A future bio-economy should not only be based on renewable raw materials but also in the raise of carbon yields of existing production routes. Microbial electrochemical technologies are gaining increased attention for this purpose. In this study, the electro-fermentative production of biobutanol with C. acetobutylicum without the use of exogenous mediators is investigated regarding the medium composition and the reactor design. It is shown that the use of an optimized synthetic culture medium allows higher product concentrations, increased biofilm formation, and higher conductivities compared to a synthetic medium supplemented with yeast extract. Moreover, the optimization of the reactor system results in a doubling of the maximum product concentrations for fermentation products. When a working electrode is polarized at −600 mV vs. Ag/AgCl, a shift from butyrate to acetone and butanol production is induced. This leads to an increased final solvent yield of Yᴀᴃᴇ = 0.202 gg⁻¹ (control 0.103 gg⁻¹), which is also reflected in a higher carbon efficiency of 37.6% compared to 23.3% (control) as well as a fourfold decrease in simplified E-factor to 0.43. The results are promising for further development of biobutanol production in bioelectrochemical systems in order to fulfil the principles of Green Chemistry.
Bacterial cell appendix formation supports cell-cell interaction, cell adhesion and cell movement. Additionally, in bioelectrochemical systems (BES), cell appendages have been shown to participate in extracellular electron transfer. In this work, the cell appendix formation of Clostridium acetobutylicum in biofilms of a BES are imaged and compared with conventional biofilms. Under all observed conditions, the cells possess filamentous appendages with a higher number and density in the BES. Differences in the amount of extracellular polymeric substance in the biofilms of the electrodes lead to the conclusion that the cathode can be used as electron donor and the anode as electron acceptor by C. acetobutylicum. When using conductive atomic force microscopy, a current response of about 15 nA is found for the cell appendages from the BES. This is the first report of conductivity for clostridial cell appendices and represents the basis for further studies on their role for biofilm formation and electron transfer.
Paracoccus denitrificans for the effluent recycling during continuous denitrification of liquid food

Nitrate is an undesirable component of several foods. A typical case of contamination with high nitrate contents is whey concentrate, containing nitrate in concentrations up to 25 l. The microbiological removal of nitrate by Paracoccus denitrificans under formation of harmless nitrogen in combination with a cell retention reactor is described here. Focus lies on the resource-conserving design of a microbal denitrification process. Two methods are compared. The application of polyvinyl alcohol-immobilized cells, which can be applied several times in whey feed, is compared with the implementation of a two step denitrification system. First, the whey concentrate's nitrate is removed by ion exchange and subsequently the eluent regenerated by microorganisms under their retention by crossflow filtration. Nitrite and nitrate concentrations were determined by reflectometric color measurement with a commercially available Reflectoquant® device. Correction factors for these media had to be determined. During the pilot development, bioreactors from 4 to 250 mg·L-1 and crossflow units with membrane areas from 0.02 to 0.80 m2 were examined. Based on the results of the pilot plants, a scaling for the exemplary process of denitrifying 1,000 tons per day is discussed.
The development prospects of the world markets for petroleum and other liquid fuels are diverse and partly contradictory. However, comprehensive changes for the energy supply of the future are essential. Notwithstanding the fact that there are still very large deposits of energy resources from a geological point of view, the finite nature of conventional oil reserves is indisputable. To reduce our dependence on oil, the EU, the USA, and other major economic zones rely on energy diversification. For this purpose, alternative materials and technologies are being sought, and is most obvious in the transport sector. The objective is to progressively replace fossil fuels with renewable and more sustainable fuels. In this respect, biofuels have a pre-eminent position in terms of their capability of blending with fossil fuels and being usable in existing cars without substantial modification. Ethanol can be considered as the primary renewable liquid fuel. In this chapter enzymes, micro-organisms, and processes for ethanol production based on renewable resources are described.
In Anbetracht des zu erwartenden Rückgangs der Verfügbarkeit fossiler Rohstoffe müssen nicht nur für den Energiesektor, sondern auch für die Herstellung industrieller Produkte alternative Rohstoffe gefunden werden. Ein Beispiel für einen nicht in Nahrungsmittelkonkurrenz stehenden nachwachsenden Rohstoff ist grüne Biomasse wie Gras und Klee. Diese lassen sich in Deutschland auf großen Flächen anbauen und enthalten eine Vielzahl potenzieller Substrate für Fermentationen.
The invention relates to a method for production of single-stranded macronucleotides by amplifying and ligating an extended monomeric single-stranded target nucleic acid sequence (targetss) into a repetitive cluster of double-stranded target nucleic acid sequences (targetds), and subsequently cloning the construct into a vector (aptagene vector). The aptagene vector is transformed into host cells for replication of the aptagene and isolated in order to optain single-stranded target sequences (targetss). The invention also relates to single-stranded nucleic acids, produced by a method of the invention.
An immunochromatographic lateral flow dipstick assay for the fast detection of microcystin-LR was developed. Colloid gold particles with diameters of 40 nm were used as red-colored antibody labels for the visual detection of the antigen. The new dipstick sensor is capable of detecting down to 5 µg·l−1 (ppb; total inversion of the color signal) or 1 ppb (observation of color grading) of microcystin-LR. The course of the labeling reaction was observed via spectrometric wave shifts caused by the change of particle size during the binding of antibodies. Different stabilizing reagents showed that especially bovine serum albumin (BSA) and casein increase the assays sensitivity and the conjugate stability. Performance of the dipsticks was quantified by pattern processing of capture zone CCD images. Storage stability of dipsticks and conjugate suspensions over 115 days under different conditions were monitored. The ready-to-use dipsticks were successfully tested with microcystin-LR-spiked samples of outdoor drinking- and salt water and applied to the tissue of microcystin-fed mussels.
Biotechnological downstream processing is usually an elaborate procedure, requiring a multitude of unit operations to isolate the target component. Besides the disadvantageous space-time yield, the risks of cross-contaminations and product loss grow fast with the complexity of the isolation procedure. A significant reduction of unit operations can be achieved by application of magnetic particles, especially if these are functionalized with affinity ligands. As magnetic susceptible materials are highly uncommon in biotechnological processes, target binding and selective separation of such particles from fermentation or reactions broths can be done in a single step. Since the magnetizable particles can be produced from iron salts and low priced polymers, a single-use implementation of these systems is highly conceivable. In this article, the principles of magnetizable particles, their synthesis and functionalization are explained. Furthermore, applications in the area of reaction engineering, microfluidics and downstream processing are discussed focusing on established single-use technologies and development potential.
Commercial materials with polyvinylpolypyrrolidone and polymeric amberlites (XAD7HP, XAD16) are commonly used for the adsorptive downstream processing of polyphenols from renewable resources. In this study, beta-zeolite-based adsorbent systems were examined, and their properties were compared to organic resins. Batch adsorption experiments were conducted with synthetic solutions of major polyphenols. Adsorption isotherms and desorption characteristics of individual adsorbent were determined based on these results. Maximum adsorption capacities were calculated using the Langmuir model. For example, the zeolites had capacities up to 203.2 mg/g for ferulic acid. To extend these results to a complex system, additional experiments were performed on rapeseed meal and wheat seed extracts as representative renewable resources. HPLC analysis showed that with 7.5% w/v, which is regarded as the optimum amount of zeolites, zeolites A and B could bind 100% of the major polyphenols as well as release polyphenols at high yields. Additionally, regeneration experiments were performed with isopropyl alcohol at 99°C to evaluate how zeolites regenerate under mild conditions. The results showed only a negligible loss of adsorption capacity and no loss of desorption capacity. In summary, it was concluded that beta-zeolites were promising adsorbents for developing new processes to isolate polyphenols from renewable resources.
The interplay of albumin (BSA) and lysozyme (LYZ) adsorbed simultaneously on titanium was analyzed by gel electrophoresis and BCA assay. It was found that BSA and lysozyme adsorb cooperatively. Additionally, the isoelectric point of the respective protein influences the adsorption. Also, the enzymatic activity of lysozyme and amylase (AMY) in mixtures with BSA was considered with respect to a possible influence of protein-protein interaction on enzyme activity. Indeed, an increase of lysozyme activity in the presence of BSA could be observed. In contrast, BSA does not influence the activity of amylase.
Durch die Kombination von Oligonukleotid-Liganden (Aptameren) hoher Bindungsaffinitäten
mit hochselektiv abtrennbaren magnetisierbaren Mikropartikeln
wird eine einstufige Separation von Zielmolekülen aus mikrobiologischen
Produktionsansätzen möglich. Die Aptamere werden hierfür reversibel
auf den Partikeloberflächen gebunden und für die spezifische Isolierung von
Bioprodukten eingesetzt. Die Abtrennung der beladenen Partikel erfolgt
durch einen neuen Rotor-Stator-Separator mit Hochgradient-Magnetfeld.
Die vorliegende Erfindung betrifft eine Vorrichtung und ein Verfahren zur Bestimmung des Kontaktwinkels eines flüssigen oder mit Flüssigkeit gefüllten Körpers. Dieser besteht aus einem Träger (1) und einer damit verbundenen, in einem Winkelbereich von mehr als 0 ° bis maximal 90 ° neigbaren Ebene (8) mit einer darin ausgebildeten Abrollbahn (9) für den flüssigen oder mit Flüssigkeit gefüllten Körper. An der Ebene (8) sind mehrere Sensoren (11,12) zur Erfassung der Rolldauer des Körpers entlang der Rollstrecke angeordnet. Erfindungsgemäß ist vorgesehen, dass die Einstellung des Neigungswinkels der Ebene (8) über ein Winkelmessgerät (10) erfolgt, wodurch ein Abrollwinkel erfassbar ist, bei dem der Körper in Bewegung gerät. Aus der Rolldauer, der Rollstrecke und dem Abrollwinkel wird der Kontaktwinkel des Körpers ermittelt.
Due to the interfering effects of acetic acid in many fermentation processes, a gas-diffusion technique was developed for the online determination of acetic acid. The measurements were accomplished with a flow diffusion analysis (FDA) unit from the TRACE Analytics GmbH, Braunschweig, Germany. The diffusion analysis is based on the UV-absorbance of acetic acid at 205 nm. The measurement was achieved by the separation of an acceptor and a carrier stream (acidified fermentation broth) using a gas permeable polytetrafluoroethylene (PTFE) membrane, whereby broth constituents that would otherwise disturb the UV-measurement of acetic acid, are held back efficiently. Merely, the fermentation by-products, e.g. formic acid, is capable of diffusing through the membrane. While formic acid can disturb the measurement, carbon dioxide does not absorb at 205 nm. The method operates with time-dependent sample enrichment. During the analysis, a small volume of the acceptor stream is stopped for a defined time interval in the acceptor chamber. During this period, the gaseous acetic acid diffuses through the membrane and is enriched in the acceptor chamber. Subsequently after the enrichment, the acceptor stream flows through a UV-detector. The intensity of the signal is proportional to the acetic acid concentration. Online measurements in bioreactors via a sterile filtration probe have been accomplished. A linear calibration in the range of 0.5–5.0 g/L acetic acid with a relative standard deviation of <5 % was obtained. A sampling rate of 8 samples per hour was possible. The system was applied for the determination of acetic acid in E. coli fermentation broth. The instrument is easy to clean, very user-friendly and does not require any toxic or expensive reagents.
The development of a cost-effective hydrolysis for crude cellulose is an essential part of biorefinery developments. To establish such high solid hydrolysis, a new solid state reactor with static mixing is used. However, concentrations >10% (w/w) cause a rate and yield reduction of enzymatic hydrolysis. By optimizing the synergetic activity of cellulolytic enzymes at solid concentrations of 9%, 17% and 23% (w/w) of crude Organosolv cellulose, glucose concentrations of 57, 113 and 152 g L⁻¹ are reached. However, the glucose yield decreases from 0.81 to 0.72gg⁻¹ at 17% (w/w). Optimal conditions for hydrolysis scale-up under minimal enzyme addition are identified. As result, at 23% (w/w) crude cellulose the glucose yield increases from 0.29 to 0.49gg⁻¹. As proof of its applicability, biobutanol, succinic and itaconic acid are produced with the crude hydrolysate. The potential of the substrate is proven e.g. by a high butanol yield of 0.33gg⁻¹.
A major part of edible oil is subjected to bleaching procedures, primarily with minerals applied as adsorbers. Their recycling is currently done either by regaining the oil via organic solvent extraction or by using the spent bleaching earth (SBE) as additive for animal feed, etc. As a new method, the reutilization of the by-product SBE for the microbiologic formation of acetone, butanol, and ethanol (ABE) is presented as proof-of-concept. The SBE was taken from a palm oil cleaning process. The recycling concept is based on the application of lipolytic clostridia strains. Due to considerably long fermentation times, co-fermentation with Candida rugosa and enzymatic hydrolyses of the bound oil with a subsequent clostridia fermentation are shown as alternative routes. Anaerobic fermentations under comparison of different clostridia strains were performed with glycerol media, enzymatically hydrolyzed palm oil and SBE. Solutes, side product compositions and productivities were quantified via HPLC. A successful production of ABE solutes from SBE has been done with a yield of 0.15 g butanol per gram of bound glycerol. Thus, the biotechnological recycling of the waste stream is possible in principle. Inhibition of the substrate suspension has been observed. A chromatographic ion-exchange of substrates increased the biomass concentration.

Der Einsatz von Weizen als nachwachsende Rohstoffquelle ist allerdings nur dann wirt-schaftlich durchfu¨hrbar, wenn eine Prozessintegration in die bestehenden industriellen Verfahren gewa¨hrleistet oder einedirekte Konkurrenz zur Mehl- und Sta¨rkeindustrie vermieden werden kann. In diesem Artikel wird ein Verfahren aufge-zeigt, welches hohe Ausbeuten ermo¨glicht und eine Konkurrenz zu bestehenden Verwertungspfaden vermeidet.
For several thousand years, biotechnology and its associated technical processes have had a great impact on the development of mankind. Based on empirical methods, in particular for the production of foodstuffs and daily commodities, these disciplines have become one of the most innovative future issues. Due to the increasing detailed understanding of cellular processes, production strains can now be optimized. In combination with modern bioprocesses, a variety of bulk and fine chemicals as well as pharmaceuticals can be produced efficiently. In this article, some of the current trends in biotechnology are discussed.
Lignocellulosic biorefinery: Process integration of hydrolysis and fermentation (SSF process)

The aim of the present work is the process integration and the optimization of the enzymatic hydrolysis of wood and the following fermentation of the products to ethanol. The substrate is a fiber fraction obtained by organosolv pre-treatment of beech wood. For the ethanol production, a co-fermentation by two different yeasts (Saccharomyces cerevisiae and Pachysolen tannophilus) was carried out to convert glucose as well as xylose. Two approaches has been followed: 1. A two step process, in which the hydrolysis of the fiber fraction and the fermentation to product are separated from each other. 2. A process, in which the hydrolysis and the fermentation are carried out in one single process step as simultaneous saccharification and fermentation (SSF). Following the first approach, a yield of about 0.15 g ethanol per gram substrate can be reached. Based on the SSF, one process step can be saved, and additionally, the gained yield can be raised up to 0.3 g ethanol per gram substrate.
Ein viel versprechender erneuerbarer Rohstoff für die Produktion von Chemikalien und Treibstoffen ist Lignocellulose aus pflanzlicher Biomasse. Die darin enthaltenen Zucker können mittels enzymatischer Hydrolyse freigesetzt und fermentativ zu Ethanol umgesetzt werden. Ein interessanter Ansatz ist dabei die simultane Verzuckerung und Fermentation. Hefen und Enzyme haben mit 30 °C bzw. 50 °C zwar unterschiedliche Temperaturoptima, es konnte aber gezeigt werden, dass auch bei den niedrigeren Temperaturen eine Umsetzung der Cellulose zu Glucose erfolgt, wenn auch langsamer als bei optimalen Bedingungen. Außerdem konnte in Vorversuchen gezeigt werden, dass Ethanol in den zu erwartenden Konzentrationen keinen Einfluss auf die enzymatische Umsetzung hat.

Die Nutzung von Biomasse aus pflanzlichen Abfällen für die stoffliche Verwertung rückt immer stärker in den Vordergrund. Dabei ist vor allem die ganzheitliche Verwertung der Stoffströme von Bedeutung, da diese einen integrativen Ansatz ermöglichen. Im Rahmen dieser Arbeit wird die Produktion von Einzellerproteinen (Single-Cell Proteins, SCPs) mithilfe von unterschiedlichen Rohsubstraten dargelegt. Somit können Reststoffströme, die in keiner Konkurrenz zur Produktion von Lebensmitteln stehen, für die Herstellung von Futter- und auch Nahrungsmitteln Verwendung finden. Die zunächst thermisch vorbehandelten Ausgangsmaterialien stammen aus forstwirtschaftlichen und grünen Abfällen und ermöglichen durch eine anschließende enzymatische Hydrolyse die Freisetzung von Monosacchariden. Aus diesen erfolgt die SCP-Produktion fermentativ mithilfe der drei Modellorganismen Bakterium, Hefe und Pilz. Hierfür wird sowohl das flüssige Hydrolysat als auch der feste Reststoff auf der Basis einer Feststofffermentation genutzt. Auf diese Weise ist eine vollständige Verwertung der Ausgangsmaterialien möglich. Mit den gewonnen Daten erfolgt abschließend eine Bewertung der SCPs aus nachwachsenden Rohstoffen als alternative Proteinquelle.
Lignine bestehen aus einem hochgradig vernetzten Polymer phenolischer Grundeinheiten. Diese Verbindungen sind eine Quelle vielversprechender chemischer Grundbausteine. Auch die enzymatische Modifikation der Materialeigenschaften des Lignins ist für dessen Anwendung von Interesse. Aufgrund der verschiedenen Bindungstypen im Lignin ist eine Auftrennung mit nur einem Enzym unwahrscheinlich. Vielmehr sind verschiedene mediatorgestützte Reaktionen notwendig. Pilze, wie z.B. T. versicolor, nutzen Enzymkombinationen zum Aufschluss des Lignins. Hierbei kommen Laccase, Ligninperoxidase und Manganperoxidase zum Einsatz. Die optimale Kombination der Enzyme und ihrer Mediatoren bzw. Stabilisatoren ist Ziel der Untersuchungen. Aufgrund der großen Parameteranzahl wurde ein genetischer Algorithmus eingesetzt. Als Versuchsparameter wurden gewählt: die Verhältnisse der Enzyme, Ligninmasse, Konzentrationen an Eisen-, Mangan-, Oxalat-Ionen, ABTS, Violursäure und H₂O₂. Somit werden elf Parameter simultan optimiert. Als Algorithmus wurde ein Programm mit variabler Genkodierung entwickelt. Die Umsetzung des Lignins wird dabei über den verfolgt. Zurzeit ist ein enzymatischer Umsatz von 12% möglich. Als Referenz wurde eine chemische Lignindegradierung mit einem Umsatzvon 37% etabliert. Die sechs Generationen des Algorithmus zeigen eine Kongruenz der Enzymkonzentrationen von LiP, MnP und VeP, während Laccase keinen Einfluss hat. Des Weiteren beeinflussen die Konzentrationen von Mangan und Oxalat die Umsetzung, während die Variation von ABTS- und H₂O₂ nur eine geringe Auswirkung hat.
The concept of energy conversion into platform chemicals using bioelectrochemical systems (BES) has gained increasing attention in recent years, as the technology simultaneously provides an opportunity for sustainable chemical production and tackles the challenge of Power-to-X technologies. There are many approaches to realize the industrial scale of BES. One concept is to equip standard bioreactors with static electrodes. However, large installations resulted in a negative influence on various reactor parameters. In this study, we present a new single-chamber BES based on a stirred tank reactor in which the stirrer was replaced by a carbon fiber brush, performing the functions of the working electrode and the stirrer. The reactor is characterized in abiotic studies and electro-fermentations with Clostridium acetobutylicum. Compared to standard reactors an increase in butanol production of 20.14±3.66 % shows that the new BES can be efficiently used for bioelectrochemical processes.
Aufgrund von EU-Regularien und Umweltinitiativen wächst der Markt für nachhaltige und abbaubare Klebstoffe stetig. Organosolv (OS)-Lignin ist ein kommerziell wenig ertragreicher Nebenstrom der Lignocellulose-Bioraffinerie. Durch das "Nachahmen" der Adhäsionseigenschaften mit strukturverwandten Muschel-Aminosäuren soll OS-Lignin in einen starkes, vollständig biobasiertes Adhäsiv umgewandelt werden. Funktionsweisend für die Adhäsion des Muschelklebstoffes ist die Catecholgruppe der Aminosäure L-DOPA. Die laccase-katalysierte Polymerisationsreaktion von Lignin und L-DOPA ist schwierig zu kontrollieren, da L-DOPA eine Ringschlussreaktion eingeht. Stattdessen wurde eine zweistufige Reaktion mit einem Diamin als Ankermolekül etabliert. Die Catecholgruppe, die im zweiten Schritt enzymatisch an das Lignin-Amin gebunden wird, kann durch Komplexbildung mit Fe(III)-Ionen sowohl zur Adhäsion als auch zur Kohäsion des Klebstoffes beitragen. Der Lignin-Catechol-Klebstoff ist frei von petrochemischen Chemikalien und biologisch abbaubar. In ersten Stirnzugversuchen konnte eine Haftkraft von 0,3 MPa erreicht werden.
In der Molkeverarbeitung dominieren Membranfiltrationsverfahren die Prozessführung. Hierbei werden üblicherweise Aufkonzentrierungen der Proteine und deren Trennung von dem Milchzucker Lactose durchgeführt. Der Prozess der adsorptiven Aufreinigung soll als kostengünstige Alternative zu den bisher gebräuchlichen Verfahren dienen. Weiterhin eröffnet sich durch das Verfahren die Möglichkeit, einzelne Proteinfraktionen während der Verarbeitung anzureichern. Als Proteinquellen wurden für die Untersuchungen Modellproteine, Lösungen aus Molkenproteinisolat, Dünnmolke und Molkekonzentrat verwendet. Die Eignung zur Proteinbindung wurden an Tonmaterialien, Silicaten und y-Aluminiumoxiden in Pulverform, in Form von Granulaten sowie Extrudaten als auch sphärischen Partikeln überprüft. Adsorbentien aus Bentonit/Silica und c-Aluminiumoxid können sowohl a-Lactalbumin (aLA) als auch b-Lactoglobulin (bLG) binden, wohingegen Materialien aus Siliciumoxid lediglich ein starkes Adsorptionsverhalten gegenüber bLG zeigen. Mischmaterialien aus Siliciumoxid und a-Aluminiumoxid zeigen dasselbe Verhalten wie Materialien aus Siliciumoxid, weisen jedoch eine geringere Kapazität auf. Die Materialen wurden hinsichtlich ihres Einsatzes in chromatographischen Verfahren und Batch-Prozessen untersucht und ein Prozessentwurf für einen zweistufigen Batch-Prozess im Rührkessel erarbeitet.
The emerging environmental issues due to the use of fossil resources are encouraging the exploration of new renewable resources. Biomasses are attracting more interest due to the low environmental impacts, low costs, and high availability on earth. In this scenario, green biorefineries are a promising platform in which green biomasses are used as feedstock. Grasses are mainly composed of cellulose and hemicellulose, and lignin is available in a small amount. In this work, a perennial ryegrass was used as feedstock to develop a green bio-refinery platform. Firstly, the grass was mechanically pretreated, thus obtaining a press juice and a press cake fraction. The press juice has high nutritional values and can be employed as part of fermentation media. The press cake can be employed as a substrate either in enzymatic hydrolysis or in solid-state fermentation. The overall aim of this work was to demonstrate different applications of both the liquid and the solid fractions. For this purpose, the filamentous fungus A. niger and the yeast Y. lipolythica were selected for their ability to produce citric acid. Finally, the possibility was assessed to use the press juice as part of fermentation media to cultivate S. cerevisiae and lactic acid bacteria for ethanol and lactic acid fermentation.
Die ökonomische Abhängigkeit von fossilen Brennstoffen und der klimatische Wandel durch die Nutzung dieser haben zu einer intensiven Suche nach erneuerbaren Rohstoffen für die Produktion von Chemikalien und Treibstoffen geführt. Ein viel versprechender Rohstoff in diesem Zusammenhang sind Zucker, die mittels enzymatischer Hydrolyse aus Lignocellulose gewonnen werden können. Die Fermentation erfolgt mit Cellulose- bzw. Hemicellulose-Fraktionen, welche durch thermo-chemische Vorbehandlung von Holz gewonnen und anschließend enzymatisch hydrolysiert werden. Die in den Hydrolysaten enthaltenen Zuckermonomere dienen als Kohlenstoffquelle für die Produktion von Ethanol. Da sowohl Glucose als auch Xylose in den unterschiedlichen Fraktionen enthalten sind, wird zur Umsetzung dieser eine Co-Fermentation zweier Hefen durchgeführt. Im Rahmen der Optimierung dieser Fermentationen werden neben der Ergänzung der Hydrolysate durch notwendige Salze auch Verfahrenweisen wie Fed-Batch-Fermentationen untersucht. Ein weiterer interessanter Ansatz, welcher in diesem Rahmen geprüft wird, ist die enzymatische Hydrolyse der Lignocellulose-Fraktionen und die simultane Fermentation der dabei entstehenden Zucker in einem Schritt. Des Weiteren wurde die Eignung der Hydrolysate für die Biomasseproduktion anderer Mikroorganismen wie Escherichia coli getestet.
b-Lactame gehören zu den wirkungsvollsten Antibiotika, jedoch lassen sich viele nur schwierig fermentativ erzeugen. Ein Problem bei der fermentativen Produktion ist die Hydrolyse des Lactamrings im wässrigen Milieu. Das Ziel des von der DBU geförderten Projekts ist die selektive In-situ-Adsorption der b-Lactamantibiotika unter anschließender magnetischer Separation. Durch die Isolation im Hochgradientenmagnetseparator (HGMS) ist eine Fest-fest-flüssig-Trennung und somit ein erheblicher Zeitgewinn im Downstreamprozess möglich. Zusätzlich kommt es zur Einsparung an Lösungsmittel und Energie, was neben Reduzierung der Antibiotikahydrolyse auch in ökologischer Hinsicht einen interessanten Aspekt darstellt. Als Trägermaterial für die Adsorbermatrix werden magnetisierbare Eisenoxidpartikel eingesetzt, die in einer Silikamatrix eingebettet sind. Diese Adsorber sollen auf Selektivität in Wasser und verschiedenen Medien getestet werden. Zusätzlich werden die Abbauprodukte des b-Lactams analysiert, um eine Aussage über die Stabilisierung des Antibiotikums durch die selektiven Adsorber treffen zu können. Diese Ergebnisse werden mit kommerziell erhältlichen Adsorbern verglichen. Die Aufreinigung der Antibiotika soll direkt aus der Fermentationsbrühe erfolgen. Um die Trennung der magnetischen, selektiven Adsorber von der Biomasse zu gewährleisten, soll der HGMS in die Fermentation integriert werden. Das filamentöse Wachstum des Mikroorganismus erfordert eine Neuauslegung der Filtermatrix.
Aktiver und passiver antimikrobieller Oberflächenschutz durch funktionalisierte Mikropartikel

Mikrobielle Verunreinigungen von Oberflächen in technischen und medizinischen Systemen sind allgegenwärtig. Sie basieren üblicherweise auf adsorptiven Oberflächenbindungen organischer Komponenten (Proteine und Fette) oder Membrankomponenten aerogener sowie wassergebundener Mikroorganismen. In laufenden Forschungsarbeiten wird eine aktive sowie passive Biomodifikation von Oberflächen zu deren Schutz vor Adsorption von Proteinen und Mikroorganismen verfolgt. Der antimikrobielle Schutz soll dabei sowohl durch die Mikrostrukturierung bzw. Rauheitsanpassung der Oberflächen durch deren Beschichtung mit Mikro-und Nanopartikeln erfolgen. Ferner werden antimikrobielle Enzyme und funktionelle Gruppen auf den Mikropartikeln gebunden, um den Oberflächenschutz zu verstärken. In ersten Versuchen wurden quartäre Ammoniumverbindungen auf eigens synthetisierten superparamagnetischen Eisenoxid-Nanopartikeln (Durchmesser 10 – 30 nm) immobilisiert und die wachstumshemmende Wirkung untersucht. Erste Ergebnisse zeigten, dass eine Konzentration von 10 mg mL⁻¹ der Ammoniumverbindung in einer Wachstumshemmung des verwendeten Gram-negativen Modell-Mikroorganismus E. coli GFPmut2 resultiert. Zurzeit werden synergistisch wirkende Kombinationen von Partikeln mit Proteasen, quartären Ammoniumverbindungen, hydrophoben Oberflächen und mikrostrukturierten Oberflächen als antimikrobieller Schutz untersucht.
In der Reihe der nachwachsenden Rohstoffe besitzt Holz als erneuerbare und umweltfreundliche Ressource ein großes Potenzial. Über 11 Mio. ha Holz, das laut der Fachagentur für nachwachsende Rohstoffe (FNR) auch für industrielle Zwecke genutzt werden kann, wuchsen im Jahr 2013 allein auf bundesdeutscher Fläche. 56,8 Mio. m³ jährlicher Holzeinschlag in den letzten zehn Jahren wurde zu knapp der Hälfte stofflich und der Rest energetisch verwertet. Im Rahmen dieser Arbeit konnte auf der Basis vom Holz der Buche, die nach Fichte und Kiefer die dritthäufigste Baumart in Deutschland ist und 15% der deutschen Waldfläche ausmacht, die Fraktionierung der polymeren Hauptbestandteile mit niedrigem energetischen Einsatz erreicht werden. Hierbei werden in einem nachgeschalteten Extraktionsprozess die beiden Komponenten Hemicellulose und Lignin in flüssiger Form von der finalen festen Cellulosefraktion abgetrennt. Die Extraktion der Hemicellulose erfolgt durch eine Liquid Hot Water (LHW)-Behandlung. Untersucht wird der katalytische Zusatz anorganischer Säuren wie H₃PO₄ und H₂SO₄. Im Hinblick auf die weitere Verwertung von Lignin zu aromatischen Synthesebausteinen kommt die Organosolv-Extraktion mit einem Ethanol/Wasser-Gemisch zum Einsatz. Von Vorteil ist die weitere Verwendung beider Stoffströme ohne Fällungsschritt und nachteiliger Verdünnung der Hemicellulose.
Die Ausbildung von Biofilmen in technischen Anlagen, wie z. B. Kühlkreisläufen, Wasseraufbereitungssystemen und Bioreaktoren, führen zu Materialschäden (Biofouling) und stark erhöhtem Energieaufwand. Im Rahmen der aktuellen Forschungsarbeiten erfolgen aktive sowie passive Bio-Modifikationen auf funktionalisierten magnetischen Mikropartikelober-flächen. Um die verschiedenen funktionalisierten magnetischen Mikropartikel zu analysieren und ihre antimikrobielle Wirkung zu testen, wird der Einsatz einer 3D-gedruckten, magnetischen Plattform für ein Fluoreszenz-basiertes Screening-System untersucht. Für den Oberflächenschutz wurden verschiedene, antimikrobiell funktionalisierte Partikelkombinationen mit dem Mikroorganismus Escherichia coli GFPmut2 in Bezug auf aktiven Oberflächenschutz verglichen. Um die antimikrobielle Oberflächeneffekte von synergistischen Kombinationen unterschiedlich funktionalisierter Partikel zu bestimmen, werden Oberflächen einem Magnetfeld ausgesetzt, das die Mikropartikel als definierte Schicht auf ihnen zurück hält. Diese modifizierten Oberflächen können sowohl durch Fluoreszenzspektroskopie als auch -mikroskopie analysiert werden.
Gräser sind in der Lage, einen großen Teil der für eine biobasierte Wirtschaft benötigten Biomasse zur Verfügung zu stellen. Wie bei anderen lignocellulosehaltigen nachwachsenden Rohstoffen erfordert die Verwertung der im Gras enthaltenen Polysaccharide einen mehrstufigen Prozess aus Vorbehandlung, Hydrolyse und Fermentation. In Gräsern ist die Hemicellulose mitP henolcarbonsäuren wie Ferula- und p-Coumarsäure verestert, die die enzymatische Hydrolyse der Cellulose und Hemicellulose ebenso effektiv behindern wie Lignin. Anders als bei holzigen Rohstoffen ermöglicht dieser Aufbau aber eine enzymatische Vorbehandlung, mit der die Phenolcarbonsäuren abgespalten werden können. Da die bei der Vorbehandlung eingesetzten Enzyme in ihrer natürlichen Funktion synergistisch mit cellulytischen Enzymen zusammenarbeiten, besitzen sie ähnliche Optima wie die für die Hydrolyse der Polysaccharide eingesetzten Cellulasen und Hemicellulasen. Diese Eigenschaft ermöglicht die Integration von Vorbehandlung und Hydrolyse in einem einzigen Verfahrensschritt. Durch die Einführung der enzymatischen Vorbehandlung konnte das in der Literatur bekannte SSF-Verfahren für die Herstellung von Ethanol aus Gräsern um die Vorbehandlungsstufe erweitert werden. Das so realisierte simultaneous pretreatment, saccharification and fermentation (SPSF)-Verfahren stellt eine vollständige Integration der drei für die Nutzung von Lignocellulose nötigen Verfahrensschritte in der grünen Bioraffinerie dar.
Biotechnologie und die mit ihr verbundenen technischen Prozesse prägen seit Jahrtausenden die Entwicklung der Menschheit. Ausgehend von empirischen Verfahren, insbesondere zur Herstellung von Lebensmitteln und täglichen Gebrauchsgütern, haben sich diese Disziplinen zu einem der innovativsten Zukunftsfelder entwickelt. Durch das immer detailliertere Verständnis zellulärer Vorgänge können mittlerweile Produktionsstämme gezielt optimiert werden. Im Zusammenspiel mit moderner Prozesstechnik können so eine Vielzahl von Bulk- und Feinchemikalien sowie Pharmazeutika effizient hergestellt werden. In diesem Artikel werden exemplarisch einige der aktuellen Trends vorgestellt.
High gradient magnetic separation (HGMS) has been established since the early 1970s. A more recent application of these systems is the use in bioprocesses. To integrate the HGMS in a fermentation process, it is necessary to optimize the separation matrix with regard to the magnetic separation characteristics and permeability of the non-magnetizable components of the fermentation broth. As part of the work presented here, a combined fluidic and magnetic force finite element model simulation was created using the software COMSOL Multiphysics and compared with separation experiments. Finally, as optimal lattice orientation of the separation matrix, a transversal rhombohedral arrangement was defined. The high suitability of the new filter matrix has been verified by separation experiments.

The invention relates to a system for the implementation of chemical, biological or physical reactions, consisting of - one or more magnetic micro-reactors, each comprising a shell made of hydrophobic magnetic nanoparticles encapsulating an aqueous core, - a plane platform comprising a surface to receive the micro-reactors, - a source that generates a magnetic field above or underneath the platform for manipulating the one or more hydrophobic magnetic micro-reactors, or for moving them along the surface of the platform from one position to another position, characterized in that the aqueous core of the one or more magnetic micro-reactors contains a reaction solution or buffer, and wherein the magnetic field generated by the source correlates to a defined position on the surface of the platform.
Due to their anion exchange characteristics, layered double hydroxides (LDHs) are suitable for the detoxification of aqueous, fatty acid containing fermentation substrates. The aim of this study is to examine the adsorption mechanism, using crude glycerol from plant oil esterification as a model system. Changes in the intercalation structure in relation to the amount of fatty acids adsorbed are monitored by X-ray diffraction and infra-red spectroscopy. Additionally, calcination of LDH is investigated in order to increase the binding capacity for fatty acids. Our data propose that, at ambient temperature, fatty acids can be bound to the hydrotalcite by adsorption or in addition by intercalation, depending on fatty acid concentration. The adsorption of fatty acids from crude glycerol shows a BET-like behavior. Above a fatty acid concentration of 3.5 g L−1, intercalation of fatty acids can be shown by the appearance of an increased interlayer spacing. This observation suggests a two phase adsorption process. Calcination of LDHs allows increasing the binding capacity for fatty acids by more than six times, mainly by reduction of structural CO32−.
