Previous studies optimized the dimensions of coaxial heat exchangers using constant mass fow rates as a boundary condition. They show a thermal optimal circular ring width of nearly zero. Hydraulically optimal is an inner to outer pipe radius ratio of 0.65 for turbulent and 0.68 for laminar fow types. In contrast, in this study, fow conditions in the circular ring are kept constant (a set of fxed Reynolds numbers) during optimization. This approach ensures fxed fow conditions and prevents inappropriately high or low mass fow rates. The optimization is carried out for three objectives: Maximum energy gain, minimum hydraulic efort and eventually optimum net-exergy balance. The optimization changes the inner pipe radius and mass fow rate but not the Reynolds number of the circular ring. The thermal calculations base on Hellström’s borehole resistance and the hydraulic optimization on individually calculated linear loss of head coefcients. Increasing the inner pipe radius results in decreased hydraulic losses in the inner pipe but increased losses in the circular ring. The net-exergy diference is a key performance indicator and combines thermal and hydraulic calculations. It is the difference between thermal exergy fux and hydraulic efort. The Reynolds number in the circular ring is instead of the mass fow rate constant during all optimizations. The result from a thermal perspective is an optimal width of the circular ring of nearly zero. The hydraulically optimal inner pipe radius is 54% of the outer pipe radius for laminar fow and 60% for turbulent fow scenarios. Net-exergetic optimization shows a predominant infuence of hydraulic losses, especially for small temperature gains. The exact result depends on the earth’s thermal properties and the fow type. Conclusively, coaxial geothermal probes’ design should focus on the hydraulic optimum and take the thermal optimum as a secondary criterion due to the dominating hydraulics.
Im Studiengang Mikrosystemtechnik des Fachhochschulstandortes Zweibrücken werden zwei neue moderne Anlagen für die Herstellung von mikrotechnischen Komponenten in Betrieb genommen: Ein Oxidationsofen für Herstellung dünner Oxidschichten auf Silizium-Einkristallen und eine Belichtungsapparatur für die Fotolithografie - das Besondere an diesen Anlagen: Sie existieren nur virtuell, d.h. als Animationen in einer Computerwelt.
Im Rahmen eines modernen Blitzschutzsystems für Stahlbeton-Bauten bietet es sich an, die Betonbewehrung zu benutzen: - Sie kann die Funktionen der Ableitungseinrichtungen und des Blitzschutz- Potentialausgleichs bei einem klassischen Gebäude-Blitzschutz übernehmen [1]; - Sie kann, ggf. bei entsprechender Ergänzung, als ein geschlossener Käfig ausgebildet werden und damit eine deutliche Reduzierung der Belastung elektrischer / elektronischer Systeme durch blitzinduzierte elektromagnetische Felder erbringen (LEMP-Schutz [2]). Die Nutzung der Bewehrung ist dabei grundsätzlich gleichermaßen bei Neubauten wie auch bei Ertüchtigungen möglich und sinnvoll. So stellt die Nutzung der Bewehrung beispielsweise im Bereich von Großkraftwerken eine wesentliche Ertüchtigungsmaßnahme für den Blitzschutz elektrischer und elektronischer Einrichtungen dar: - Einerseits wird der Blitzschutz-Potentialausgleich durch den Anschluss metallener Einrichtungen wie Elektronik-Schränke, Kabeltrag-Konstruktionen, Rohrleitungen, etc. an die Bewehrung deutlich verbessert. - Andererseits kann bei größeren Gebäuden die elektromagnetische Schirmwirkung durch die elektrische Überbrückung von vorhandenen Dehnfugen bei Stahlbetonbauten optimiert werden. Diese Dehnfugen sind teilweise nur unzureichend überbrückt, so dass bei Blitzeinschlag in das betreffende oder ein benachbartes Gebäude an Kabelstrecken, die über die Dehnfuge hinwegführen, rel. hohe Spannungen induziert werden können [2, 3]. Die sich um das gesamte Gebäude herumziehende oder zwischen zwei Gebäuden befindliche Dehnfuge muss deshalb im Abstand von maximal einigen Metern überbrückt werden. Im Falle von Blitzschutz-Ertüchtigungen in vorhandenen Gebäuden wird bisher an jeder geplanten Anschlussstelle die Bewehrung großflächig (∅ wenige 10 cm) freigelegt, dort ein elektrischer Anschluss zu dem Bewehrungsstab hergestellt, z.B. mittels eines Erdungsfestpunkts, und dann die Betonoberfläche wieder geschlossen. Je nach prognostizierter Strombelastung wird teilweise versucht, den über den Anschluss fließenden Strom bereits auf mehrere Bewehrungsstäbe zu verteilen. Dazu sind entweder die kreuzenden Stäbe zu verschweißen oder es sind direkt Anschlüsse an zwei Bewehrungsstäbe herzustellen. All dieses bedeutet einen hohen Aufwand bei der Freilegung der Bewehrung und auch wieder bei der Schließung der entstandenen Betonlöcher. Es soll deshalb hier untersucht werden, ob es beispielsweise zum Zwecke des Blitzschutz-Potentialausgleichs und auch zur Überbrückung von Dehnfugen ausreichend ist, den Anschluss an die Bewehrung nach einfachen Verfahren nur jeweils an einen Bewehrungsstab herzustellen. Damit würde der finanzielle und administrative Aufwand an Betonarbeiten deutlich reduziert. Die hier dargestellten Verfahren sind dabei insbesondere für den Einsatz bei Blitzschutz-Ertüchtigungen in bestehenden Gebäuden vorgesehen. Abschließend sollen deshalb die Möglichkeiten zur Prüfung korrekter Anschlüsse, die Grenzen der Verfahren sowie auch die Grenzen der Anwendbarkeit bei Neuanlagen diskutiert werden.
The aim of the present study was the characterisation of three true subtilisins and one phylogenetically intermediate subtilisin from halotolerant and halophilic microorganisms. Considering the currently growing enzyme market for efficient and novel biocatalysts, data mining is a promising source for novel, as yet uncharacterised enzymes, especially from halophilic or halotolerant Bacillaceae, which offer great potential to meet industrial needs. Both halophilic bacteria Pontibacillus marinus DSM 16465ᵀ and Alkalibacillus haloalkaliphilus DSM 5271ᵀ and both halotolerant bacteria Metabacillus indicus DSM 16189 and Litchfieldia alkalitelluris DSM 16976ᵀ served as a source for the four new subtilisins SPPM, SPAH, SPMI and SPLA. The protease genes were cloned and expressed in Bacillus subtilis DB104. Purification to apparent homogeneity was achieved by ethanol precipitation, desalting and ion-exchange chromatography. Enzyme activity could be observed between pH 5.0–12.0 with an optimum for SPPM, SPMI and SPLA around pH 9.0 and for SPAH at pH 10.0. The optimal temperature for SPMI and SPLA was 70 °C and for SPPM and SPAH 55 °C and 50 °C, respectively. All proteases showed high stability towards 5% (w/v) SDS and were active even at NaCl concentrations of 5 M. The four proteases demonstrate potential for future biotechnological applications.
Amino acid-based surfactants are valuable compounds for cosmetic formulations. The chemical synthesis of acyl-amino acids is conventionally performed by the Schotten-Baumann reaction using fatty acyl chlorides, but aminoacylases have also been investigated for use in biocatalytic synthesis with free fatty acids. Aminoacylases and their properties are diverse; they belong to different peptidase families and show differences in substrate specificity and biocatalytic potential. Bacterial aminoacylases capable of synthesis have been isolated from Burkholderia, Mycolicibacterium, and Streptomyces. Although several proteases and peptidases from S. griseus have been described, no aminoacylases from this species have been identified yet. In this study, we investigated two novel enzymes produced by S. griseus DSM 40236ᵀ . We identified and cloned the respective genes and recombinantly expressed an α-aminoacylase (EC 3.5.1.14), designated SgAA, and an ε-lysine acylase (EC 3.5.1.17), designated SgELA, in S. lividans TK23. The purified aminoacylase SgAA was biochemically characterized, focusing on its hydrolytic activity to determine temperature- and pH optima and stabilities. The aminoacylase could hydrolyze various acetyl-amino acids at the Nα -position with a broad specificity regarding the sidechain. Substrates with longer acyl chains, like lauroyl-amino acids, were hydrolyzed to a lesser extent. Purified aminoacylase SgELA specific for the hydrolysis of Nε -acetyl-L-lysine was unstable and lost its enzymatic activity upon storage for a longer period but could initially be characterized. The pH optimum of SgELA was pH 8.0. While synthesis of acyl-amino acids was not observed with SgELA, SgAA catalyzed the synthesis of lauroyl-methionine.
Novel organic membrane-based thin-film microsensors for the determination of heavy metal cations

A first step towards the fabrication and electrochemical evaluation of thin-film microsensors based on organic PVC membranes for the determination of Hg(II), Cd(II), Pb(II) and Cu(II) ions in solutions has been realised. The membrane-coating mixture used in the preparation of this new type of microsensors is incorporating PVC as supporting matrix, o-nitrophenyloctylether (o-NPOE) as solvent mediator and a recently synthesized Hg[dimethylglyoxime(phene)]2+ and Bis-(4-hydroxyacetophenone)-ethylenediamine as electroactive materials for Hg(II) and Cd(II), respectively. A set of three commercialised ionophores for Cd(II), Pb(II) and Cu(II) has been also used for comparison. Thin-film microsensors based on these membranes showed a Nernstian response of slope (26-30 mV/dec.) for the respective tested cations. The potentiometric response characteristics (linear range, pH range, detection limit and response time) are comparable with those obtained by conventional membranes as well as coated wire electrodes prepared from the same membrane. The realisation of the new organic membrane-based thin-film microsensors overcomes the problem of an insufficient selectivity of solid-state-based thinfilm sensors.
N-Acyl-amino acids can act as mild biobased surfactants, which are used, e.g., in baby shampoos. However, their chemical synthesis needs acyl chlorides and does not meet sustainability criteria. Thus, the identification of biocatalysts to develop greener synthesis routes is desirable. We describe a novel aminoacylase from Paraburkholderia monticola DSM 100849 (PmAcy) which was identified, cloned, and evaluated for its N-acyl-amino acid synthesis potential. Soluble protein was obtained by expression in lactose autoinduction medium and co-expression of molecular chaperones GroEL/S. Strep-tag affinity purification enriched the enzyme 16-fold and yielded 15 mg pure enzyme from 100 mL of culture. Biochemical characterization revealed that PmAcy possesses beneficial traits for industrial application like high temperature and pH-stability. A heat activation of PmAcy was observed upon incubation at temperatures up to 80 °C. Hydrolytic activity of PmAcy was detected with several N-acyl-amino acids as substrates and exhibited the highest conversion rate of 773 U/mg with N-lauroyl-L-alanine at 75 °C. The enzyme preferred long-chain acyl-amino-acids and displayed hardly any activity with acetyl-amino acids. PmAcy was also capable of N-acyl-amino acid synthesis with good conversion rates. The best synthesis results were obtained with the cationic L-amino acids L-arginine and L-lysine as well as with L-leucine and L-phenylalanine. Exemplarily, L-phenylalanine was acylated with fatty acids of chain lengths from C8 to C18 with conversion rates of up to 75%. N-lauroyl-L-phenylalanine was purified by precipitation, and the structure of the reaction product was verified by LC–MS and NMR.

The aerodynamic performance of propellers strongly depends on their geometry and, consequently, on aeroelastic deformations. Knowledge of the extent of the impact is crucial for overall aircraft performance. An integrated simulation environment for steady aeroelastic propeller simulations is presented. The simulation environment is applied to determine the impact of elastic deformations on the aerodynamic propeller performance. The aerodynamic module includes a blade element momentum approach to calculate aerodynamic loads. The structural module is based on finite beam elements, according to Timoshenko theory, including moderate deflections. Several fixed-pitch propellers with thin-walled cross sections made of both isotropic and non-isotropic materials are investigated. The essential parameters are varied: diameter, disc loading, sweep, material, rotational, and flight velocity. The relative change of thrust between rigid and elastic blades quantifies the impact of propeller elasticity. Swept propellers of large diameters or low disc loadings can decrease the thrust significantly. High flight velocities and low material stiffness amplify this tendency. Performance calculations without consideration of propeller elasticity can lead to decreased efficiency. To avoid cost- and time-intense redesigns, propeller elasticity should be considered for swept planforms and low disc loadings.
Die Auswirkungen psychischer Erkrankungen stellen ein zunehmendes Thema in unserer Gesellschaft dar. Allein in Deutschland erkranken jedes Jahr ein Viertel aller Menschen psychisch. Besonders seit der Pandemie wächst die Nachfrage nach psychotherapeutischer Behandlung stetig an, wodurch die Wartelisten in den Praxen immer länger werden. Betroffene müssen oft ein halbes Jahr oder länger auf einen Therapieplatz warten und sind gezwungen zu nehmen, was gerade frei ist. Dabei ist es für den Therapieerfolg essenziell, dass nicht nur die Therapie auf einen abgestimmt ist, sondern auch die Chemie zwischen Therapeut:in und Patient:in passt. OpenCouch ist eine Anwendung, die Betroffene bei der Therapieplatzsuche unterstützen soll, indem sie Informationen und Tools zur Verfügung stellt, um trotz der geringen Verfügbarkeit einen Therapieplatz finden zu können, der einen Behandlungserfolg garantiert.
The present work aimed to study the mainstream feasibility of the deammonifying sludge of side stream of municipal wastewater treatment plant (MWWTP) in Kaster, Germany. For this purpose, the deammonifying sludge available at the side stream was investigated for nitrogen (N) removal with respect to the operational factors temperature (15–30°C), pH value (6.0–8.0) and chemical oxygen demand (COD)/N ratio (≤1.5–6.0). The highest and lowest N-removal rates of 0.13 and 0.045 kg/(m³ d) are achieved at 30 and 15°C, respectively. Different conditions of pH and COD/N ratios in the SBRs of Partial nitritation/anammox (PN/A) significantly influenced both the metabolic processes and associated N-removal rates. The scientific insights gained from the current work signifies the possibility of mainstream PN/A at WWTPs. The current study forms a solid basis of operational window for the upcoming semi-technical trails to be conducted prior to the full-scale mainstream PN/A at WWTP Kaster and WWTPs globally.

Mittels eines operationalen Ansatzes zur Semantikdefinition wird am Bei-spiel des konzeptuellen Gebäudeentwurfs ein Regelsystem formalisiert. Dazu werdenzwei Teile, zum einen das Regelwissen, zum anderen ein konzeptueller Entwurfsplan zunächst informell eingeführt und dann formal beschrieben. Darauf aufbauend wird die Grundlage für eine Konsistenzprüfung des konzeptuellen Entwurfs gegen das Regel-wissen formal angeben
Ziel war die technische Überarbeitung und Optimierung des vorliegenden Prototypen, um eine Basis für ein E-Learning System (Internet unterstütztes Lernen) zu schaffen. Zu diesem Zweck wurde das Programm der Robotersteuerung neu erstellt, notwendige mechanische Änderungen an Roboterequipment und Materialförderung vorgenommen sowie eine neue Internet-Anbindung geschaffen. Wesentliche Ergebnisse der Arbeit sind: • Erzeugen roboterbasierter kreisinterpolierter Bahnschweißnähte • Kommunikation des Lernsystems via Internet • Lernorientierte Steuerungssoftware

Low-end-Embedded-Plattformen stellen eine hohe Anforderung an die Entscheidungsfähigkeit des Entwicklers: Zum nächstgrößeren Prozessor greifen und ein Betriebssystem benutzen oder doch besser auf das Betriebssystem verzichten? Die Frage lässt sich einfach beantworten: Einen Nanokernel verwenden und das Embedded-System mit einem minimalen Footprint realisieren. Adam Dunkels Protothreads sind eine ausgesprochen effiziente Art, Mikrocontroller gut strukturiert zu programmieren und gleichzeitig auf Overhead zu verzichten. So können auch mit kleinen 8-bit-Prozessoren anspruchsvolle Aufgaben in einem Thread-Modell bearbeitet werden. Man muss also nicht immer das Rad neu erfinden oder gleich auf Linux-basierte Systeme zurückgreifen.

Bis vor kurzem basierte eine erschwingliche Hardware meist auf Mikrocontrollern mit den entsprechenden Leistungseinschränkungen z.B. im Bereich der Bildverarbeitung. Die Wahrnehmung einer 3D-Umgebung und somit die Möglichkeit zur autonomen Navigation wurde mit relativ kostenintensiver Hardware, z.B. Stereo-Vision-Systemen und Laserscannern gelöst. Die zur Auswertung der Sensorik notwendige Rechenleistung stand - entweder aufgrund des Stromverbrauchs oder der Performance meist für mobile Plattformen (lokal) - nicht zur Verfügung. (S. 136-137) Eine nachhaltige Sicherung der Funktionalität und der ökologischen Verträglichkeit eines mittleren bis großen Kanalnetzes erfordert eine umfassende und detaillierte Modellierung in Raum und Zeit. Um den in den Richtlinien geforderten statistischen Anforderungen gerecht zu werden und die jährlichen Häufigkeiten, Mengen und Zeiträume der Belastungen erwartungstreu abschätzen zu können, ist es zielführend und notwendig, lange Zeiträume und die Gesamtheit der Einzugsgebiete möglichst detailliert zu betrachten. Die hydraulische Funktionalität und Sicherheit soll meistens mit Hilfe zeitsymmetrischer (hydrodynamischer) Verfahren nur durch Betrachtung von Modellregen, allenfalls Regenserien, sichergestellt werden. Für die Abschätzung der jährlich zu erwartenden Emissionen in unsere natürlichen Gewässer mit ihren Mengen, Frequenzen und Dauern werden normalerweise Langzeitsimulationen natürlicher Regenreihen über möglichst große Zeiträume mit zeitasymmetrischen (hydrologischen) Verfahren durchgeführt. Die betrachteten Kanalnetze werden zumeist vereinfacht (Grobnetze), um die Rechenzeiten erträglicher zu gestalten. Wünschenswert wäre jedoch eine allen Anforderungen gerecht werdende wirklichkeitsnahe Modellierung des gesamten Kanalnetzes in all seinen Details, Vermaschungen und Wechselwirkungen (Feinnetz) und dessen zeitsymmetrische und damit verlässliche Simulation mit langjährigen Regenreihen. Bereits vor 15 Jahren wurde im Hause Tandler begonnen, die Berechnungssoftware durch Parallelisierung auf symmetrische Multiprozessortechnologien auszurichten. In neuerer Zeit hält diese Technik durch die Mehrkernprozessoren in normalen Notebooks und PCs Einzug in die Ingenieurbüros und Abwasserbetriebe und sorgt schon für wesentliche Einsparungen an Rechenzeit. Doch erst durch die Kombination der Parallelisierung mit dem Prinzip des verteilten Rechnens (d.h. die Einbeziehung mehrerer PCs eines Netzwerkes in die Berechnung) erhält man die Chance ausreichend Rechenkapazität zur Verfügung zu stellen, um nicht nur eine einzelne Langzeitsimulation eines Feinnetzes durchzuführen, sondern sogar mehrere Sanierungsalternativen zu überprüfen. Die zukunftsweisenden Arbeiten von Dipl. Math. R. Tandler auf diesem Gebiet sind Thema dieses Vortrags.
Applications of Graph Transformations with Industrial Relevance Lecture Notes in Computer Science, 2004, Volume 3062/2004, 90-105, DOI: 10.1007/978-3-540-25959-6_7 In this paper we discuss how tools for conceptual design in civil engineering can be developed using graph transformation specifications. These tools consist of three parts: (a) for elaborating specific conceptual knowledge (knowledge engineer), (b) for working out conceptual design results (architect), and (c) automatic consistency analyses which guarantee that design results are consistent with the underlying specific conceptual knowledge. For the realization of such tools we use a machinery based on graph transformations. In a traditional PROGRES tool specification the conceptual knowledge for a class of buildings is hard-wired within the specification. This is not appropriate for the experimentation platform approach we present in this paper, as objects and relations for conceptual knowledge are due to many changes, implied by evaluation of their use and corresponding improvements. Therefore, we introduce a parametric specification method with the following characteristics: (1) The underlying specific knowledge for a class of buildings is not fixed. Instead, it is built up as a data base by using the knowledge tools. (2) The specification for the architect tools also does not incorporate specific conceptual knowledge. (3) An incremental checker guarantees whether a design result is consistent with the current state of the underlying conceptual knowledge (data base).
In this paper, methods of sample preparation for potentiometric measurement of phenylalanine are presented. Basing on the spectrophotometric measurements of phenylalanine, the concentrations of reagents of the enzymatic reaction (10 mM L-Phe, 0,4 mM NAD+, 2U L-PheDH) were determined. Then, the absorption spectrum of the reaction product, NADH, was monitored (maximum peak at 340 nm). The results obtained by the spectrophotometric method were compared with the results obtained by the colourimetry, using pH indicators. The above-mentioned two methods will be used as references for potentiometric measurements of phenylalanine concentration.
Photoelectrochemical (PEC) biosensors are a rather novel type of biosensors thatutilizelighttoprovideinformationaboutthecompositionofananalyte,enablinglight-controlled multi-analyte measurements. For enzymatic PEC biosensors,amperometric detection principles are already known in the literature. In con-trast, there is only a little information on H+-ion sensitive PEC biosensors. Inthis work, we demonstrate the detection of H+ions emerged by H+-generatingenzymes, exemplarily demonstrated with penicillinase as a model enzyme on atitanium dioxide photoanode. First, we describe the pH sensitivity of the sensorand study possible photoelectrocatalytic reactions with penicillin. Second, weshow the enzymatic PEC detection of penicillin.

Recently, SHARP corporation has developed the world’s first “Plasma Cluster Ions (PCI)” air purification technology, which uses plasma discharge to generate cluster ions. The new plasma cluster device releases into the air positive and negative ions, which are harmless to humans and are able to decompose and deactivate airborne substances by chemical reactions. A lot of phenomenological tests of the PCI air purification technology on microbial cells have been conducted. And, in most cases, it has been shown that PCI demonstrate strongly pronounced killing effect. Although, the particular mechanisms of PCI action are still not evident. We studied variations in resistance to PCI among gram-positive airborne microorganisms, as well as some dose-dependent, spatial, cultural and biochemical properties of PCI action in respect of Staphylococcus spp, Enterococcus spp, Micrococcus spp.
