Humanized UGT2 and CYP3A transchromosomic rats for improved prediction of human drug metabolism

Mouse nongenotoxic hepatocarcinogens phenobarbital (PB) and chlordane induce hepatomegaly characterized by hypertrophy and hyperplasia. Increased cell proliferation is implicated in the mechanism of tumor induction. The relevance of these tumors to human health is unclear. The xenoreceptors, constitutive androstane receptors (CARs), and pregnane X receptor (PXR) play key roles in these processes. Novel “humanized” and knockout models for both receptors were developed to investigate potential species differences in hepatomegaly. The effects of PB (80 mg/kg/4 days) and chlordane (10 mg/kg/4 days) were investigated in double humanized PXR and CAR (huPXR/huCAR), double knockout PXR and CAR (PXRKO/CARKO), and wild-type (WT) C57BL/6J mice. In WT mice, both compounds caused increased liver weight, hepatocellular hypertrophy, and cell proliferation. Both compounds caused alterations to a number of cell cycle genes consistent with induction of cell proliferation in WT mice. However, these gene expression changes did not occur in PXRKO/CARKO or huPXR/huCAR mice. Liver hypertrophy without hyperplasia was demonstrated in the huPXR/huCAR animals in response to both compounds. Induction of the CAR and PXR target genes, Cyp2b10 and Cyp3a11, was observed in both WT and huPXR/huCAR mouse lines following treatment with PB or chlordane. In the PXRKO/CARKO mice, neither liver growth nor induction of Cyp2b10 and Cyp3a11 was seen following PB or chlordane treatment, indicating that these effects are CAR/PXR dependent. These data suggest that the human receptors are able to support the chemically induced hypertrophic responses but not the hyperplastic (cell proliferation) responses. At this time, we cannot be certain that hCAR and hPXR when expressed in the mouse can function exactly as the genes do when they are expressed in human cells. However, all parameters investigated to date suggest that much of their functionality is maintained.
Der Erhalt möglichst hoher Zuckerkonzentrationen für nachfolgende Fermentationen und eine Steigerung der Produktivität sind Ziele der Hydrolyse bei hohen Feststoffkonzentrationen im Rahmen des Projekts „Lignocellulose Bioraffinerie“. Verwendet wird durch ein Organosolv-Verfahren aufgeschlossenes Buchenholz. Die Hydrolyse des Faserstoffes erfolgt mithilfe von CTec2-Enzymen (Fa. Novozymes). Zurzeit können unter Einsatz eines neuen Feststoffreaktors Cellulosefasern in einer Konzentration bis 400 g L⁻¹ enzymatisch hydrolysiert werden. Dabei werden Ausbeuten (g Glucose/g Cellulose im Faserstoff) bis 0,86 g g⁻¹ und Glucosekonzentrationenvon 120 g L⁻¹ erreicht. Ein Nachteil ist jedoch die hierbei auftretende Abnahme der Hydrolyseausbeuten. Zahlreiche Limitierungen bezüglich der Hydrolysierbarkeit von Lignocellulose werden zurzeit diskutiert und publiziert. Ziel der Untersuchungen ist die Identifizierung hydrolysehemmender Substanzen sowie die Erhöhung der Ausbeute an Zuckermonomeren durch den Einsatz lignolytischer Enzyme. Hierbei wird eine HPLC-MS-Methode zur Charakterisierung hemmender Substanzen eingesetzt, um potenzielle Inhibitoren zu erfassen.
Durch den Einsatz magnetisierbarer Partikel lassen sich Stoffwechselprodukte direkt und selektiv aus feststoffreichen Fermentationssuspensionen abtrennen. Im Gegensatz zu klassischen Adsorbermaterialien können magnetisierbare Partikel mit sehr geringen Durchmessern verwendet werden. Zur deren Abtrennung ist jedoch ein hoher Magnetfeldgradient notwendig. Dieser wird in der Regel durch in der Trennkammer bzw. dem Magnetfeld eingebrachte magnetisierbare Drähte realisiert. Bei der Auslegung der Drahtgitter ist ein Kompromiss zwischen Abtrennrate und Durchlässigkeit nötig. Die Ausrichtung der Drähte in Relation zum Magnetfeld, deren Abstand sowie die geometrische Anordnung können hierbei variiert werden. Zum Verständnis der Einflüsse auf das sich ausbildende Magnetfeld und die Fluiddynamik wurden Simulationen mit der Finite-Elemente-Methode durchgeführt und experimentell überprüft. Hierfür wurden die Drähte unter Variation von Anzahl, Richtung und Anordnung in den Hochgradient-Magnetseparator eingebracht. Erste Verifizierungen der Simulationen zeigen, dass die in Magnetfeldrichtung ausgerichteten Drähte (x-Achse) über die geringste Partikelrückhaltefähigkeit verfügen. Die Drähte der y- und z-Achse halten den größten Anteil der Magnetpartikel zurück, wobei die Drähte in y-Richtung den höchsten Feldgradienten ausbilden. Des Weiteren konnte gezeigt werden, dass eine rhomboedrische Drahtanordnung der kubischen vorzuziehen ist.
Clostridium propionicum is the only organism known to ferment β-alanine, a constituent of coenzyme A (CoA) and the phosphopantetheinyl prosthetic group of holo-acyl carrier protein. The first step in the fermentation is a CoA-transfer to β-alanine. Subsequently, the resulting β-alanyl-CoA is deaminated by the enzyme β-alanyl-CoA:ammonia lyase (Acl) to reversibly form ammonia and acrylyl-CoA. We have determined the crystal structure of Acl in its apo-form at a resolution of 0.97 Å as well as in complex with CoA at a resolution of 1.59 Å. The structures reveal that the enyzme belongs to a superfamily of proteins exhibiting a so called “hot dog fold” which is characterized by a five-stranded antiparallel β-sheet with a long α-helix packed against it. The functional unit of all “hot dog fold” proteins is a homodimer containing two equivalent substrate binding sites which are established by the dimer interface. In the case of Acl, three functional dimers combine to a homohexamer strongly resembling the homohexamer formed by YciA-like acyl-CoA thioesterases. Gräser sind in der Lage, einen großen Teil der für eine biobasierte Wirtschaft benötigten Biomasse zur Verfügung zustellen. Um eine ganzjährige Nutzung des Grases zu gewährleisten, muss eine stabile Lagerung des Grases erreicht werden, was z. B. durch Silieren möglich ist. Die konservierende Wirkung der Silierung beruht auf der Bildung organischer Säuren. Um diese zu gewinnen, wird die Silage gepresst, die organischen Säuren über Flüssig/Flüssig-Extraktion aus dem Presssaft abgetrenntund mittels chromatographischer Methoden weiter aufgereinigt. Im präsentierten Konzept werden die im Presskuchen enthaltenen Lignocellulosen hydrolysiert und die erhaltenen Monosaccharide zu Ethanol fermentiert. Die Phenolsäuren, die in Gräsern die Rolle des Lignins übernehmen, können simultan mit der Hydrolyse der Polysaccharide enzymatisch abgetrennt und als Nebenprodukt gewonnen werden. Die nach der Abtrennung des Ethanols verbleibenden Fermentationsreststoffe werden für die Herstellung von Biogas verwendet.

Gras hat ein hohes Potenzial als nachwachsender Rohstoff. Bei ungeeigneter Lagerung verderben Gräser allerdings innerhalb weniger Tage. Dieser Nachteil kann durch die Silierung des Grasschnittes behoben werden. Eines der wichtigsten in der Silage enthaltenen Produkte ist die Milchsäure. Um diese für weitere Aufarbeitungsschritte zugänglich zu machen, wird aus der Silage ein Presssaft hergestellt. Die Milchsäure wird aus einem Silagepresssaft mittels Extraktion durch ionische Flüssigkeiten isoliert. Dabei wird zum einen eine reine Milchsäure hergestellt, die z. B. für die Herstellung von Polymilchsäure genutzt werden kann. Zum anderen wird ein weniger aufgereinigter Extrakt gewonnen, der für die fermentative Produktion von L-Lysin und 1,2-Propandiol genutzt werden soll. Im Rahmen des Projekts erfolgt die gentechnische Optimierung von Corynebacterium glutamicum für die Umsetzung von Milchsäurezu L-Lysin. Die im nach der Pressung verbleibenden Presskuchen enthaltenen Grasfasern bestehen zu einem großen Teil aus Polysacchariden. Diese werden hydrolysiert und die dabei freigesetzten Zucker zu Grundchemikalien wie Ethanol oder Itakonsäure fermentiert. Im Rahmen einer vollständigen Nutzung der Silage wird das Raffinat aus der Milchsäureextraktion als Mediumsupplement in der Fermentation eingesetzt, was die Zugabe weiterer Medienbestandteile überflüssig macht. Die Rückstände der Hydrolysen und Fermentationen sollen darüberhinaus für die Herstellung von Biogas genutzt werden.
Reconstructive surgery and tissue replacements like ureters or bladders reconstruction have been recently studied, taking into account growth and remodelling of cells since living cells are capable of growing, adapting, remodelling or degrading and restoring in order to deform and respond to stimuli. Hence, shapes of ureters or bladders and their microstructure change during growth and these changes strongly depend on external stimuli such as training. We present the mechanical stimulation of smooth muscle cells in a tubular fibrin-PVDFA scaffold and the modelling of the growth of tissue by stimuli. To this end, mechanotransduction was performed with a kyphoplasty balloon catheter that was guided through the lumen of the tubular structure. The bursting pressure was examined to compare the stability of the incubated tissue constructs. The results showed the significant changes on tissues with training by increasing the burst pressure as a characteristic mechanical property and the smooth muscle cells were more oriented with uniformly higher density. Besides, the computational growth models also exhibited the accurate tendencies of growth of the cells under different external stimuli. Such models may lead to design standards for the better layered tissue structure in reconstructing of tubular organs characterized as composite materials such as intestines, ureters and arteries.

Der zunehmende Bedarf an fossilen Rohstoffen bei gleichzeitig abnehmender Versorgungssicherheit führt zu einer intensiven Suche nach erneuerbaren Ressourcen. Ein vielversprechendes Ausgangsmaterial mit einer weltweiten Verfügbarkeit stellt Gras dar. In 2012 wurden in Deutschland 33 Millionen Tonnen (Heugewicht) Gras auf 4,82 Millionen Hektar Ackerland produziert, davon wurden 60,5 % siliert. Durch die Silierung kann Gras als Substrat zeitlich uneingeschränkt verfügbar sein, ohne dem Risiko des schnellen Verderbs ausgesetzt zu sein. Eine Schlüsselrolle im Rahmen des Silierprozesses nimmt die Produktion von Milchsäure ein. Milchsäure ist einbedeutendes biotechnologisches Produkt für die Lebensmittel- und die chemische Industrie. Im Rahmen dieser Arbeit wird die vollständige Umwandlung der fermentierbaren Zucker in der Silage zu Milchsäure angestrebt, um die maximale Ausbeute der organischen Säure zu erreichen. Im ersten Verfahrensschritt wird die Silage gepresst und der erhaltene Presskuchen einer Liquid-Hot-Water-Behandlung unterzogen. Durch diese einfache Vorbehandlung können hohe Glucoseausbeuten im nachfolgenden SSF-Schritt bei gleichzeitig geringem Enzymeinsatz und Chemikalienverbrauch realisiert werden. Zur Aufreinigung der Milchsäure wurden extraktive und chromatographische Methoden untersucht.
Grassilage stellt einen nachwachsenden Rohstoff mit großem Potenzial dar. Neben Cellulose und Hemicellulose enthält sie auch organische Säuren, insbesondere Milchsäure. In einem Bioraffinerie-Projekt wird die Milchsäure aus der Silage isoliert und mit gentechnisch optimierten Stämmen zu L-Lysin weiterverarbeitet. Die Lignocellulose wird hydrolysiert und zu Ethanol fermentiert. Ein besonderes Augenmerk liegt auf der Integration der unterschiedlichen Prozesse sowie der einzelnen Prozessschritte zu einem Gesamtprozess, der sämtliche Inhaltsstoffe der Silage verwertet.
1. Drug metabolizing enzymes and transporters play important roles in the absorption, metabolism, tissue distribution and excretion of various compounds and their metabolites and thus can significantly affect their efficacy and safety. Furthermore, they can be involved in drug–drug interactions which can result in adverse responses, life-threatening toxicity or impaired efficacy. Significant species differences in the interaction of compounds with drug metabolizing enzymes and transporters have been described.
2. In order to overcome the limitation of animal models in accurately predicting human responses, a large variety of mouse models humanized for drug metabolizing enzymes and to a lesser extent drug transporters have been created.
3. This review summarizes the literature describing these mouse models and their key applications in studying the role of drug metabolizing enzymes and transporters in drug bioavailability, tissue distribution, clearance and drug–drug interactions as well as in human metabolite testing and risk assessment.
4. Though such humanized mouse models have certain limitations, there is great potential for their use in basic research and for testing and development of new medicines. These limitations and future potentials will be discussed.
Genetically humanized mice for proteins involved in drug metabolism and toxicity and mice engrafted with human hepatocytes are emerging as promising in vivo models for improved prediction of the pharmacokinetic, drug–drug interaction, and safety characteristics of compounds in humans. This is an overview on the genetically humanized and chimeric liver-humanized mouse models, which are illustrated with examples of their utility in drug metabolism and toxicity studies. The models are compared to give guidance for selection of the most appropriate model by highlighting advantages and disadvantages to be carefully considered when used for studies in drug discovery and development.
Disruption experiments targeted at the Bacillus licheniformis degSU operon and GFP-reporter analysis provided evidence for promoter activity immediately upstream of degU. pMutin mediated concomitant introduction of the degU32 allele – known to cause hypersecretion in Bacillus subtilis – resulted in a marked increase in protease activity. Application of 5-fluorouracil based counterselection through establishment of a phosphoribosyltransferase deficient Δupp strain eventually facilitated the marker-free introduction of degU32 leading to further protease enhancement achieving levels as for hypersecreting wild strains in which degU was overexpressed. Surprisingly, deletion of rapG – known to interfere with DegU DNA-binding in B. subtilis – did not enhance protease production neither in the wild type nor in the degU32 strain. The combination of degU32 and Δupp counterselection in the type strain is not only equally effective as in hypersecreting wild strains with respect to protease production but furthermore facilitates genetic strain improvement aiming at biological containment and effectiveness of biotechnological processes.
Compared with rodents and many other animal species, the human cytochrome P450 (P450) Cyp2c gene cluster varies significantly in the multiplicity of functional genes and in the substrate specificity of its enzymes. As a consequence, the use of wild-type animal models to predict the role of human CYP2C enzymes in drug metabolism and drug-drug interactions is limited. Within the human CYP2C cluster CYP2C9 is of particular importance, because it is one of the most abundant P450 enzymes in human liver, and it is involved in the metabolism of a wide variety of important drugs and environmental chemicals. To investigate the in vivo functions of cytochrome P450 Cyp2c genes and to establish a model for studying the functions of CYP2C9 in vivo, we have generated a mouse model with a deletion of the murine Cyp2c gene cluster and a corresponding humanized model expressing CYP2C9 specifically in the liver. Despite the high number of functional genes in the mouse Cyp2c cluster and the reported roles of some of these proteins in different biological processes, mice deleted for Cyp2c genes were viable and fertile but showed certain phenotypic alterations in the liver. The expression of CYP2C9 in the liver also resulted in viable animals active in the metabolism and disposition of a number of CYP2C9 substrates. These mouse lines provide a powerful tool for studying the role of Cyp2c genes and of CYP2C9 in particular in drug disposition and as a factor in drug-drug interaction.
Generation and Characterization of a Novel Multidrug Resistance Protein 2 Humanized Mouse Line

The multidrug resistance protein (MRP) 2 is predominantly expressed in liver, intestine, and kidney, where it plays an important role in the excretion of a range of drugs and their metabolites or endogenous compounds into bile, feces, and urine. Mrp knockout [Mrp2(−/−)] mice have been used recently to study the role of MRP2 in drug disposition. Here, we describe the first generation and initial characterization of a mouse line humanized for MRP2 (huMRP2), which is nulled for the mouse Mrp2 gene and expresses the human transporter in the organs and cell types where MRP2 is normally expressed. Analysis of the mRNA expression for selected cytochrome P450 and transporter genes revealed no major changes in huMRP2 mice compared with wild-type controls. We show that human MRP2 is able to compensate functionally for the loss of the mouse transporter as demonstrated by comparable bilirubin levels in the humanized mice and wild-type controls, in contrast to the hyperbilirubinemia phenotype that is observed in MRP2(−/−) mice. The huMRP2 mouse provides a model to study the role of the human transporter in drug disposition and in assessing the in vivo consequences of inhibiting this transporter by compounds interacting with human MRP2.
Breast cancer resistance protein (BCRP) is expressed in various tissues, such as the gut, liver, kidney and blood brain barrier (BBB), where it mediates the unidirectional transport of substrates to the apical/luminal side of polarized cells. Thereby BCRP acts as an efflux pump, mediating the elimination or restricting the entry of endogenous compounds or xenobiotics into tissues and it plays important roles in drug disposition, efficacy and safety. Bcrp knockout mice (Bcrp−/−) have been used widely to study the role of this transporter in limiting intestinal absorption and brain penetration of substrate compounds. Here we describe the first generation and characterization of a mouse line humanized for BCRP (hBCRP), in which the mouse coding sequence from the start to stop codon was replaced with the corresponding human genomic region, such that the human transporter is expressed under control of the murine Bcrp promoter. We demonstrate robust human and loss of mouse BCRP/Bcrp mRNA and protein expression in the hBCRP mice and the absence of major compensatory changes in the expression of other genes involved in drug metabolism and disposition. Pharmacokinetic and brain distribution studies with several BCRP probe substrates confirmed the functional activity of the human transporter in these mice. Furthermore, we provide practical examples for the use of hBCRP mice to study drug-drug interactions (DDIs). The hBCRP mouse is a promising model to study the in vivo role of human BCRP in limiting absorption and BBB penetration of substrate compounds and to investigate clinically relevant DDIs involving BCRP.
Dargestellt und beansprucht ist eine Funkeinheit mit einem Funkbauteil, einer elektrisch leitfähigen Antenne und einem Kunststoffverbindungsmittel. Das Funkbauteil weist einen Antennenanschluss auf. Das Kunststoffverbindungsmittel weist ein erstes Elastomermaterial mit elektrisch leitfähigem Zusatzmaterial auf, so dass das Kunststoffverbindungsmittel elektrisch leitfähig ist. Das Kunststoffverbindungsmittel bildet eine mechanische und elektrisch leitfähige Verbindung zwischen dem Antennenanschluss und der Antenne. Weiterhin wird eine Vorrichtung mit einem ein Matrixmaterial mit oder aus einem zweiten Elastomermaterial und einer Funkeinheit beschrieben und beansprucht, wobei die Funkeinheit vollständig in das Matrixmaterial eingebettet ist. Die Vorrichtung kann insbesondere ein Fahrzeugreifen sein.
