Capacitive field-effect electrolyte-insulator-semiconductor sensors consisting of an Al-p-Si-SiO2 structure have been used for the electrical detection of unlabelled single- and double-stranded DNA (dsDNA) molecules by their intrinsic charge. A simple functionalization protocol based on the layer-by-layer (LbL) technique was used to prepare a weak polyelectrolyte/probe-DNA bilayer, followed by the hybridization with complementary target DNA molecules. Due to the flat orientation of the LbL-adsorbed DNA molecules, a high sensor signal has been achieved. In addition, direct label-free detection of in-solution hybridized dsDNA molecules has been studied.
Use of textile structures for reinforcement of pelvic floor structures has to consider mechanical forces to the implant, which are quite different to the tension free conditions of the abdominal wall. Thus, biomechanical analysis of textile devices has to include the impact of strain on stretchability and effective porosity. Prolift® and Prolift + M®, developed for tension free conditions, were tested by measuring stretchability and effective porosity applying mechanical strain. For comparison, we used Dynamesh-PR4®, which was designed for pelvic floor repair to withstand mechanical strain. Prolift® at rest showed moderate porosity with little stretchability but complete loss of effective porosity at strain of 4.9 N/cm. Prolift + M® revealed an increased porosity at rest, but at strain showed high stretchability, with subsequent loss of effective porosity at strain of 2.5 N/cm. Dynamesh PR4® preserved its high porosity even under strain, but as consequence of limited stretchability. Though in tension free conditions Prolift® and Prolift + M® can be considered as large pore class I meshes, application of mechanical strain rapidly lead to collapse of pores. The loss of porosity at mechanical stress can be prevented by constructions with high structural stability. Assessment of porosity under strain was found helpful to define requirements for pelvic floor devices. Clinical studies have to prove whether devices with high porosity as well as high structural stability can improve the patients' outcome.
Background
Culture media containing complex compounds like yeast extract or peptone show numerous disadvantages. The chemical composition of the complex compounds is prone to significant variations from batch to batch and quality control is difficult. Therefore, the use of chemically defined media receives more and more attention in commercial fermentations. This concept results in better reproducibility, it simplifies downstream processing of secreted products and enable rapid scale-up. Culturing bacteria with unknown auxotrophies in chemically defined media is challenging and often not possible without an extensive trial-and-error approach. In this study, a respiration activity monitoring system for shake flasks and its recent version for microtiter plates were used to clarify unknown auxotrophic deficiencies in the model organism Bacillus pumilus DSM 18097.
Results
Bacillus pumilus DSM 18097 was unable to grow in a mineral medium without the addition of complex compounds. Therefore, a rich chemically defined minimal medium was tested containing basically all vitamins, amino acids and nucleobases, which are essential ingredients of complex components. The strain was successfully cultivated in this medium. By monitoring of the respiration activity, nutrients were supplemented to and omitted from the rich chemically defined medium in a rational way, thus enabling a systematic and fast determination of the auxotrophic deficiencies. Experiments have shown that the investigated strain requires amino acids, especially cysteine or histidine and the vitamin biotin for growth.
Conclusions
The introduced method allows an efficient and rapid identification of unknown auxotrophic deficiencies and can be used to develop a simple chemically defined tailor-made medium. B. pumilus DSM 18097 was chosen as a model organism to demonstrate the method. However, the method is generally suitable for a wide range of microorganisms. By combining a systematic combinatorial approach based on monitoring the respiration activity with cultivation in microtiter plates, high throughput experiments with high information content can be conducted. This approach facilitates media development, strain characterization and cultivation of fastidious microorganisms in chemically defined minimal media while simultaneously reducing the experimental effort.

The results of a statistical investigation of 42 fixed-wing, small to medium sized (20 kg−1000 kg) reconnaissance unmanned air vehicles (UAVs) are presented. Regression analyses are used to identify correlations of the most relevant geometry dimensions with the UAV’s maximum take-off mass. The findings allow an empirical based geometry-build up for a complete unmanned aircraft by referring to its take-off mass only. This provides a bridge between very early design stages (initial sizing) and the later determination of shapes and dimensions. The correlations might be integrated into a UAV sizing environment and allow designers to implement more sophisticated drag and weight estimation methods in this process. Additional information on correlation factors for a rough drag estimation methodology indicate how this technique can significantly enhance the accuracy of early design iterations.
The 2012 Emilia-Romagna earthquake, that mainly struck the homonymous Italian region provoking 28 casualties and damage to thousands of structures and infrastructures, is an exceptional source of information to question, investigate, and challenge the validity of seismic fragility functions and loss curves from an empirical standpoint. Among the most recent seismic events taking place in Europe, that of Emilia-Romagna is quite likely one of the best documented, not only in terms of experienced damages, but also for what concerns occurred losses and necessary reconstruction costs. In fact, in order to manage the compensations in a fair way both to citizens and business owners, soon after the seismic sequence, the regional administrative authority started (1) collecting damage and consequence-related data, (2) evaluating information sources and (3) taking care of the cross-checking of various reports. A specific database—so-called Sistema Informativo Gestione Europa (SFINGE)—was devoted to damaged business activities. As a result, 7 years after the seismic events, scientists can rely on a one-of-a-kind, vast and consistent database, containing information about (among other things): (1) buildings’ location and dimensions, (2) occurred structural damages, (3) experienced direct economic losses and (4) related reconstruction costs. The present work is focused on a specific data subset of SFINGE, whose elements are Long-Span-Beam buildings (mostly precast) deployed for business activities in industry, trade or agriculture. With the available set of data, empirical fragility functions, cost and loss ratio curves are elaborated, that may be included within existing Performance Based Earthquake Engineering assessment toolkits.
On the basis of independent and identically distributed bivariate random vectors, where the components are categorial and continuous variables, respectively, the related concomitants, also called induced order statistic, are considered. The main theoretical result is a functional central limit theorem for the empirical process of the concomitants in a triangular array setting. A natural application is hypothesis testing. An independence test and a two-sample test are investigated in detail. The fairly general setting enables limit results under local alternatives and bootstrap samples. For the comparison with existing tests from the literature simulation studies are conducted. The empirical results obtained confirm the theoretical findings.
Subject of this case is Deutsche Telekom Services Europe (DTSE), a service center for administrative processes. Due to the high volume of repetitive tasks (e.g., 100k manual uploads of offer documents into SAP per year), automation was identified as an important strategic target with a high management attention and commitment. DTSE has to work with various backend application systems without any possibility to change those systems. Furthermore, the complexity of administrative processes differed. When it comes to the transfer of unstructured data (e.g., offer documents) to structured data (e.g., MS Excel files), further cognitive technologies were needed.
Due to the high number of customer contacts, fault clearances, installations, and product provisioning per year, the automation level of operational processes has a significant impact on financial results, quality, and customer experience. Therefore, the telecommunications operator Deutsche Telekom (DT) has defined a digital strategy with the objectives of zero complexity and zero complaint, one touch, agility in service, and disruptive thinking. In this context, Robotic Process Automation (RPA) was identified as an enabling technology to formulate and realize DT’s digital strategy through automation of rule-based, routine, and predictable tasks in combination with structured and stable data.

(2012)
ETHICS is concerned with evaluating, measuring and making improvements in the thermal and energy performance of steel-clad and steel-framed buildings. It addresses basic building physics performance at a laboratory and full-scale level, and the preparation of design guidance for commercial, industrial and residential buildings. It includes the development of design tools to assist users in assessing whole-building performance, and calibrates these tools against whole-building measurements, which will be obtained from this research. Opportunities for renewable energy and other energy-saving features will be assessed. This project focuses on objectives that are of particular interest for the design of new steel constructions regarding energy efficiency. ETHICS investigates the as-built performance by on-site tests regarding air tightness and heat transfer properties of the building envelope and by monitoring the energy consumption and thermal comfort of selected up-to-date steel buildings. As energy efficiency is a key requirement for design and construction of buildings in the future, this project provides well-founded scientific data, which prove the high energy performance of current steel constructions and work out details for further improvements to maintain and extend the position of steel products in the construction sector.
New information regarding the influence of a stepped chute on the hydraulic performance of the United States Bureau of Reclamation (Reclamation) Type III hydraulic jump stilling basin is presented for design (steady) and adverse (decreasing tailwater) conditions. Using published experimental data and computational fluid dynamics (CFD) models, this paper presents a detailed comparison between smooth-chute and stepped-chute configurations for chute slopes of 0.8H:1V and 4H:1V and Froude numbers (F) ranging from 3.1 to 9.5 for a Type III basin designed for F = 8. For both stepped and smooth chutes, the relative role of each basin element was quantified, up to the most hydraulic extreme case of jump sweep-out. It was found that, relative to a smooth chute, the turbulence generated by a stepped chute causes a higher maximum velocity decay within the stilling basin, which represents an enhancement of the Type III basin’s performance but also a change in the relative role of the basin elements. Results provide insight into the ability of the CFD models [unsteady Reynolds-averaged Navier-Stokes (RANS) equations with renormalization group (RNG) k-ϵ turbulence model and volume-of-fluid (VOF) for free surface tracking] to predict the transient basin flow structure and velocity profiles. Type III basins can perform adequately with a stepped chute despite the effects steps have on the relative role of each basin element. It is concluded that the classic Type III basin design, based upon methodology by reclamation specific to smooth chutes, can be hydraulically improved for the case of stepped chutes for design and adverse flow conditions using the information presented herein.
The energy-efficiency of technical systems can be improved by a systematic design approach. Technical Operations Research (TOR) employs methods known from Operations Research to find a global optimal layout and operation strategy of technical systems. We show the practical usage of this approach by the systematic design of a decentralized water supply system for skyscrapers. All possible network options and operation strategies are modeled by a Mixed-Integer Nonlinear Program. We present the optimal system found by our approach and highlight the energy savings compared to a conventional system design.

For fuel flexibility enhancement hydrogen represents a possible alternative gas turbine fuel within future low emission power generation, in case of hydrogen production by the use of renewable energy sources such as wind energy or biomass. Kawasaki Heavy Industries, Ltd. (KHI) has research and development projects for future hydrogen society; production of hydrogen gas, refinement and liquefaction for transportation and storage, and utilization with gas turbine / gas engine for the generation of electricity. In the development of hydrogen gas turbines, a key technology is the stable and low NOx hydrogen combustion, especially Dry Low Emission (DLE) or Dry Low NOx (DLN) hydrogen combustion. Due to the large difference in the physical properties of hydrogen compared to other fuels such as natural gas, well established gas turbine combustion systems cannot be directly applied for DLE hydrogen combustion. Thus, the development of DLE hydrogen combustion technologies is an essential and challenging task for the future of hydrogen fueled gas turbines. The DLE Micro-Mix combustion principle for hydrogen fuel has been in development for many years to significantly reduce NOx emissions. This combustion principle is based on cross-flow mixing of air and gaseous hydrogen which reacts in multiple miniaturized “diffusion-type” flames. The major advantages of this combustion principle are the inherent safety against flashback and the low NOx-emissions due to a very short residence time of the reactants in the flame region of the micro-flames.
The chemical imaging sensor, which is based on the principle of the light-addressable potentiometric sensor (LAPS), is a powerful tool to visualize the spatial distribution of chemical species on the sensor surface. The spatial resolution of this sensor depends on the diffusion of photocarriers excited by a modulated light. In this study, a novel hybrid fiber-optic illumination was developed to enhance the spatial resolution. It consists of a modulated light probe to generate a photocurrent signal and a ring of constant light, which suppresses the lateral diffusion of minority carriers excited by the modulated light. It is demonstrated that the spatial resolution was improved from 92 μm to 68 μm.

Residential and commercial buildings account for more than one-third of global energy-related greenhouse gas emissions. Integrated multi-energy systems at the district level are a promising way to reduce greenhouse gas emissions by exploiting economies of scale and synergies between energy sources. Planning district energy systems comes with many challenges in an ever-changing environment. Computational modelling established itself as the state-of-the-art method for district energy system planning. Unfortunately, it is still cumbersome to combine standalone models to generate insights that surpass their original purpose. Ideally, planning processes could be solved by using modular tools that easily incorporate the variety of competing and complementing computational models. Our contribution is a vision for a collaborative development and application platform for multi-energy system planning tools at the district level. We present challenges of district energy system planning identified in the literature and evaluate whether this platform can help to overcome these challenges. Further, we propose a toolkit that represents the core technical elements of the platform. Lastly, we discuss community management and its relevance for the success of projects with collaboration and knowledge sharing at their core.
The development of a cost-effective hydrolysis for crude cellulose is an essential part of biorefinery developments. To establish such high solid hydrolysis, a new solid state reactor with static mixing is used. However, concentrations >10% (w/w) cause a rate and yield reduction of enzymatic hydrolysis. By optimizing the synergetic activity of cellulolytic enzymes at solid concentrations of 9%, 17% and 23% (w/w) of crude Organosolv cellulose, glucose concentrations of 57, 113 and 152 g L⁻¹ are reached. However, the glucose yield decreases from 0.81 to 0.72gg⁻¹ at 17% (w/w). Optimal conditions for hydrolysis scale-up under minimal enzyme addition are identified. As result, at 23% (w/w) crude cellulose the glucose yield increases from 0.29 to 0.49gg⁻¹. As proof of its applicability, biobutanol, succinic and itaconic acid are produced with the crude hydrolysate. The potential of the substrate is proven e.g. by a high butanol yield of 0.33gg⁻¹.
A semiconductor field-effect device has been used for an enzymatically catalyzed degradation of biopolymers for the first time. This novel technique is capable to monitor the degradation process of multiple samples in situ and in real-time. As model system, the degradation of the biopolymer poly(D, L-lactic acid) has been monitored in the degradation medium containing the enzyme lipase from Rhizomucor miehei. The obtained results demonstrate the potential of capacitive field-effect sensors for degradation studies of biodegradable polymers.
Capacitive field-effect sensors modified with a multi-enzyme membrane have been applied for an electronic transduction of biochemical signals processed by enzyme-based AND-Reset and OR-Reset logic gates. The local pH change at the sensor surface induced by the enzymatic reaction was used for the activation of the Reset function for the first time.
Enzyme-based logic gates and circuits - analytical applications and interfacing with electronics
(2017)
The paper is an overview of enzyme-based logic gates and their short circuits, with specific examples of Boolean AND and OR gates, and concatenated logic gates composed of multi-step enzyme-biocatalyzed reactions. Noise formation in the biocatalytic reactions and its decrease by adding a “filter” system, converting convex to sigmoid response function, are discussed. Despite the fact that the enzyme-based logic gates are primarily considered as components of future biomolecular computing systems, their biosensing applications are promising for immediate practical use. Analytical use of the enzyme logic systems in biomedical and forensic applications is discussed and exemplified with the logic analysis of biomarkers of various injuries, e.g., liver injury, and with analysis of biomarkers characteristic of different ethnicity found in blood samples on a crime scene. Interfacing of enzyme logic systems with modified electrodes and semiconductor devices is discussed, giving particular attention to the interfaces functionalized with signal-responsive materials. Future perspectives in the design of the biomolecular logic systems and their applications are discussed in the conclusion.
This article describes the fabrication, characterization and application of an epidermal temporary-transfer tattoo-based potentiometric sensor, coupled with a miniaturized wearable wireless transceiver, for real-time monitoring of sodium in the human perspiration. Sodium excreted during perspiration is an excellent marker for electrolyte imbalance and provides valuable information regarding an individual's physical and mental wellbeing. The realization of the new skin-worn non-invasive tattoo-like sensing device has been realized by amalgamating several state-of-the-art thick film, laser printing, solid-state potentiometry, fluidics and wireless technologies. The resulting tattoo-based potentiometric sodium sensor displays a rapid near-Nernstian response with negligible carryover effects, and good resiliency against various mechanical deformations experienced by the human epidermis. On-body testing of the tattoo sensor coupled to a wireless transceiver during exercise activity demonstrated its ability to continuously monitor sweat sodium dynamics. The real-time sweat sodium concentration was transmitted wirelessly via a body-worn transceiver from the sodium tattoo sensor to a notebook while the subjects perspired on a stationary cycle. The favorable analytical performance along with the wearable nature of the wireless transceiver makes the new epidermal potentiometric sensing system attractive for continuous monitoring the sodium dynamics in human perspiration during diverse activities relevant to the healthcare, fitness, military, healthcare and skin-care domains.

The industrial revolution especially in the IR4.0 era have driven many states of the art technologies to be introduced.
The automotive industry as well as many other key industries have also been greatly influenced. The rapid development of automotive industries in Europe have created wide industry gap between European Union (EU) and developing countries such as in South East Asia (SEA). Indulging this situation, FH JOANNEUM, Austria together with European partners from FH Aachen, Germany and Politecnico di Torino, Italy are taking initiative to close down the gap utilizing the Erasmus+ United Capacity Building in Higher Education grant from EU. A consortium was founded to engage with automotive technology transfer using the European framework to Malaysian, Indonesian and Thailand Higher Education Institutions (HEI) as well as automotive industries in respective countries. This could be achieved by establishing Engineering Knowledge Transfer Unit (EKTU) in respective SEA institutions guided by the industry partners in their respective countries. This EKTU could offer updated, innovative and high-quality training courses to increase graduate’s employability in higher education institutions and strengthen relations between HEI and the wider economic and social environment by addressing University-industry cooperation which is the regional priority for Asia. It is expected that, the Capacity Building Initiative would improve the quality of higher education and enhancing its relevance for the labor market and society in the SEA partners. The outcome of this project would greatly benefit the partners in strong and complementary partnership targeting the automotive industry and enhanced larger scale international cooperation between the European and SEA partners. It would also prepare the SEA HEI in sustainable partnership with Automotive industry in the region as a mean of income generation in the future.
In this work, we report on our attempt to design and implement an early introduction to basic robotics principles for children at kindergarten age. One of the main challenges of this effort is to explain complex robotics contents in a way that pre-school children could follow the basic principles and ideas using examples from their world of experience. What sets apart our effort from other work is that part of the lecturing is actually done by a robot itself and that a quiz at the end of the lesson is done using robots as well. The humanoid robot Pepper from Softbank, which is a great platform for human–robot interaction experiments, was used to present a lecture on robotics by reading out the contents to the children making use of its speech synthesis capability. A quiz in a Runaround-game-show style after the lecture activated the children to recap the contents they acquired about how mobile robots work in principle. In this quiz, two LEGO Mindstorm EV3 robots were used to implement a strongly interactive scenario. Besides the thrill of being exposed to a mobile robot that would also react to the children, they were very excited and at the same time very concentrated. We got very positive feedback from the children as well as from their educators. To the best of our knowledge, this is one of only few attempts to use a robot like Pepper not as a tele-teaching tool, but as the teacher itself in order to engage pre-school children with complex robotics contents.

Because of customer churn, strong competition, and operational inefficiencies, the telecommunications operator ME Telco (fictitious name due to confidentiality) launched a strategic transformation program that included a Business Process Management (BPM) project. Major problems were silo-oriented process management and missing cross-functional transparency. Process improvements were not consistently planned and aligned with corporate targets. Measurable inefficiencies were observed on an operational level, e.g., high lead times and reassignment rates of the incident management process.
We compare four different algorithms for automatically estimating the muscle fascicle angle from ultrasonic images: the vesselness filter, the Radon transform, the projection profile method and the gray level cooccurence matrix (GLCM). The algorithm results are compared to ground truth data generated by three different experts on 425 image frames from two videos recorded during different types of motion. The best agreement with the ground truth data was achieved by a combination of pre-processing with a vesselness filter and measuring the angle with the projection profile method. The robustness of the estimation is increased by applying the algorithms to subregions with high gradients and performing a LOESS fit through these estimates.
Grass silage provides a great potential as renewable feedstock. Two fractions of the grass silage, a press juice and the fiber fraction, were evaluated for their possible use for bioethanol production. Direct production of ethanol from press juice is not possible due to high concentrations of organic acids. For the fiber fraction, alkaline peroxide or enzymatic pretreatment was used, which removes the phenolic acids in the cell wall. In this study, we demonstrate the possibility to integrate the enzymatic pretreatment with a simultaneous saccharification and fermentation to achieve ethanol production from grass silage in a one-process step. Achieved yields were about 53 g ethanol per kg silage with the alkaline peroxide pretreatment and 91 g/kg with the enzymatic pretreatment at concentrations of 8.5 and 14.6 g/L, respectively. Furthermore, it was shown that additional supplementation of the fermentation medium with vitamins, trace elements and nutrient salts is not necessary when the press juice is directly used in the fermentation step.
