Due to their anion exchange characteristics, layered double hydroxides (LDHs) are suitable for the detoxification of aqueous, fatty acid containing fermentation substrates. The aim of this study is to examine the adsorption mechanism, using crude glycerol from plant oil esterification as a model system. Changes in the intercalation structure in relation to the amount of fatty acids adsorbed are monitored by X-ray diffraction and infra-red spectroscopy. Additionally, calcination of LDH is investigated in order to increase the binding capacity for fatty acids. Our data propose that, at ambient temperature, fatty acids can be bound to the hydrotalcite by adsorption or in addition by intercalation, depending on fatty acid concentration. The adsorption of fatty acids from crude glycerol shows a BET-like behavior. Above a fatty acid concentration of 3.5 g L−1, intercalation of fatty acids can be shown by the appearance of an increased interlayer spacing. This observation suggests a two phase adsorption process. Calcination of LDHs allows increasing the binding capacity for fatty acids by more than six times, mainly by reduction of structural CO32−.
Die fermentative Verwertung von Rohglycerin setzt je nach Herstellungsmethode und Produktionsorganismus eine Vorbehandlung des Glycerins zur Entfernung von Produktinhibitoren voraus. Durch den Einsatz von Hydrotalcit-Adsorbern können die im Rohglycerin enthaltenen Fettsäuren entfernt werden. Durch diese einfache Aufarbeitungsmethode ist ein mit reinem Glycerin vergleichbarer Umsatz von stark mit Fettsäuren verunreinigtem Rohglycerin zu 1,3-Propandiol (PDO) möglich. Die durch den Hydrotalcit gebundenen Fettsäuren lassen sich mit einem Ethanol-Wasser-Gemisch eluieren. Somit kann der Adsorber regeneriert und die Fettsäuren wieder der Wertschöpfungskette zugeführt werden. Im Fed-Batch-Experiment kann mit C. diolis eine PDO-Konzentration von über 50 g L⁻¹ unter Verwendung des aufgereinigten Rohglycerins erzielt werden. In der industriellen Produktion wird PDO momentan destillativ aufgearbeitet. Ein adsorptives Aufarbeitungsverfahren kann den Energiebedarf des Herstellungsprozesses drastisch senken. Auf der Suche nach einem geeigneten Material wurde ein Adsorberscreening in Bezug auf die Bindungseigenschaften durchgeführt. Mit einem b-Zeolith der Firma Süd ChemieAG konnte bisher die höchste Beladung im Modellsystem von 120 mg PDO/gAdsorber erreicht werden.
Die Lösungsmittelherstellung durch Clostridien konnte wirtschaftlich nicht mit der chemischen Synthese von Lösungsmitteln auf Erdölbasis konkurrieren und wurde in den frühen 1960er Jahren nahezu vollständig eingestellt. Das Interesse an nachwachsenden Rohstoffen hat in den letzten Jahren zu einem Wiederaufleben der ABE-Fermentation geführt. Aufgrund seiner höheren Energiedichte im Vergleich zu Ethanol ist Biobutanol als Energieträgerbesonders interessant und bietet sich z. B. als Produkt einer Bioraffinerie der 2. Generation an. Für die beschriebenen Experimente wird durch das Organosolv-Verfahren aufgeschlossenes Buchenholz verwendet. Der Faserstoff wird mithilfe von CTec2-Enzymen hydrolysiert, wobei der erhaltene Überstand eine Glucosekonzentration von 66 g L⁻¹ aufweist. Auf der Basis dieses Materials können mit Clostridium acetobutylicum Butanol-Ausbeuten erzielt werden, die mit denen unter Verwendung von reinen Zuckern vergleichbar sind. Dem Problem der hohen Produktinhibierung wird mit einer In-situ-Produktaufarbeitung begegnet. Mithilfe von Lösungsmittelimprägnierten Partikeln (SIPs) kann die Produktausbeute drastisch gesteigert werden, indem die gebildeten Lösungsmittel durch das auf dem Partikel imprägnierte Lösungsmittel während der Fermentation extrahiert werden. Zudem wird hierdurch die weitere Produktaufarbeitungstark vereinfacht.

The feasibility of light-addressed detection and manipulation of pH gradients inside an electrochemical microfluidic cell was studied. Local pH changes, induced by a light-addressable electrode (LAE), were detected using a light-addressable potentiometric sensor (LAPS) with different measurement modes representing an actuator-sensor system. Biosensor functionality was examined depending on locally induced pH gradients with the help of the model enzyme penicillinase, which had been immobilized in the microfluidic channel. The surface morphology of the LAE and enzyme-functionalized LAPS was studied by scanning electron microscopy. Furthermore, the penicillin sensitivity of the LAPS inside the microfluidic channel was determined with regard to the analyte’s pH influence on the enzymatic reaction rate. In a final experiment, the LAE-controlled pH inhibition of the enzyme activity was monitored by the LAPS.
Acetoin and diacetyl have a major impact on the flavor of alcoholic beverages such as wine or beer. Therefore, their measurement is important during the fermentation process. Until now, gas chromatographic techniques have typically been applied; however, these require expensive laboratory equipment and trained staff, and do not allow for online monitoring. In this work, a capacitive electrolyte–insulator–semiconductor sensor modified with tobacco mosaic virus (TMV) particles as enzyme nanocarriers for the detection of acetoin and diacetyl is presented. The enzyme acetoin reductase from Alkalihalobacillus clausii DSM 8716ᵀ is immobilized via biotin–streptavidin affinity, binding to the surface of the TMV particles. The TMV-assisted biosensor is electrochemically characterized by means of leakage–current, capacitance–voltage, and constant capacitance measurements. In this paper, the novel biosensor is studied regarding its sensitivity and long-term stability in buffer solution. Moreover, the TMV-assisted capacitive field-effect sensor is applied for the detection of diacetyl for the first time. The measurement of acetoin and diacetyl with the same sensor setup is demonstrated. Finally, the successive detection of acetoin and diacetyl in buffer and in diluted beer is studied by tuning the sensitivity of the biosensor using the pH value of the measurement solution.
Utilizing an appropriate enzyme immobilization strategy is crucial for designing enzyme-based biosensors. Plant virus-like particles represent ideal nanoscaffolds for an extremely dense and precise immobilization of enzymes, due to their regular shape, high surface-to-volume ratio and high density of surface binding sites. In the present work, tobacco mosaic virus (TMV) particles were applied for the co-immobilization of penicillinase and urease onto the gate surface of a field-effect electrolyte-insulator-semiconductor capacitor (EISCAP) with a p-Si-SiO₂-Ta₂O₅ layer structure for the sequential detection of penicillin and urea. The TMV-assisted bi-enzyme EISCAP biosensor exhibited a high urea and penicillin sensitivity of 54 and 85 mV/dec, respectively, in the concentration range of 0.1–3 mM. For comparison, the characteristics of single-enzyme EISCAP biosensors modified with TMV particles immobilized with either penicillinase or urease were also investigated. The surface morphology of the TMV-modified Ta₂O₅-gate was analyzed by scanning electron microscopy. Additionally, the bi-enzyme EISCAP was applied to mimic an XOR (Exclusive OR) enzyme logic gate.
Characterizing volcanic ash elements from the 2015 eruptions of bromo and raung volcanoes, Indonesia

The volcanic eruptions of Mt. Bromo and Mt. Raung in East Java, Indonesia, in 2015 perturbed volcanic materials and affected surface-layer air quality at surrounding locations. During the episodes, the volcanic ash from the eruptions influenced visibility, traffic accidents, flight schedules, and human health. In this research, the volcanic ash particles were collected and characterized by relying on the detail of physical observation. We performed an assessment of the volcanic ash elements to characterize the volcanic ash using two different methods which are aqua regia extracts followed by MP-AES and XRF laboratory test of bulk samples. The analysis results showed that the volcanic ash was mixed of many materials, such as Al, Si, P, K, Ca, Ti, V, Cr, Mn, Fe, Ni, and others. Fe, Si, Ca, and Al were found as the major elements, while the others were the trace elements Ba, Cr, Cu, Mn, P, Mn, Ni, Zn, Sb, Sr, and V with the minor concentrations. XRF analyses showed that Fe dominated the elements of the volcanic ash. The XRF analysis showed that Fe was at 35.40% in Bromo and 43.00% in Raung of the detected elements in bulk material. The results of aqua regia extracts analyzed by MP-AES were 1.80% and 1.70% of Fe element for Bromo and Raung volcanoes, respectively.
The composition and physiochemical properties of aquatic-phase natural organic matter (NOM) are most important problems for both environmental studies and water industry. Laser desorption/ionization (LDI) mass spectrometry facilitated successful examinations of NOM, as humic and fulvic acids in NOM are readily ionized by the nitrogen laser. In this study, hydrophobic NOMs (HPO NOMs) from river, reservoir and waste water were characterized by this technique. The effect of analytical variables like concentration, solvent composition and laser energy was investigated. The exact masses of small molecular NOM moieties in the range of 200–1200 m/z were determined in reflectron mode. In addition, spectra of post-source-decay experiments in this range showed that some compounds from different natural NOMs had the same fragmental ions. In the large mass range of 1200–15 000 Da, macromolecules and their aggregates were found in HPO NOMs from natural waters. Highly humic HPO exhibited mass peaks larger than 8000 Da. On the other hand, the waste water and reservoir water mainly had relatively smaller molecules of about 2000 Da. The LDI-MS measurements indicated that highly humic river waters were able to form large aggregates and membrane foulants, while the HPO NOMs from waste water and reservoir water were unlikely to form large aggregates. This short introduction is intended to provide a delimitation and clarification of the terminology as well as a classification of current biorefinery concepts. The basic process diagrams of the most important biorefinery types are shown.
The response of Bacillus licheniformis to heat and ethanol stress and the role of the SigB regulon

The emerging environmental issues due to the use of fossil resources are encouraging the exploration of new renewable resources. Biomasses are attracting more interest due to the low environmental impacts, low costs, and high availability on earth. In this scenario, green biorefineries are a promising platform in which green biomasses are used as feedstock. Grasses are mainly composed of cellulose and hemicellulose, and lignin is available in a small amount. In this work, a perennial ryegrass was used as feedstock to develop a green bio-refinery platform. Firstly, the grass was mechanically pretreated, thus obtaining a press juice and a press cake fraction. The press juice has high nutritional values and can be employed as part of fermentation media. The press cake can be employed as a substrate either in enzymatic hydrolysis or in solid-state fermentation. The overall aim of this work was to demonstrate different applications of both the liquid and the solid fractions. For this purpose, the filamentous fungus A. niger and the yeast Y. lipolythica were selected for their ability to produce citric acid. Finally, the possibility was assessed to use the press juice as part of fermentation media to cultivate S. cerevisiae and lactic acid bacteria for ethanol and lactic acid fermentation.

Ein viel versprechender erneuerbarer Rohstoff für die Produktion von Chemikalien und Treibstoffen ist Lignocellulose aus pflanzlicher Biomasse. Die darin enthaltenen Zucker können mittels enzymatischer Hydrolyse freigesetzt und fermentativ zu Ethanol umgesetzt werden. Ein interessanter Ansatz ist dabei die simultane Verzuckerung und Fermentation. Hefen und Enzyme haben mit 30 °C bzw. 50 °C zwar unterschiedliche Temperaturoptima, es konnte aber gezeigt werden, dass auch bei den niedrigeren Temperaturen eine Umsetzung der Cellulose zu Glucose erfolgt, wenn auch langsamer als bei optimalen Bedingungen. Außerdem konnte in Vorversuchen gezeigt werden, dass Ethanol in den zu erwartenden Konzentrationen keinen Einfluss auf die enzymatische Umsetzung hat.
Biomass from various types of organic waste was tested for possible use in hydrogen production. The composition consisted of lignified samples, green waste, and kitchen scraps such as fruit and vegetable peels and leftover food. For this purpose, the enzymatic pretreatment of organic waste with a combination of five different hydrolytic enzymes (cellulase, amylase, glucoamylase, pectinase and xylase) was investigated to determine its ability to produce hydrogen (H2) with the hydrolyzate produced here. In course, the anaerobic rod-shaped bacterium T. neapolitana was used for H2 production. First, the enzymes were investigated using different substrates in preliminary experiments. Subsequently, hydrolyses were carried out using different types of organic waste. In the hydrolysis carried out here for 48 h, an increase in glucose concentration of 481% was measured for waste loads containing starch, corresponding to a glucose concentration at the end of hydrolysis of 7.5 g·L−1. In the subsequent set fermentation in serum bottles, a H2 yield of 1.26 mmol H2 was obtained in the overhead space when Terrific Broth Medium with glucose and yeast extract (TBGY medium) was used. When hydrolyzed organic waste was used, even a H2 yield of 1.37 mmol could be achieved in the overhead space. In addition, a dedicated reactor system for the anaerobic fermentation of T. neapolitana to produce H2 was developed. The bioreactor developed here can ferment anaerobically with a very low loss of produced gas. Here, after 24 h, a hydrogen concentration of 83% could be measured in the overhead space.
Lignine bestehen aus einem hochgradig vernetzten Polymer phenolischer Grundeinheiten. Diese Verbindungen sind eine Quelle vielversprechender chemischer Grundbausteine. Auch die enzymatische Modifikation der Materialeigenschaften des Lignins ist für dessen Anwendung von Interesse. Aufgrund der verschiedenen Bindungstypen im Lignin ist eine Auftrennung mit nur einem Enzym unwahrscheinlich. Vielmehr sind verschiedene mediatorgestützte Reaktionen notwendig. Pilze, wie z.B. T. versicolor, nutzen Enzymkombinationen zum Aufschluss des Lignins. Hierbei kommen Laccase, Ligninperoxidase und Manganperoxidase zum Einsatz. Die optimale Kombination der Enzyme und ihrer Mediatoren bzw. Stabilisatoren ist Ziel der Untersuchungen. Aufgrund der großen Parameteranzahl wurde ein genetischer Algorithmus eingesetzt. Als Versuchsparameter wurden gewählt: die Verhältnisse der Enzyme, Ligninmasse, Konzentrationen an Eisen-, Mangan-, Oxalat-Ionen, ABTS, Violursäure und H₂O₂. Somit werden elf Parameter simultan optimiert. Als Algorithmus wurde ein Programm mit variabler Genkodierung entwickelt. Die Umsetzung des Lignins wird dabei über den verfolgt. Zurzeit ist ein enzymatischer Umsatz von 12% möglich. Als Referenz wurde eine chemische Lignindegradierung mit einem Umsatzvon 37% etabliert. Die sechs Generationen des Algorithmus zeigen eine Kongruenz der Enzymkonzentrationen von LiP, MnP und VeP, während Laccase keinen Einfluss hat. Des Weiteren beeinflussen die Konzentrationen von Mangan und Oxalat die Umsetzung, während die Variation von ABTS- und H₂O₂ nur eine geringe Auswirkung hat.
In diesem Beitrag wird die NIR- und MIR-Spektrometrie in Kombination mit multivariaten Kalibrationsmodellen zur Analyse von Monosacchariden und Cellulose aus Biomasse etabliert. Spektrengemischter Standardlösungen mit definierten Glucose- und Xylosekonzentrationen in Wasser werden im NIR-(Lambda 750, Perkin Elmer, USA) und MIR-Bereich (Spektrum 100, PerkinElmer) in Gegenwart von entweder Carboxymethylcellulose oder Grasfasern aufgenommen. Darauf basierend werden Kalibrationsmodelle (Unscrambler®, CAMO-Software AS, Norwegen) entwickelt und zur Vorhersage der Zuckerkonzentration in den Hydrolyseproben und der Celluloseanteile angewendet. Darüber hinaus wird die Partikelgröße der Rohstoffe bestimmt. Die Messergebnisse bilden die experimentelle Basis für die numerische Modellierung der Reaktionskinetik der enzymatischen Hydrolyse von Lignocellulose. Das Modell kombiniert die Bilanzierung der Partikelgrößenverteilungen mit der Multienzymkinetik. Dabei werden neben der Partikelgrößenverteilung und der Substratkonzentration die Zusammensetzung der Rohstoffe nach Vorbehandlung sowie die Produktinhibierung und mehrere enzymatische Aktivitäten berücksichtigt. Das Modell ermöglicht es, die Partikelgrößenverteilungen und die Konzentrationen der Substrate und Produkte während der Hydrolyse vorherzusagen und die kinetischen Parameter im Batch- sowie im Fed-Batch-Reaktor zu bestimmen.
A major part of edible oil is subjected to bleaching procedures, primarily with minerals applied as adsorbers. Their recycling is currently done either by regaining the oil via organic solvent extraction or by using the spent bleaching earth (SBE) as additive for animal feed, etc. As a new method, the reutilization of the by-product SBE for the microbiologic formation of acetone, butanol, and ethanol (ABE) is presented as proof-of-concept. The SBE was taken from a palm oil cleaning process. The recycling concept is based on the application of lipolytic clostridia strains. Due to considerably long fermentation times, co-fermentation with Candida rugosa and enzymatic hydrolyses of the bound oil with a subsequent clostridia fermentation are shown as alternative routes. Anaerobic fermentations under comparison of different clostridia strains were performed with glycerol media, enzymatically hydrolyzed palm oil and SBE. Solutes, side product compositions and productivities were quantified via HPLC. A successful production of ABE solutes from SBE has been done with a yield of 0.15 g butanol per gram of bound glycerol. Thus, the biotechnological recycling of the waste stream is possible in principle. Inhibition of the substrate suspension has been observed. A chromatographic ion-exchange of substrates increased the biomass concentration.

Bioprodukten eingesetzt. Die Abtrennung der beladenen Partikel erfolgt
durch einen neuen Rotor-Stator-Separator mit Hochgradient-Magnetfeld.
Biotechnologische Wertstoffgewinnung entlang der Prozessketten Grüner und Pflanzenöl-Bioraffinerien

Der nachwachsende Rohstoff Raps ist in großen Mengen verfügbar und eine Quelle für Biomoleküle mit hohem Wertschöpfungspotenzial. Entwicklungen zur biotechnologischen Wertstoffgewinnung werden dabei schwerpunktmäßig in den Bereichen Aufarbeitung und Funktionalisierung von Polyphenolen und Fetten betrieben. Bei der Verarbeitung der Pflanzenmaterialien werden dabei insbesondere Verfahren zur adsorptiven Aufreinigung und Auftrennung mittels Materialien mit modifizierten Bleicherden und anderen organischen oder anorganischen Adsorbentien untersucht. Ferner wurden für die Aufreinigung von Polyphenolen adsorptive sowie extraktive Prozesse entwickelt. Bei den Entwicklungen wird berücksichtigt, dass Bioraffinerien auf eine fortwährende Gewährleistung eines hohen Produktions- bzw. Lieferbedarfs nachwachsender Rohstoffe angewiesen sind. Somit werden Optionen dezentraler regionaler Vorbehandlungs- und Wertschöpfungsketten in der Nähe landwirtschaftlicher Betriebe einbezogen. Neben neuen Aufreinigungsverfahren werden mikrobielle und enzymatische Prozesse zur wertsteigernden Umsetzung von Glycerin, Polyphenolen und Zuckermonomeren vorgestellt sowie Limitierungen nachwachsender Rohstoffe der 2. Generation diskutiert.
Die Teilefertigung durch Rapid Prototyping (RP) verkürzt den Weg von der Idee bis zum Produkt, wobei unter anderem Optimierungszyklen in geringer Zeit durchlaufen werden können. Ferner eröffnen neue Entwicklungen in diesem Bereich die Möglichkeit individueller Produktionsverfahren. Im Unterschied zur klassischen Fertigung von Prototypen wird beim RP mit additiver Schichtfertigung (Additive Layer Manufacturing, ALM) gearbeitet. Je nach Methode werden Flüssigkeiten oder Pulver nach Vorgaben eines 3D-Computermodells sequentiell aufgetragen. Diese Verfahren existieren seit ca. 25 Jahren, jedoch sind seit kurzem ausgesprochen günstige Geräte verfügbar, die Objekte mit Genauigkeiten bis 20 lm fertigen können. Das RP hat in klinischen Anwendungsgebieten bzw. im Bereich des Tissue Engineering bereits vielfach Einzug gefunden. Aber auch chemisch-biotechnologische Entwicklungen können von den Verfahren profitieren. So wurden Mikrofluidiksysteme und Bioreaktoren bereits erfolgreich durch RP gefertigt. Durch ALM ist ebenso die Herstellung von Reaktionseinheiten aus biokompatiblen Materialien wie ionotropen Gelen möglich. Ferner sind sehr komplexe Strukturierungen von Oberflächen im Nanometerbereich realisierbar, die für die Auftragung heterogener Katalysatoren oder auch Mikroorganismen eingesetzt werden können. Auch der Bereich Reaktoren- und Apparatebau kann von den Fortschritten in der additiven Fertigung profitieren. Verfahren wie selektives Laser- oder Elektronenstrahlschmelzen erlauben es, metallische Komponenten in nahezu beliebigen Geometrien zu fertigen. Somit können Strukturen verwirklicht werden, die mit konventionellen Fertigungstechniken nur sehr schwer oder überhauptnicht herstellbar wären. Durch Anwendung von rechnergestützter Modellierung können optimale Strukturen identifiziert und additiv gefertigt werden. Eine anschließende katalytische Funktionalisierung der Oberfläche ermöglicht die Herstellung strukturierter Reaktoren mit maßgeschneiderten Eigenschaften.
