisive role. Accordingly, it is questionable to what extent sustainability in its triad of social, ecological, and economic aspects is taken into account holistically at all, and thus to what extent the industry contributes to achieving the 17 goals defined by the UN.
This paper presents a qualitative study that explores these questions. Interviewing 31 representatives from the manufacturing industry in Germany, results indicate a Paradox of Sustainable Production expressed by a theoretical reflection of the need for focusing on people in production processes on the one hand and a lack of addressing the social pillar of sustainability in concepts on the other hand. However, while it is a troublesome finding given the striking need for sustainable development (The-Sustainable-Development-Goals-Report-2022; Kropp 2019; von Hauff 2021; Roy and Singh 2017), the paradox directly lays out a path of resolving it. This is because, given its nature, we can see that we could resolve it via the implementation of strong educational efforts trying to help the respective people of the manufacturing industry to understand the holistic and interdependent character of sustainable development (The-Sustainable-Development-Goals-Report-2022).
In Folge der Globalisierung und der Zuwanderung von verschiedensten ethnischen Gruppen und deren Kulturen entstehen in Deutschland immer mehr sogenannte Transkulturen. Um diesen mehr Aufmerksamkeit zu schenken und zur Verständigung verschiedenster Gruppen beizutragen, wurde das Konzept dieser Bachelorarbeit: „BRIDGE: Ein Magazin über Transkulturen in Deutschland" entwickelt, welches sich mit den Schnittstellen der Deutschen und anderen Kulturen auseinandersetzt. Diese Transkulturen sollen jeweils repräsentativ und aufklärend dargestellt werden. Jede Ausgabe wird eine dieser Transkulturen porträtieren und zeigen, wie diese in Deutschland besteht. Für die erste Ausgabe, welche im Rahmen der Bachelorarbeit ausgearbeitet wurde, fiel die Entscheidung auf die Deutsch-Habesha-Kultur, die eine Völkergruppe aus Äthiopien und Eritrea darstellt.
Im Umgang mit dem Schweizer Historiker Daniele Ganser lässt sich ein ungewöhnliches Phänomen beobachten. Während die Vorträge und Bücher des bekannten Publizisten bei zahlreichen Fans auf großes Interesse stoßen, zeichnen die meisten Medien das Bild eines gefährlichen Verschwörungstheoretikers. Schnell wird seine Person zum Symbolbild einer ganzen Gegenöffentlichkeit. Ein Dokumentarfilm über Daniele Ganser, seine Kritiker:innen und alternative Medien.
Kinder sind bei der Suche nach Antworten auf ihre Fragen in unserer medialen Gesellschaft stark mit dem digitalen Raum konfrontiert. Diese Medien können bei einer nicht kindergerechten Benutzung wichtige Entwicklungsprozesse hindern. Im Gegensatz dazu geben diese aber auch die Möglichkeit, Kindern wichtige Denkgewohnheiten beizubringen und sie besser auf die Zukunft vorzubereiten. Ziel der Arbeit ist es, eine Verbindung zwischen der analogen und digitalen Welt herzustellen und ein vielfältiges Lernen zu ermöglichen. Die App „miela“ schafft eine neue Ebene der Wissensvermittlung durch die Kombination von digitaler und analoger Welt. Das Userinterface ist altersentsprechend gestaltet und bietet unterschiedliche Themenfelder an, wobei die Wissensvermittlung und die Kreativität im Fokus stehen. Eine Zusammenarbeit mit der Toniebox als digitale Erweiterung kann weitere Themenfelder anbieten.
Seit Jahrhunderten tragen Hebammen die Verantwortung für neues Leben und geben ihre Erfahrungen und ihr Können weiter. Trotz dieser ehrwürdigen Aufgabe gehören sie zu einer kleinen Berufsgruppe, die viel zu oft unterschätzt, überlastet und unterbezahlt wird. Sie sind Begleiter*innen in einer der sensibelsten und emotionalsten Zeiten des Lebens. Die Unterstützung einer Hebamme gewährleistet Aufklärung und Beistand während der Schwangerschaft, der Geburt und der Zeit im Wochenbett.
Das Projekt »Die Hebamme« ist ein Berufsportrait. Die Arbeit gibt Einblicke in die Tätigkeit und seine verschiedenen Bereiche und erzählt dabei hauptsächlich in Bildern. Zusätzlich berichten Hebammen aus unterschiedlichen Blickwinkeln über ihre Arbeit. Dieses Portrait stellt die Realtität der Arbeit dar und soll die Bedeutung der Tätigkeit näher bringen.
Dass der Stress im Krankenhaus eine Belastung für das Klinikpersonal darstellt, ist eine allseits bekannte Tatsache. Das liegt an dem anstrengenden Klinikalltag, welcher die Mitarbeiter:innen auf Belastungsprobe stellt. Lange Dienste und Überstunden, wenig Pausen und hohe Arbeitsverdichtung, Nacht- und Schichtdienst, zudem auch die aufwendige Dokumentation und ̈Bürokratie. Timewise behandelt die Thematik der Dienstplanerstellung im Krankenhaus unter der Berücksichtigung der psychischen und physischen Gesundheit des Klinikpersonals im Nacht- und Schichtdienst. Essenzielle Funktionen sind kürzere Kommunikationswege und eine gute Übersicht über den Soll-Ist-Abgleich inklusive Überstunden, Minusstunden, Abwesenheiten und den monatlichen Lohn. Der einfache Schichttausch und die Option der Berücksichtigung individueller Präferenzen und Wünsche ermöglicht es dem Personal, die Arbeit und das private Leben besser miteinander vereinen zu können, was sich positiv auf die Arbeitszufriedenheit auswirkt.
