The CellDrum technology (The term 'CellDrum technology' includes a couple of slightly different technological setups for measuring lateral mechanical tension in various types of cell monolayers or 3D-tissue constructs) was designed to quantify the contraction rate and mechanical tension of self-exciting cardiac myocytes. Cells were grown either within flexible, circular collagen gels or as monolayer on top of respective 1-mum thin silicone membranes. Membrane and cells were bulged outwards by air pressure. This biaxial strain distribution is rather similar the beating, blood-filled heart. The setup allowed presetting the mechanical residual stress level externally by adjusting the centre deflection, thus, mimicking hypertension in vitro. Tension was measured as oscillating differential pressure change between chamber and environment. A 0.5-mm thick collagen-cardiac myocyte tissue construct induced after 2 days of culturing (initial cell density 2 x 10(4) cells/ml), a mechanical tension of 1.62 +/- 0.17 microN/mm(2). Mechanical load is an important growth regulator in the developing heart, and the orientation and alignment of cardiomyocytes is stress sensitive. Therefore, it was necessary to develop the CellDrum technology with its biaxial stress-strain distribution and defined mechanical boundary conditions. Cells were exposed to strain in two directions, radially and circumferentially, which is similar to biaxial loading in real heart tissues. Thus, from a biomechanical point of view, the system is preferable to previous setups based on uniaxial stretching.
Design and initial performance of PlanTIS: a high-resolution positron emission tomograph for plants

Positron emitters such as 11C, 13N and 18F and their labelled compounds are widely used in clinical diagnosis and animal studies, but can also be used to study metabolic and physiological functions in plants dynamically and in vivo. A very particular tracer molecule is 11CO2 since it can be applied to a leaf as a gas. We have developed a Plant Tomographic Imaging System (PlanTIS), a high-resolution PET scanner for plant studies. Detectors, front-end electronics and data acquisition architecture of the scanner are based on the ClearPET™ system. The detectors consist of LSO and LuYAP crystals in phoswich configuration which are coupled to position-sensitive photomultiplier tubes. Signals are continuously sampled by free running ADCs, and data are stored in a list mode format. The detectors are arranged in a horizontal plane to allow the plants to be measured in the natural upright position. Two groups of four detector modules stand face-to-face and rotate around the field-of-view. This special system geometry requires dedicated image reconstruction and normalization procedures. We present the initial performance of the detector system and first phantom and plant measurements.
For more than a decade up to now there is an ongoing interest in small gas turbines downsized to micro-scale. With their high energy density they offer a great potential as a substitute for today’s unwieldy accumulators, found in a variety of applications like laptops, small tools etc. But micro-scale gas turbines could not only be used for generating electricity, they could also produce thrust for powering small unmanned aerial vehicles (UAVs) or similar devices. Beneath all the great design challenges with the rotating parts of the turbomachinery at this small scale, another crucial item is in fact the combustion chamber needed for a safe and reliable operation. With the so called regular micromix burning principle for hydrogen successfully downscaled in an initial combustion chamber prototype of 10 kW energy output, this paper describes a new design attempt aimed at the integration possibilities in a μ-scale gas turbine. For manufacturing the combustion chamber completely out of stainless steel components, a recuperative wall cooling was introduced to keep the temperatures in an acceptable range. Also a new way of an integrated ignition was developed. The detailed description of the prototype’s design is followed by an in depth report about the test results. The experimental investigations comprise a set of mass flow variations, coupled with a variation of the equivalence ratio for each mass flow at different inlet temperatures and pressures. With the data obtained by an exhaust gas analysis, a full characterisation concerning combustion efficiency and stability of the prototype chamber is possible. Furthermore the data show a full compliance with the expected operating requirements of the designated μ-scale gas turbine.
The idea of component-based software engineering was proposed more that 40 years ago, yet only few robotics software frameworks follow these ideas. The main problem with robotics software usually is that it runs on a particular platform and transferring source code to another platform is crucial. In this paper, we present our software framework Fawkes which follows the component-based software design paradigm by featuring a clear component concept with well-defined communication interfaces. We deployed Fawkes on several different robot platforms ranging from service robots to biped soccer robots. Following the component concept with clearly defined communication interfaces shows great benefit when porting robot software from one robot to the other. Fawkes comes with a number of useful plugins for tasks like timing, logging, data visualization, software configuration, and even high-level decision making. These make it particularly easy to create and to debug productive code, shortening the typical development cycle for robot software.
A handheld sensor system for the online measurement of hydrogen peroxide (H2O2) in aseptic sterilisation processes has been developed. It is based on a calorimetric-type gas sensor that consists of a differential set-up of two temperature sensors, of which one is catalytically activated and the second one is passivated and used as reference. The sensor principle relies in detecting a rise in temperature on the active sensor due to the exothermic reaction of H2O2 on the catalytic surface. To characterise the sensor system towards H2O2 sensitivity and other influencing factors, measurements have been carried out both at an experimental set-up and a manufacturer's sterilisation machine. Physical sensor characterisation was done by means of the optical microscopy.
Chalcogenide glass materials as membranes for potentiometric sensors for chemical analysis in solutions have been studied since more than 20 years. In this work, an electrolyte–insulator–semiconductor structure was combined with chalcogenide glass membranes prepared by means of the pulsed laser deposition technique. Depending on the membrane composition a selectivity to different ions (Cd2+ and Pb2+) is achieved. The different sensor membranes have been physically characterised using microscopy, ellipsometry, profilometry, atomic force microscopy (AFM), scanning electron microscopy (SEM) and Rutherford backscattering spectrometry (RBS). The electrochemical behaviour has been investigated via capacitance/voltage (C/V) and constant capacitance (ConCap) measurements and results in a Cd2+ sensitivity of 23.1 ± 0.6 mV per decade in a linear range from 7 × 10−6 to 10−2 mol/l and 24.4 ± 0.5 mV per decade in a linear range from 5 × 10−6 to 10−2 mol/l for Pb2+, respectively.

In der biopharmazeutischen Industrie werden rekombinante Proteine und monoklonale Antikörper in Zellkulturfermentationen produziert, da nur humane oder tierische Zelllinien über die Fähigkeit der Glykosylierung verfügen. Um hohe Produktausbeuten in ausgezeichneter Qualität zu erzielen, ist eine funktionstüchtige Prozesskontrolle unerlässlich. Hierzu wurde in Kooperation mit der Firma Hitec Zang GmbH die HiSense Präzisionsabgasanalytik entwickelt, die auf Basis der vollautomatischen Ermittlung des Respirationsquotienten (RQ; Verhältnis vonKohlendioxidbildungsrate (CER) zu Sauerstoffaufnahmerate (OTR)) einen Fermentationsprozess nicht-invasiv überwacht. Der RQ kann in Hybridoma- und CHO-Zellen (s. Abb.) in sowohl serumhaltigen als auch serumfreien Medien erfolgreich ermittelt werden. Hier spiegeln die CER und die OTR das Wachstumsverhalten der kultivierten CHO-Zellen wider. Der RQ nimmt dabei Werte zwischen 0,9 und 1,2 an. Dies lässt auf verschiedene Stoffwechselaktivitäten schließen. Da die momentane industrielle Prozesskontrolle auf gemessenen Sauerstoffaufnahmeraten oder entsprechende Offline-Analytiken der Metaboliten basieren, soll durch die vollautomatische RQ-Ermittlung ein neues Verfahren zur Fermentationsüberwachung etabliert werden. Bisher war diese, in bakteriellen Kultivierungen standardisierte Methode, aufgrund der schwierigen CER-Berechnung bei Zellkulturen keine adäquate Alternative.
Das verbraucherschützende Widerrufsrecht ist in die Jahre gekommen. Als Ergebnis der zunehmenden Rechtszersplitterung im europäischen Richtlinienrecht hat sich eine unüberschaubare Bandbreite an Ausgestaltungsformen in den mitgliedstaatlichen Rechtssystemen herausgebildet. Effektivitätseinbußen, Wettbewerbsverzerrungen und ein hohes Maß an Rechtsunsicherheit sind die Folgen. Zurückführen lässt sich die kostenintensive Fragmentierung des Verbrauchervertragsrechts auf die bisherige Regelungspolitik der Mindestharmonisierung, die es den Mitgliedstaaten erlaubt, von dem durch die Richtlinien gesetzten Mindeststandard durch „überschießende Umsetzung” abzuweichen. Der Unionsgesetzgeber versucht dem vermehrt durch einen Prozess der Vollharmonisierung zu begegnen: Neben Reformen der Richtlinie über Verbraucherkredite (VerbrKrRL) sowie der Timesharing-Richtlinie (TimesharingRL), liegt seit dem 8. Oktober 2008 nunmehr auch der Entwurf einer Rahmenrichtlinie über die Rechte der Verbraucher (VRRL-E) vor. Danach sollen die bisherigen Richtlinien über Haustürgeschäfte (HaustürgeschäfteRL), Fernabsatzverträge (FARL), missbräuchliche Klauseln sowie Verbrauchsgüterkäufe vereinigt werden. Für das verbraucherschützende Widerrufsrecht ist damit zum einen die Herausbildung einheitlicher Kernelemente verbunden, zum anderen tritt es in Konkurrenz zu umfassenden Informationspflichten, die ebenfalls den Schutz des Verbrauchers vor dem unüberlegten Abschluss riskanter bzw. nachteiliger Verträge zum Ziel haben.
Angesichts dieser Entwicklung stellt sich im Folgenden die Frage nach der Zukunft des unionsrechtlichen Widerrufsrechts. Von besonderer Bedeutung ist dabei die Klärung des Verhältnisses zu den verbraucherschützenden Informationspflichten. Dazu bedarf es zunächst der Herausarbeitung einheitlicher Kernelemente des Widerrufsrechts im vollharmonisierten Richtlinienrecht (II.) sowie einer Systematisierung der sekundärrechtlichen Informationspflichten im Anschluss eine kritische Gegenüberstellung der Vor- und Nachteile beider Schutzinstrumente erfolgen kann (IV.).
Im Herbst 2009 wurde das “Deutsche Gütesiegel Nachhaltiges Bauen” für die Gebäudetypen Industriebauten und Handelsbauten offiziell vorgestellt. Die Ökobilanz als wichtiger Baustein der Nachhaltigkeitsbewertung wird somit auch im Industrie- und Gewerbebau zukünftig stark an Bedeutung gewinnen.
Der vorliegende Beitrag beinhaltet einen Vergleich der Umweltleistung verschiedener Bauweisen des Industrie- und Gewerbebaus, die durch die Ökobilanzierung quantifizierbar wird, anhand von momentan verfügbaren Daten und Methoden. Es werden ausgewählte Ergebnisse sowie vorhandene Unsicherheiten und daraus resultierender Forschungsbedarf aufgezeigt.
Differential modulation of valence and arousal in high-alexithymic and low-alexithymic individuals

High-alexithymic individuals are characterized by an impaired ability to identify and communicate emotions whereas low-alexithymic individuals have a wide-ranging ability to deal with emotions. This study examined the hypothesis that valence and arousal modifications of emotional stimuli differentially modulate cortical regions in high-alexithymic and low-alexithymic individuals. To this end, 28 high-alexithymic and 25 low-alexithymic individuals were investigated with event-related fMRI using visual emotional stimuli. We found differential neural activations in the dorsal anterior cingulate, the insula and the amygdala. We suggest that these differences may account for the impaired ability of high-alexithymic individuals to appropriately handle emotional stimuli.
Bilaterale stromale Hornhauttrübungen sind für den Augenarzt eine differenzialdiagnostische Herausforderung. Im folgenden Beitrag werden 2 Patieninnen (30 und 36 Jahre) mit unterschiedlich stark ausgeprägter stromaler diffuser Hornhauttrübung vorgestellt. Patientin 1 war kleinwüchsig (114 cm) und Patientin 2 normal groß (172 cm). Beide Patientinnen wiesen veränderte Gelenkstrukturen an Hand und Fußgelenken sowie diffuse stromale Hornhauttrübungen auf. Des Weiteren lagen eine Mitral- und Aorteninsuffizienz (Patientin 1) bzw. eine Aorteninsuffizienz (Patientin 2) vor. Die stromalen diffusen Hornhauttrübungen ließen im Zusammenhang mit den Gelenkveränderungen ein Scheie-Syndrom vermuten. Therapeutisch ist bei Patienten mit Visusminderung eine (lamelläre) Keratoplastik sinnvoll.
In Valais, Switzerland, Scots pines (Pinus sylvestris L.) are declining, mainly following drought. To assess the impact of drought on tree growth and survival, an irrigation experiment was initiated in 2003 in a mature pine forest, approximately doubling the annual precipitation. Tree crown transparency (lack of foliage) and leaf area index (LAI) were annually assessed. Seven irrigated and six control trees were felled in 2006, and needles, stem discs and branches were taken for growth analysis. Irrigation in 2004 and 2005, both with below-average precipitation, increased needle size, area and mass, stem growth and, with a 1-year delay, shoot length. This led to a relative decrease in tree crown transparency (−14%) and to an increase in stand LAI (+20%). Irrigation increased needle length by 70%, shoot length by 100% and ring width by 120%, regardless of crown transparency. Crown transparency correlated positively with mean needle size, shoot length and ring width and negatively with specific leaf area. Trees with high crown transparency (low growth, short needles) experienced similar increases in needle mass and growth with irrigation than trees with low transparency (high growth, long needles), indicating that seemingly declining trees were able to ‘recover’ when water supply became sufficient. A simple drought index before and during the irrigation explained most of the variation found in the parameters for both irrigated and control trees.
Unter Energieeffizienz wird verstanden, dass ein definiertes Ziel mit möglichst wenig Energie erreicht wird. Im Beitrag wird zunächst eine Formel zur Berechnung der Gesamtenergieeffizienz eines Gebäudes angegeben. Darauf aufbauend wird ein 3-Säulen-Modell zur Energieeinsparung vorgeschlagen. Es beinhaltet vereinfachte Verfahren zur Berechnung der Betriebseigenschaften von Heizanlagen, raumlufttechnischen Anlagen und Kühl- und Kaltwasseranlagen. Am Beispiel einer RLT-Anlage wird die Vorgehensweise der Bewertung näher erläutert. Zusammenfassend wird festgestellt, dass das vereinfachte Verfahren eine schnelle und sichere energetische Bewertung der genannten Anlagen erlaubt,, zu Präsenzwissen führt, die Haupteinflussgrößen aufzeigt und eine Anlagenbewertung ohne Gebäudebewertung gewährleistet. Das vereinfachte Verfahren wird im Frühjahr 2011 verfügbar sein. Es soll Bestandteil der DIN V 18599 werden, und die EnEV 2012 soll darauf Bezug nehmen.
Energiebilanzen und -potenziale von Abwasseranlagen : Vorstellung neuer Untersuchungsvorhaben in NRW

Moderne Bauwerke müssen heute eine hohe energetische Leistungsfähigkeit aufweisen und gleichzeitig alle einwirkenden Lasten sicher abtragen. Dies stellt insbesondere in Erdbebengebieten hohe Anforderungen an die verwendeten Baustoffe. Am baupraktischen Beispiel einer Doppelhaushälfte wird demonstriert, dass die Symbiose aus energieeffizientem und gleichzeitig erdbebensicherem Bauen in der höchsten deutschen Erdbebenzone mit monolithischem Ziegelmauerwerk gut realisierbar ist. Als Ziegelmauerwerk werden für die Außenwände wärmetechnisch optimierte Hochlochziegel verwendet, die sowohl die Anforderungen der Energieeinsparverordnung 2009 als auch die Anforderungen an Mauerwerkbaustoffe nach den aktuellen Erdbebennormen erfüllen. Der Erdbebennachweis der Doppelhaushälfte erfolgt mit einem nichtlinearen Nachweisverfahren, das für eine einfache praktische Anwendung programmtechnisch umgesetzt wurde. Für den Nachweis wurden aus zyklischen Schubwandversuchen ermittelte Last-Verformungskurven verwendet. Das gesamte in Deutschland noch nicht normativ geregelte Nachweiskonzept wurde im Rahmen einer Zustimmung im Einzelfall geprüft und genehmigt.
In diesem Beitrag werden Ergebnisse der Entwicklung eines modularen festkörperbasierten Sensorsystems für die Überwachung von Zellkulturfermentationen präsentiert. Zur Messung der Elektrolytleitfähigkeit wurde das Layout von Interdigitalelektroden angepasst, um in vergleichsweise gut leitenden Elektrolyten zu messen. Durch Quervernetzung von Glucose-Oxidase mit Glutaraldehyd und Immobilisierung auf einer Platinelektrode wurde ein amperometrischer Glucosesensor mit einem linearen Messbereich von bis zu 2 mM und einer Sensitivität von 168 nA/mM realisiert.
Lignine bestehen aus einem hochgradig vernetzten Polymer phenolischer Grundeinheiten. Diese Verbindungen sind eine Quelle vielversprechender chemischer Grundbausteine. Auch die enzymatische Modifikation der Materialeigenschaften des Lignins ist für dessen Anwendung von Interesse. Aufgrund der verschiedenen Bindungstypen im Lignin ist eine Auftrennung mit nur einem Enzym unwahrscheinlich. Vielmehr sind verschiedene mediatorgestützte Reaktionen notwendig. Pilze, wie z.B. T. versicolor, nutzen Enzymkombinationen zum Aufschluss des Lignins. Hierbei kommen Laccase, Ligninperoxidase und Manganperoxidase zum Einsatz. Die optimale Kombination der Enzyme und ihrer Mediatoren bzw. Stabilisatoren ist Ziel der Untersuchungen. Aufgrund der großen Parameteranzahl wurde ein genetischer Algorithmus eingesetzt. Als Versuchsparameter wurden gewählt: die Verhältnisse der Enzyme, Ligninmasse, Konzentrationen an Eisen-, Mangan-, Oxalat-Ionen, ABTS, Violursäure und H₂O₂. Somit werden elf Parameter simultan optimiert. Als Algorithmus wurde ein Programm mit variabler Genkodierung entwickelt. Die Umsetzung des Lignins wird dabei über den verfolgt. Zurzeit ist ein enzymatischer Umsatz von 12% möglich. Als Referenz wurde eine chemische Lignindegradierung mit einem Umsatzvon 37% etabliert. Die sechs Generationen des Algorithmus zeigen eine Kongruenz der Enzymkonzentrationen von LiP, MnP und VeP, während Laccase keinen Einfluss hat. Des Weiteren beeinflussen die Konzentrationen von Mangan und Oxalat die Umsetzung, während die Variation von ABTS- und H₂O₂ nur eine geringe Auswirkung hat.

Biomechanics studies biological soft tissue materials (growth, remodeling) in vivo. For this objective, the detailed information of material properties must be well defined to construct reliable constitutive models. In the paper, the bulge test is carried out with elastomers in order to develop a test method. Then, application of the test for soft tissue materials is straightforward due to the similarities between elastomers with soft tissue materials as proved in Holzapfel 2005, Ogden 2009. It means, after the preliminary experiments and parameter identification with rubber materials has been setup, experiments on soft tissue materials can be similarly carried out. Elastomers have a complex behavior which strongly depends on the largest previous load cycle. For simplicity we consider only the first loading.
Various models have been proposed for the prediction of the necessary support pressure at the face of a shallow tunnel. To assess their quality, the collapse of a tunnel face was modelled with small-scale model tests at single gravity. The development of the failure mechanism and the support force at the face in dry sand were investigated. The observed displacement patterns show a negligible influence of overburden on the extent and evolution of the failure zone. The latter is significantly influenced, though, by the initial density of the sand: in dense sand a chimney-wedge-type collapse mechanism developed, which propagated towards the soil surface. Initially, loose sand did not show any discrete collapse mechanism. The necessary support force was neither influenced by the overburden nor the initial density. A comparison with quantitative predictions by several theoretical models showed that the measured necessary support pressure is overestimated by most of the models. Those by Vermeer/Ruse and Léca/Dormieux showed the best agreement to the measurements.
With the final objective of optimizing the "Micromix" hydrogen combustion principle, a round jet in a laminar cross-flow prior to its combustion is investigated experimentally using Stereoscopic Particle Image Velocimetry. Measurements are performed at a jet to cross-stream momentum ratio of 1 and a Reynolds number, based on the jet diameter and jet velocity, of 1600. The suitability to combine side, top and end views is analyzed statistically. The statistical theory of testing hypotheses, pertaining to the joint distribution of the averaged velocity along intersecting observation planes, is employed. Overall, the averaged velocity fields of the varying observation planes feature homogeneity at a 0.05 significance level. Minor discrepancies are related to the given experimental conditions. By use of image maps, averaged and instantaneous velocity fields, an attempt is made to elucidate the flow physics and a kinematically consistent vortex model is proposed. In the time-averaged flow field, the principal vortical systems were identified and the associated mixing visualized. The jet trajectory and physical dimensions scale with the momentum ratio times the jet diameter. The jet/cross-flow mixture converging upon the span-wise centre-line, the lifting action of the Counter Rotating Vortex Pair and the reversed flow region contribute to the high entrainment and mixedness. It is shown that the jet width is larger on the downstream side as compared to the upstream side of the centre-streamline. The deepest penetration of the particles on the outer boundary occurs in the centre-plane. Meanwhile, with increasing off-centre position, the boundaries all lay further from the centre-line position than does the boundary in the centre-plane, corresponding to a kidney-like shape of the flow cross-section. The generation of the Counter Rotating Vortex Pair and the instability mechanism is documented by instantaneous image maps and vector fields. The necessary circulation for the Counter Rotating Vortex Pair originates from a combined effect of steady in-hole, hanging and wake vortices. The strong cross-flow and jet interaction induces a three-dimensional waving, the stream-wise Counter Rotating Vortex Pair pair, leading to the formation of Ring Like Vortices. A secondary Counter Rotating Vortex Pair forms on top of the primary Counter Rotating Vortex Pair, resulting in mixing by "puffs". Overall, Stereoscopic Particle Image Velocimetry proofed capable of elucidating the Jet in Cross-Flow complex flow field. The gained insight in the mixing process will definitely contribute to the "Micromix" hydrogen combustion optimization.
Purpose
To assess potential cognitive deficits under the influence of static magnetic fields at various field strengths some studies already exist. These studies were not focused on attention as the most vulnerable cognitive function. Additionally, mostly no magnetic resonance imaging (MRI) sequences were performed.
Materials and Methods
In all, 25 right-handed men were enrolled in this study. All subjects underwent one MRI examination of 63 minutes at 1.5 T and one at 7 T within an interval of 10 to 30 days. The order of the examinations was randomized. Subjects were referred to six standardized neuropsychological tests strictly focused on attention immediately before and after each MRI examination. Differences in neuropsychological variables between the timepoints before and after each MRI examination were assessed and P-values were calculated
Results
Only six subtests revealed significant differences between pre- and post-MRI. In these tests the subjects achieved better results in post-MRI testing than in pre-MRI testing (P = 0.013–0.032). The other tests revealed no significant results.
Conclusion
The improvement in post-MRI testing is only explicable as a result of learning effects. MRI examinations, even in ultrahigh-field scanners, do not seem to have any persisting influence on the attention networks of human cognition immediately after exposure.
Fast response of Scots pine to improved water availability reflected in tree-ring width and δ13C

Drought-induced forest decline, like the Scots pine mortality in inner-Alpine valleys, will gain in importance as the frequency and severity of drought events are expected to increase. To understand how chronic drought affects tree growth and tree-ring δ13C values, we studied mature Scots pine in an irrigation experiment in an inner-Alpine valley. Tree growth and isotope analyses were carried out at the annual and seasonal scale. At the seasonal scale, maximum δ13C values were measured after the hottest and driest period of the year, and were associated with decreasing growth rates. Inter-annual δ13C values in early- and latewood showed a strong correlation with annual climatic conditions and an immediate decrease as a response to irrigation. This indicates a tight coupling between wood formation and the freshly produced assimilates for trees exposed to chronic drought. This rapid appearance of the isotopic signal is a strong indication for an immediate and direct transfer of newly synthesized assimilates for biomass production. The fast appearance and the distinct isotopic signal suggest a low availability of old stored carbohydrates. If this was a sign for C-storage depletion, an increasing mortality could be expected when stressors increase the need for carbohydrate for defence, repair or regeneration.
Autonomous robotic systems for penetrating thick ice shells with simultaneous collecting of scientific data are very promising devices in both terrestrial (glacier, climate research) and extra-terrestrial applications. Technical challenges in development of such systems are numerous and include 3D-navigation, an appropriate energy source, motion control, etc. Not less important is the problem of forward contamination of the pristine glacial environments with microorganisms and biomolecules from the surface of the probe. This study was devoted to establishing a laboratory model for microbial contamination of a newly constructed ice-melting probe called IceMole and to analyse the viability and amount of the contaminating microorganisms as a function of distance. The used bacterial strains were Bacillus subtilis (ATCC 6051) and Escherichia coli (ATCC 11775). The main objective was development of an efficient and reliable in-situ decontamination method of the melting probe. Therefore, several chemical substances were tested in respect of their efficacy to eliminate bacteria on the surface of the melting probe at low temperature (0 - 5 °C) and at continuous dilution by melted water. Our study has shown that at least 99.9% decontamination of the IceMole can be successfully achieved by the injection of 30% (v/v) hydrogen peroxide and 3% (v/v) sodium hypochlorite into the drilling site. We were able to reproduce this result in both time-dependent and depth-dependent experiments. The sufficient amount of 30% (v/v) H₂O₂ or 3% (v/v) NaClO has been found to be approximately 18 L per cm² of the probe’s surface.
Urinary stone formation has been evolved to a widespread disease during the last years. The reason for the formation of urinary stones are little crystals, mostly composed of calcium oxalate, which are formed in human kidneys. The early diagnosis of the risk for urinary stone formation of patients can be determined by the “Bonn-Risk-Index” method based on the potentiometric detection of the Ca2+-ion concentration and an optical determination of the triggered crystallisation of calcium oxalate in unprocessed urine. In this work, miniaturised capacitive field-effect EMIS (electrolyte-membrane-insulator-semiconductor) sensors have been developed for the determination of the Ca2+-ion concentration in human native urine. The Ca2+-sensitive EMIS sensors have been systematically characterised by impedance spectroscopy, capacitance–voltage and constant–capacitance method in terms of sensitivity, signal stability and response time in both CaCl2 solutions and in native urine. The obtained results demonstrate the suitability of EMIS sensors for the measurement of the Ca2+-ion concentration in native urine of patients.
The mission of the COMPASS-1 picosatellite is to take pictures of the earth, to validate a space-borne GPS receiver developed by the German Aerospace Center, and to verify the proper operation of the magnetic attitude control system in orbit. The spacecraft was launched on April 28, 2008 from the Indian space port Sriharikota, as part of the PSLV-C9 world record launch that simultaneously brought ten satellites into orbit. The mission operations were carried out from the ground stations in Aachen and Tainan. Arising difficulties in the communication link were overcome with the support of individuals from the amateur radio community. After several months of mission operation, abundant housekeeping and mission data has been commanded, received and analyzed and is presented in this paper.

Sie können die Beziehungen zwischen den Disziplinen fördern, indem sie bereits erzielte Arbeitsergebnisse disziplinenübergreifend bekannt machen und dadurch dazu beitragen, Synergiepotenziale und mögliche Kooperationspartner zu identifizieren.
The development of new interfaces for (bio-)chemical sensors requires comprehensive analyses and testing. The light-addressable potentiometric sensor (LAPS) can be used as a platform to investigate the sensitivity of a newly developed interface towards (bio-)chemical agents. LAPS measurements are spatially resolved by utilisation of focused light beams to define individual measurement spots. In this work, a new digitally modulated LAPS set-up based on an FPGA design will be introduced to increase the number of measurement spots, to shorten the measurement time and to improve the measurement accuracy.
We consider recent reports on small-world topologies of interaction networks derived from the dynamics of spatially extended systems that are investigated in diverse scientific fields such as neurosciences, geophysics, or meteorology. With numerical simulations that mimic typical experimental situations, we have identified an important constraint when characterizing such networks: indications of a small-world topology can be expected solely due to the spatial sampling of the system along with the commonly used time series analysis based approaches to network characterization.
Gas sensor investigation based on a catalytically activated thin-film thermopile for H2O2 detection

In aseptic filling systems, hydrogen peroxide vapour is commonly used for the reduction of microbial contaminations in carton packages. In this process, the germicidal efficiency of the vapour depends especially on the H₂O₂ concentration. To monitor the H₂O₂ concentration, a calorimetric H₂O₂ gas sensor based on a catalytically activated thin-film thermopile is investigated. Two different sensor layouts, namely a circular and a linear form, as well as two various material pairs such as tungsten/nickel and gold/nickel, have been examined for the realization of a thin-film thermopile. Additionally, manganese oxide and palladium particles have been compared as responsive catalysts towards H₂O₂. The thin-film sensors have been investigated at various H₂O₂ concentrations, gas temperatures and flow rates.
Among many approaches to address the high-level decision making problem for autonomous robots and agents, the robot program¬ming and plan language Golog follows a logic-based deliberative approach, and its successors were successfully deployed in a number of robotics applications over the past ten years. Usually, Golog interpreter are implemented in Prolog, which is not available for our target plat¬form, the bi-ped robot platform Nao. In this paper we sketch our first approach towards a prototype implementation of a Golog interpreter in the scripting language Lua. With the example of the elevator domain we discuss how the basic action theory is specified and how we implemented fluent regression in Lua. One possible advantage of the availability of a Non-Prolog implementation of Golog could be that Golog becomes avail¬able on a larger number of platforms, and also becomes more attractive for roboticists outside the Cognitive Robotics community.
Die vorliegende Arbeit zeichnet sich dadurch aus, dass registrierte unsegmentierte Volumina aus multimodalen Bilddatensätzen (z.B. MRT, PET) gleichzeitig in einer 3D-Rekonstruktion visualisiert werden und in Echtzeit manipuliert werden können. Ermöglicht wird die Echtzeitfähigkeit durch die Programmierung der Algorithmen zur direkten Volumenvisualisierung auf der Grafikkarte mittels der neuen CUDA-Technologie. Die Zuordnung der Farbeigenschaften wird über 1D-Transferfunktionen für jedes Volumen getrennt gesteuert. So können durch die interaktive Veränderung der 1D-Transferfunktion Detailinformationen aus den zwei Bilddatensätzen getrennt kontrolliert werden und die Vorteile der verschiedenen Bildmodalitäten in einer Visualisierung genutzt werden. Mittels dieses interaktiven Frameworks können neue Erkenntnisse insbesondere über neurodegenerativen Erkrankungen gewonnen werden.
Grassilage stellt einen nachwachsenden Rohstoff mit großem Potenzial dar. Neben Cellulose und Hemicellulose enthält sie auch organische Säuren, insbesondere Milchsäure. In einem Bioraffinerie-Projekt wird die Milchsäure aus der Silage isoliert und mit gentechnisch optimierten Stämmen zu L-Lysin weiterverarbeitet. Die Lignocellulose wird hydrolysiert und zu Ethanol fermentiert. Ein besonderes Augenmerk liegt auf der Integration der unterschiedlichen Prozesse sowie der einzelnen Prozessschritte zu einem Gesamtprozess, der sämtliche Inhaltsstoffe der Silage verwertet.
Die Anforderungen an das energiesparende Bauen sind mit der Einführung der Energieeinsparverordnung (EnEV) 2009 auch im Industrie- und Gewerbebau deutlich verschärft worden. Einen wesentlichen Beitrag zur Energieeinsparung liefert die Minimierung des Transmissionswärmetransfers. Analysiert man Gebäudehüllen in Metallleichtbauweise stellt man fest, dass eine Erhöhung der Wärmedämmstärke allein noch nicht zielführend ist, zusätzlich sind Wärmebrückeneffekte zu berücksichtigen und deren Einflüsse auf die Wärmetransmission zu reduzieren. Neben der Bedeutung für die Energieeinsparung ist eine wärmetechnisch optimierte Detailausbildung auch erforderlich, um einen ausreichenden Feuchteschutz (Vermeidung von Tauwasser und Schimmelpilz) zu realisieren und so Schäden zu vermeiden. Ein wichtiges Hilfsmittel stellt hierzu der vom Industrieverband für Bausysteme im Metallleichtbau (IFBS) herausgegebene Wärmebrückenatlas der Metall-Sandwichbauweise dar.
Gras hat ein hohes Potenzial als nachwachsender Rohstoff. Bei ungeeigneter Lagerung verderben Gräser allerdings innerhalb weniger Tage. Dieser Nachteil kann durch die Silierung des Grasschnittes behoben werden. Eines der wichtigsten in der Silage enthaltenen Produkte ist die Milchsäure. Um diese für weitere Aufarbeitungsschritte zugänglich zu machen, wird aus der Silage ein Presssaft hergestellt. Die Milchsäure wird aus einem Silagepresssaft mittels Extraktion durch ionische Flüssigkeiten isoliert. Dabei wird zum einen eine reine Milchsäure hergestellt, die z. B. für die Herstellung von Polymilchsäure genutzt werden kann. Zum anderen wird ein weniger aufgereinigter Extrakt gewonnen, der für die fermentative Produktion von L-Lysin und 1,2-Propandiol genutzt werden soll. Im Rahmen des Projekts erfolgt die gentechnische Optimierung von Corynebacterium glutamicum für die Umsetzung von Milchsäurezu L-Lysin. Die im nach der Pressung verbleibenden Presskuchen enthaltenen Grasfasern bestehen zu einem großen Teil aus Polysacchariden. Diese werden hydrolysiert und die dabei freigesetzten Zucker zu Grundchemikalien wie Ethanol oder Itakonsäure fermentiert. Im Rahmen einer vollständigen Nutzung der Silage wird das Raffinat aus der Milchsäureextraktion als Mediumsupplement in der Fermentation eingesetzt, was die Zugabe weiterer Medienbestandteile überflüssig macht. Die Rückstände der Hydrolysen und Fermentationen sollen darüberhinaus für die Herstellung von Biogas genutzt werden.

High Spatial Resolution 3D MRI of the Larynx Using a Dedicated TX/RX Phased Array Coil at 7.0T

MRI holds great potential for elucidating laryngeal and vocal fold anatomy together with the assessment of physiological processes associated in human phonation. However, MRI of human phonation remains very challenging due to the small size of the targeted structures, interfering signal from fat, air between the vocal folds and surrounding muscles and physiological motion. These anatomical/physiological constraints translate into stringent technical requirements in balancing, scan time, image contrast, immunity to physiological motion, temporal resolution and spatial resolution. Motivated by these challenges and limitations this study is aiming at translating the sensitivity gain at ultra-high magnetic fields for enhanced high spatial resolution 3D imaging of the larynx and vocal tract. To approach this goal a dedicated two channel TX/RX larynx coil is being proposed.
Durch den Einsatz magnetisierbarer Partikel lassen sich Stoffwechselprodukte direkt und selektiv aus feststoffreichen Fermentationssuspensionen abtrennen. Im Gegensatz zu klassischen Adsorbermaterialien können magnetisierbare Partikel mit sehr geringen Durchmessern verwendet werden. Zur deren Abtrennung ist jedoch ein hoher Magnetfeldgradient notwendig. Dieser wird in der Regel durch in der Trennkammer bzw. dem Magnetfeld eingebrachte magnetisierbare Drähte realisiert. Bei der Auslegung der Drahtgitter ist ein Kompromiss zwischen Abtrennrate und Durchlässigkeit nötig. Die Ausrichtung der Drähte in Relation zum Magnetfeld, deren Abstand sowie die geometrische Anordnung können hierbei variiert werden. Zum Verständnis der Einflüsse auf das sich ausbildende Magnetfeld und die Fluiddynamik wurden Simulationen mit der Finite-Elemente-Methode durchgeführt und experimentell überprüft. Hierfür wurden die Drähte unter Variation von Anzahl, Richtung und Anordnung in den Hochgradient-Magnetseparator eingebracht. Erste Verifizierungen der Simulationen zeigen, dass die in Magnetfeldrichtung ausgerichteten Drähte (x-Achse) über die geringste Partikelrückhaltefähigkeit verfügen. Die Drähte der y- und z-Achse halten den größten Anteil der Magnetpartikel zurück, wobei die Drähte in y-Richtung den höchsten Feldgradienten ausbilden. Des Weiteren konnte gezeigt werden, dass eine rhomboedrische Drahtanordnung der kubischen vorzuziehen ist.
Mouse nongenotoxic hepatocarcinogens phenobarbital (PB) and chlordane induce hepatomegaly characterized by hypertrophy and hyperplasia. Increased cell proliferation is implicated in the mechanism of tumor induction. The relevance of these tumors to human health is unclear. The xenoreceptors, constitutive androstane receptors (CARs), and pregnane X receptor (PXR) play key roles in these processes. Novel “humanized” and knockout models for both receptors were developed to investigate potential species differences in hepatomegaly. The effects of PB (80 mg/kg/4 days) and chlordane (10 mg/kg/4 days) were investigated in double humanized PXR and CAR (huPXR/huCAR), double knockout PXR and CAR (PXRKO/CARKO), and wild-type (WT) C57BL/6J mice. In WT mice, both compounds caused increased liver weight, hepatocellular hypertrophy, and cell proliferation. Both compounds caused alterations to a number of cell cycle genes consistent with induction of cell proliferation in WT mice. However, these gene expression changes did not occur in PXRKO/CARKO or huPXR/huCAR mice. Liver hypertrophy without hyperplasia was demonstrated in the huPXR/huCAR animals in response to both compounds. Induction of the CAR and PXR target genes, Cyp2b10 and Cyp3a11, was observed in both WT and huPXR/huCAR mouse lines following treatment with PB or chlordane. In the PXRKO/CARKO mice, neither liver growth nor induction of Cyp2b10 and Cyp3a11 was seen following PB or chlordane treatment, indicating that these effects are CAR/PXR dependent. These data suggest that the human receptors are able to support the chemically induced hypertrophic responses but not the hyperplastic (cell proliferation) responses. At this time, we cannot be certain that hCAR and hPXR when expressed in the mouse can function exactly as the genes do when they are expressed in human cells. However, all parameters investigated to date suggest that much of their functionality is maintained.
Hybrid control for autonomous systems — Integrating learning, deliberation and reactive control

The chemical imaging sensor is a semiconductor-based chemical sensor that can visualize the spatial distribution of chemical species. For the practical application of this sensor, artifacts in the chemical images due to defects of the semiconductor substrate and contamination of the sensing surface etc. have been a major problem. An image correction method was developed to eliminate the influence of nonuniformity of individual sensor plate.
Dexamethasone (DEX) is a potent and widely used anti-inflammatory and immunosuppressant glucocorticoid. It can bind and activate the pregnane X receptor (PXR), which plays a critical role as xenobiotic sensor in mammals to induce the expression of many enzymes, including cytochromes P450 in the CYP3A family. This induction results in its own metabolism. We have used a series of transgenic mouse lines, including a novel, improved humanized PXR line, to compare the induction profile of PXR-regulated drug-metabolizing enzymes after DEX administration, as well as looking at hepatic responses to rifampicin (RIF). The new humanized PXR model has uncovered further intriguing differences between the human and mouse receptors in that RIF only induced Cyp2b10 in the new humanized model. DEX was found to be a much more potent inducer of Cyp3a proteins in wild-type mice than in mice humanized for PXR. To assess whether PXR is involved in the detoxification of DEX in the liver, we analyzed the consequences of high doses of the glucocorticoid on hepatotoxicity on different PXR genetic backgrounds. We also studied these effects in an additional mouse model in which functional mouse Cyp3a genes have been deleted. These strains exhibited different sensitivities to DEX, indicating a protective role of the PXR and CYP3A proteins against the hepatotoxicity of this compound.
Background: One of the most prominent neurobiological models of alexithymia assumes an altered function of the anterior cingulate cortex (ACC) as the crucial neural correlate of alexithymia. So far functional imaging studies have yielded inconclusive results. Therefore, we tested this hypothesis in healthy alexithymics and nonalexithymics in an event-related fMRI study.
Methods: Thirty high- and 30 low-alexithymic right-handed male subjects (selected by the 20-item Toronto Alexithymia Scale, TAS-20) were investigated with event-related fMRI using a picture viewing paradigm. The stimuli consisted of happy, fearful and neutral facial expressions (Ekman-Friesen) as well as positive, negative and neutral pictures from the International Affective Picture System.
Results: Contrasting the high-alexithymic with the low-alexithymic group we observed increased activation of the supragenual ACC for different emotional valences as well as for different emotional stimuli. Moreover, there was a positive correlation of the ACC with the individual TAS-20 scores but no correlations with the individual Beck Depression Inventory scores. Additionally, there was no difference in activity of the amygdala.
Conclusions: We demonstrated that the supragenual ACC is constantly activated more strongly in alexithymic subjects and that this activation is related to the symptoms of alexithymia and not to associated symptoms such as depression. Therefore, our findings support the hypothesis of an altered function of the ACC in alexithymia.
