Halophilic and halotolerant microorganisms represent a promising source of salt-tolerant enzymes suitable for various biotechnological applications where high salt concentrations would otherwise limit enzymatic activity. Considering the current growing enzyme market and the need for more efficient and new biocatalysts, the present study aimed at the characterization of a high-alkaline subtilisin from Alkalihalobacillus okhensis Kh10-101T. The protease gene was cloned and expressed in Bacillus subtilis DB104. The recombinant protease SPAO with 269 amino acids belongs to the subfamily of high-alkaline subtilisins. The biochemical characteristics of purified SPAO were analyzed in comparison with subtilisin Carlsberg, Savinase, and BPN'. SPAO, a monomer with a molecular mass of 27.1 kDa, was active over a wide range of pH 6.0–12.0 and temperature 20–80 °C, optimally at pH 9.0–9.5 and 55 °C. The protease is highly oxidatively stable to hydrogen peroxide and retained 58% of residual activity when incubated at 10 °C with 5% (v/v) H2O2 for 1 h while stimulated at 1% (v/v) H2O2. Furthermore, SPAO was very stable and active at NaCl concentrations up to 5.0 m. This study demonstrates the potential of SPAO for biotechnological applications in the future.
Subtilisins from microbial sources, especially from the Bacillaceae family, are of particular interest for biotechnological applications and serve the currently growing enzyme market as efficient and novel biocatalysts. Biotechnological applications include use in detergents, cosmetics, leather processing, wastewater treatment and pharmaceuticals. To identify a possible candidate for the enzyme market, here we cloned the gene of the subtilisin SPFA from Fictibacillus arsenicus DSM 15822ᵀ (obtained through a data mining-based search) and expressed it in Bacillus subtilis DB104. After production and purification, the protease showed a molecular mass of 27.57 kDa and a pI of 5.8. SPFA displayed hydrolytic activity at a temperature optimum of 80 °C and a very broad pH optimum between 8.5 and 11.5, with high activity up to pH 12.5. SPFA displayed no NaCl dependence but a high NaCl tolerance, with decreasing activity up to concentrations of 5 m NaCl. The stability enhanced with increasing NaCl concentration. Based on its substrate preference for 10 synthetic peptide 4-nitroanilide substrates with three or four amino acids and its phylogenetic classification, SPFA can be assigned to the subgroup of true subtilisins. Moreover, SPFA exhibited high tolerance to 5% (w/v) SDS and 5% H₂O₂ (v/v). The biochemical properties of SPFA, especially its tolerance of remarkably high pH, SDS and H₂O₂, suggest it has potential for biotechnological applications.

Bestimmung der metabolischen Aktivität von Mikroorganismen während des Biogasbildungsprozesses

Lignin is a promising renewable biopolymer being investigated worldwide as an environmentally benign substitute of fossil-based aromatic compounds, e.g. for the use as an excipient with antioxidant and antimicrobial properties in drug delivery or even as active compound. For its successful implementation into process streams, a quick, easy, and reliable method is needed for its molecular weight determination. Here we present a method using 1H spectra of benchtop as well as conventional NMR systems in combination with multivariate data analysis, to determine lignin’s molecular weight (Mw and Mn) and polydispersity index (PDI). A set of 36 organosolv lignin samples (from Miscanthus x giganteus, Paulownia tomentosa and Silphium perfoliatum) was used for the calibration and cross validation, and 17 samples were used as external validation set. Validation errors between 5.6% and 12.9% were achieved for all parameters on all NMR devices (43, 60, 500 and 600 MHz). Surprisingly, no significant difference in the performance of the benchtop and high-field devices was found. This facilitates the application of this method for determining lignin’s molecular weight in an industrial environment because of the low maintenance expenditure, small footprint, ruggedness, and low cost of permanent magnet benchtop NMR systems.
In diesem Beitrag geht es um die Integration von Stoffströmen einer Lignocellulose-Bioraffinerie in Verfahren zur Batterieherstellung. Pflanzliche Reststoffe aus der Biokraftstoffherstellung wie Lignin sollen zur Herstellung neuer Batteriematerialien verwendet werden. Hierbei wird das Lignin als Matrix für die vorgraphitischen C-haltigen Einlagerungsverbindungen in den Elektroden genutzt. Die Si-C-Komposite werden durch das Einbetten von Si in eine Ligninmatrix mit anschließender Carbonisierung hergestellt. Das Lignin hierfür wird durch die sequentielle hydrothermale Vorbehandlung von Buchenholz bei variablen Bedingungen gewonnen und mit Si-Nanopartikel sowie als Referenz ohne Si-Nanopartikel gefällt. Die Ergebnisse zeigen, dass die sequenzielle Vorbehandlung höhere Ausbeuten im Vergleich zum LHW- oder Organosolv-Aufschluss liefert. Um eine Anode herzustellen, wurde das resultierende Si–C-Kompositmaterial carbonisiert, auf einen Stromsammler aufgetragen und elektro-chemisch charakterisiert. Der Einfluss der Vorbehandlungsschritte auf den Herstellungsprozess und die ökonomische Bewertung des untersuchten Bioraffinerie-Prozesses wurde mithilfe eines Stoffstrommodells analysiert.
Bacillus pumilus reveals a remarkably high resistance to hydrogen peroxide provoked oxidative stress

Bacillus pumilus is characterized by a higher oxidative stress resistance than other comparable industrially relevant Bacilli such as B. subtilis or B. licheniformis. In this study the response of B. pumilus to oxidative stress was investigated during a treatment with high concentrations of hydrogen peroxide at the proteome, transcriptome and metabolome level. Genes/proteins belonging to regulons, which are known to have important functions in the oxidative stress response of other organisms, were found to be upregulated, such as the Fur, Spx, SOS or CtsR regulon. Strikingly, parts of the fundamental PerR regulon responding to peroxide stress in B. subtilis are not encoded in the B. pumilus genome. Thus, B. pumilus misses the catalase KatA, the DNA-protection protein MrgA or the alkyl hydroperoxide reductase AhpCF. Data of this study suggests that the catalase KatX2 takes over the function of the missing KatA in the oxidative stress response of B. pumilus. The genome-wide expression analysis revealed an induction of bacillithiol (Cys-GlcN-malate, BSH) relevant genes. An analysis of the intracellular metabolites detected high intracellular levels of this protective metabolite, which indicates the importance of bacillithiol in the peroxide stress resistance of B. pumilus.
Molke als Nebenprodukt der Käseherstellung wurde lange Zeit als Abfall betrachtet. Bedingt durch ihren hohen BOD (biological oxygen demand) war die direkte Einleitung in Gewässer, aber auch der mikrobielle Abbau in Kläranlagen bedenklich. Falls eine Weiterverarbeitung der Molke stattfand, so geschah dies meist zu Molkepulver oder Proteinkonzentrat. Als Untersuchungen der Molkeproteine jedoch unter pharmazeutischen Gesichtspunkten interessante Eigenschaften nahelegten, stieg das Interesse am Bioprodukt Molke und ihren Proteinen an. So stehen beispielsweise für die Molkeproteine a-Lactalbumin (ala) und b-Lactoglobulin (blg) antibakterielle, anticancerogene und diverse andere physiologische Effekte in der Diskussion. Gegenwärtig finden meist Membranverfahren zur Aufreinigung von Molkeproteinen Anwendung. Als alternatives Verfahren wurde am Institut für Bioverfahrenstechnik in Kaiserslautern ein chromatographisches Verfahren entwickelt, bei dem natürliche Tonminerale zum Einsatz kamen. Nach chemischer und physikalischer Modifikation des Ausgangsmaterials durch den Hersteller Süd-Chemie wurden drei der Adsorber für nähere Untersuchungen zur Auftrennung von Molkeproteinen aus Molkekonzentrat herangezogen. Nach einer Cross-Flow-Filtration des Molkekonzentrats erfolgte die Aufreinigung der Molkeproteine in einem FPLC-System.
Cytochrom P450 sind Häm-Proteine, die zur Enzymklasse der Oxidoreduktasen (EC 1.14.xy) gehören. Eine wichtige Reaktion ist die Hydroxylierung nichtaktivierter C–H-Bindungen, die in technischen Systemen von großem Interesse ist. Durch die Verwendung von M-IDA-2-Partikeln ist eine direkte Aufreinigung mit gleichzeitiger Immobilisierung und die Applikation der Enzyme aus dem Zelllysat möglich. Damit ist das Verfahren mehr als fünf Stunden schneller als die konventionelle Chromatographie und mehr als 80 % der Aufreinigungszeit wird gespart. Mit dem isolierten nativen Enzym konnte die Plattformchemikalie 9,10-Dihydroxystearinsäure aus Ölsäure hergestellt werden. Unter anderem für die Kunststoffindustrie können aus diesem Produkt wichtige Monomere wie z. B. Azelainsäure hergestellt werden. Die Bildung des Produkts erfolgt in einem zweiphasigen Reaktionssystem an der Grenzfläche zwischen dem Öl und der wässrigen Phase als Feststoff. Um das immobilisierte Enzym aktiv in die obere Phase zu transportieren, wurde eine neue magnetische Mischvorrichtung entwickelt. Das Reaktionsprodukt wurde mit NMR, GC-MS und HPLC-MS analysiert und mit einem chemisch synthetisierten Standard von 9,10-Dihydroxystearinsäure verglichen. Derzeit werden Studien des immobilisierten Häms des Enzyms durchgeführt.
Die selektive Isolierung von Cephalosporin C (CPC) aus komplexen Fermentationssuspensionen unter Einsatz magnetischer Separation ist das Ziel dieser Arbeit. Das Verfahren wird im frühen Stadium der Aufarbeitung genutzt, um CPC zu stabilisieren und somit die Produktausbeute zu erhöhen. Als Adsorbersysteme für CPC wurden neben einem projektinternen magnetischen Material ND 10322, dessen Oberflächenladungen spezifisch für die Bindung des Zielmoleküls synthetisiert wurden, verschiedene kommerzielle Partikelsysteme verglichen. Es konnten massenspezifische Maximalbeladungen von 51 mg g⁻¹ erreicht werden. Weiterhin wurde die Stabilität von CPC untersucht. Unter optimalen Adsorptionsbedingungen kann CPC stabilisiert werden, so dass die Geschwindigkeitskonstante der Degradation des b-Lactam-Rings unter diesen Bedingungen unter 0,005 h⁻¹ liegt. Untersuchungen zur Wiederverwertbarkeit der neuen Adsorbers zeigten eine irreversible Bindung geringer CPC-Mengen nach dem ersten Einsatz. Nach zwölf Zyklen tritt eine irreversible Bindung von CPC ein, was zu einer signifikanten Reduktion der Adsorptionsfähigkeit führt. Die Anhäufung des CPC auf dem Adsorber konnte durch IR-Untersuchungen auf die Bildung einer Peptidbindung zwischen Carboxylgruppen des CPC und Aminogruppe der Adsorberoberfläche zurückgeführt werden.
b-Lactame gehören zu den wirkungsvollsten Antibiotika, jedoch lassen sich viele nur schwierig fermentativ erzeugen. Ein Problem bei der fermentativen Produktion ist die Hydrolyse des Lactamrings im wässrigen Milieu. Das Ziel des von der DBU geförderten Projekts ist die selektive In-situ-Adsorption der b-Lactamantibiotika unter anschließender magnetischer Separation. Durch die Isolation im Hochgradientenmagnetseparator (HGMS) ist eine Fest-fest-flüssig-Trennung und somit ein erheblicher Zeitgewinn im Downstreamprozess möglich. Zusätzlich kommt es zur Einsparung an Lösungsmittel und Energie, was neben Reduzierung der Antibiotikahydrolyse auch in ökologischer Hinsicht einen interessanten Aspekt darstellt. Als Trägermaterial für die Adsorbermatrix werden magnetisierbare Eisenoxidpartikel eingesetzt, die in einer Silikamatrix eingebettet sind. Diese Adsorber sollen auf Selektivität in Wasser und verschiedenen Medien getestet werden. Zusätzlich werden die Abbauprodukte des b-Lactams analysiert, um eine Aussage über die Stabilisierung des Antibiotikums durch die selektiven Adsorber treffen zu können. Diese Ergebnisse werden mit kommerziell erhältlichen Adsorbern verglichen. Die Aufreinigung der Antibiotika soll direkt aus der Fermentationsbrühe erfolgen. Um die Trennung der magnetischen, selektiven Adsorber von der Biomasse zu gewährleisten, soll der HGMS in die Fermentation integriert werden. Das filamentöse Wachstum des Mikroorganismus erfordert eine Neuauslegung der Filtermatrix.
Aufarbeitung von Polyphenolen aus Weizenmittels Zeolithen am Beispiel der Ferulasa¨ ureAlexander Thiel1, Kai Muffler1, Nils Tippko¨ tter1, Kirstin Suck2, Ulrich Sohling2, Friedrich Ruf3und Roland Ulber1,*DOI: 10.1002/cite.201400031Bei der Ferulasa¨ure handelt es sich um einen Wertstoff, der aus Weizen gewonnen und in der Lebensmittel- und Pharma-industrie eingesetzt werden kann. Der Einsatz von Weizen als nachwachsende Rohstoffquelle ist allerdings nur dann wirt-schaftlich durchfu¨hrbar, wenn eine Prozessintegration in die bestehenden industriellen Verfahren gewa¨hrleistet oder einedirekte Konkurrenz zur Mehl- und Sta¨rkeindustrie vermieden werden kann. In diesem Artikel wird ein Verfahren aufge-zeigt, welches hohe Ausbeuten ermo¨glicht und eine Konkurrenz zu bestehenden Verwertungspfaden vermeidet.
The Monte Carlo code FLUKA is used to simulate the production of a number of positron emitting radionuclides, ¹⁸F, ¹³N, ⁹⁴Tc, ⁴⁴Sc, ⁶⁸Ga, ⁸⁶Y, ⁸⁹Zr, ⁵²Mn, ⁶¹Cu and ⁵⁵Co, on a small medical cyclotron with a proton beam energy of 13 MeV. Experimental data collected at the TR13 cyclotron at TRIUMF agree within a factor of 0.6 ± 0.4 with the directly simulated data, except for the production of ⁵⁵Co, where the simulation underestimates the experiment by a factor of 3.4 ± 0.4. The experimental data also agree within a factor of 0.8 ± 0.6 with the convolution of simulated proton fluence and cross sections from literature. Overall, this confirms the applicability of FLUKA to simulate radionuclide production at 13 MeV proton beam energy.
Air- and water-stable phenyl complexes with nitridotechnetium(V) cores can be prepared by straightforward procedures. [TcNPh2(PPh3)2] is formed by the reaction of [TcNCl2(PPh3)2] with PhLi. The analogous N-heterocyclic carbene (NHC) compound [TcNPh2(HLPh)2], where HLPh is 1,3,4-triphenyl-1,2,4-triazol-5-ylidene, is available from (NBu4)[TcNCl4] and HLPh or its methoxo-protected form. The latter compound allows the comparison of different Tc–C bonds within one compound. Surprisingly, the Tc chemistry with such NHCs does not resemble that of corresponding Re complexes, where CH activation and orthometalation dominate.
For the successful implementation of microfluidic reaction systems, such as PCR and electrophoresis, the movement of small liquid volumes is essential. In conventional lab-on-a-chip-platforms, solvents and samples are passed through defined microfluidic channels with complex flow control installations. The droplet actuation platform presented here is a promising alternative. With it, it is possible to move a liquid drop (microreactor) on a planar surface of a reaction platform (lab-in-a-drop). The actuation of microreactors on the hydrophobic surface of the platform is based on the use of magnetic forces acting on the outer shell of the liquid drops which is made of a thin layer of superhydrophobic magnetite particles. The hydrophobic surface of the platform is needed to avoid any contact between the liquid core and the surface to allow a smooth movement of the microreactor. On the platform, one or more microreactors with volumes of 10 µL can be positioned and moved simultaneously. The platform itself consists of a 3 x 3 matrix of electrical double coils which accommodate either neodymium or iron cores. The magnetic field gradients are automatically controlled. By variation of the magnetic field gradients, the microreactors' magnetic hydrophobic shell can be manipulated automatically to move the microreactor or open the shell reversibly. Reactions of substrates and corresponding enzymes can be initiated by merging the microreactors or bringing them into contact with surface immobilized catalysts.
We present a concise mini overview on the approaches to the disposal of nuclear waste currently used or deployed. The disposal of nuclear waste is the end point of nuclear waste management (NWM) activities and is the emplacement of waste in an appropriate facility without the intention to retrieve it. The IAEA has developed an internationally accepted classification scheme based on the end points of NWM, which is used as guidance. Retention times needed for safe isolation of waste radionuclides are estimated based on the radiotoxicity of nuclear waste. Disposal facilities usually rely on a multi-barrier defence system to isolate the waste from the biosphere, which comprises the natural geological barrier and the engineered barrier system. Disposal facilities could be of a trench type, vaults, tunnels, shafts, boreholes, or mined repositories. A graded approach relates the depth of the disposal facilities’ location with the level of hazard. Disposal practices demonstrate the reliability of nuclear waste disposal with minimal expected impacts on the environment and humans.
The use of transgenic animal models has transformed our knowledge of complex biochemical pathways in vivo. It has allowed disease processes to be modelled and used in the development of new disease prevention and treatment strategies. They can also be used to define cell- and tissue-specific pathways of gene regulation. A further major application is in the area of preclinical development where such models can be used to define pathways of chemical toxicity, and the pathways that regulate drug disposition. One major application of this approach is the humanisation of mice for the proteins that control drug metabolism and disposition. Such models can have numerous applications in the development of drugs and in their more sophisticated use in the clinic.
Multi-analyte biosensors may offer the opportunity to perform cost-effective and rapid analysis with reduced sample volume, as compared to electrochemical biosensing of each analyte individually. This work describes the development of an enzyme-based biosensor system for multi-parametric determination of four different organic acids. The biosensor array comprises five working electrodes for simultaneous sensing of ethanol, formate, d-lactate, and l-lactate, and an integrated counter electrode. Storage stability of the biosensor was evaluated under different conditions (stored at +4 °C in buffer solution and dry at −21 °C, +4 °C, and room temperature) over a period of 140 days. After repeated and regular application, the individual sensing electrodes exhibited the best stability when stored at −21 °C. Furthermore, measurements in silage samples (maize and sugarcane silage) were conducted with the portable biosensor system. Comparison with a conventional photometric technique demonstrated successful employment for rapid monitoring of complex media.
Die Ausbildung von Biofilmen in technischen Anlagen, wie z. B. Kühlkreisläufen, Wasseraufbereitungssystemen und Bioreaktoren, führen zu Materialschäden (Biofouling) und stark erhöhtem Energieaufwand. Im Rahmen der aktuellen Forschungsarbeiten erfolgen aktive sowie passive Bio-Modifikationen auf funktionalisierten magnetischen Mikropartikelober-flächen. Um die verschiedenen funktionalisierten magnetischen Mikropartikel zu analysieren und ihre antimikrobielle Wirkung zu testen, wird der Einsatz einer 3D-gedruckten, magnetischen Plattform für ein Fluoreszenz-basiertes Screening-System untersucht. Für den Oberflächenschutz wurden verschiedene, antimikrobiell funktionalisierte Partikelkombinationen mit dem Mikroorganismus Escherichia coli GFPmut2 in Bezug auf aktiven Oberflächenschutz verglichen. Um die antimikrobielle Oberflächeneffekte von synergistischen Kombinationen unterschiedlich funktionalisierter Partikel zu bestimmen, werden Oberflächen einem Magnetfeld ausgesetzt, das die Mikropartikel als definierte Schicht auf ihnen zurück hält. Diese modifizierten Oberflächen können sowohl durch Fluoreszenzspektroskopie als auch -mikroskopie analysiert werden.
In most beers, producers strive to minimize haze to maximize visual appeal. To detect the formation of particulates, a measurement system for sub-micron particles is required. Beer haze is naturally occurring, composed of protein or polyphenol particles; in their early stage of growth their size is smaller than 2 µm. Microscopy analysis is time and resource intensive; alternatively, backscattering is an inexpensive option for detecting particle sizes of interest.
Autoradiography is a well-established method of nuclear imaging. When different radionuclides are present simultaneously, additional processing is needed to distinguish distributions of radionuclides. In this work, a method is presented where aluminium absorbers of different thickness are used to produce images with different cut-off energies. By subtracting images pixel-by-pixel one can generate images representing certain ranges of β-particle energies. The method is applied to the measurement of irradiated reactor graphite samples containing several radionuclides to determine the spatial distribution of these radionuclides within pre-defined energy windows. The process was repeated under fixed parameters after thermal treatment of the samples. The greyscale images of the distribution after treatment were subtracted from the corresponding pre-treatment images. Significant changes in the intensity and distribution of radionuclides could be observed in some samples. Due to the thermal treatment parameters the most significant differences were observed in the ³H and ¹⁴C inventory and distribution.
The composition and physiochemical properties of aquatic-phase natural organic matter (NOM) are most important problems for both environmental studies and water industry. Laser desorption/ionization (LDI) mass spectrometry facilitated successful examinations of NOM, as humic and fulvic acids in NOM are readily ionized by the nitrogen laser. In this study, hydrophobic NOMs (HPO NOMs) from river, reservoir and waste water were characterized by this technique. The effect of analytical variables like concentration, solvent composition and laser energy was investigated. The exact masses of small molecular NOM moieties in the range of 200–1200 m/z were determined in reflectron mode. In addition, spectra of post-source-decay experiments in this range showed that some compounds from different natural NOMs had the same fragmental ions. In the large mass range of 1200–15 000 Da, macromolecules and their aggregates were found in HPO NOMs from natural waters. Highly humic HPO exhibited mass peaks larger than 8000 Da. On the other hand, the waste water and reservoir water mainly had relatively smaller molecules of about 2000 Da. The LDI-MS measurements indicated that highly humic river waters were able to form large aggregates and membrane foulants, while the HPO NOMs from waste water and reservoir water were unlikely to form large aggregates.
